Rolland, Benjamin in OpenAIREBackground: Sodium oxybate (SMO) has been shown to be effective in the maintenance of abstinence (MoA) in alcohol-dependent patients in a series of small randomized controlled trials (RCTs). These results needed to be confirmed by a large trial investigating the treatment effect and its sustainability after medication discontinuation. Aims: To confirm the SMO effect on (sustained) MoA in detoxified alcohol-dependent patients. Methods: Large double-blind, randomized, placebo-controlled trial in detoxified adult alcohol-dependent outpatients (80% men) from 11 sites in four European countries. Patients were randomized to 6 months SMO (3.3–3.9 g/day) or placebo followed by a 6-month medication-free period. Primary outcome was the cumulative abstinence duration (CAD) during the 6-month treatment period defined as the number of days with no alcohol use. Secondary outcomes included CAD during the 12-month study period. Results: Of the 314 alcohol-dependent patients randomized, 154 received SMO and 160 received placebo. Based on the pre-specified fixed-effect two-way analysis of variance including the treatment-by-site interaction, SMO showed efficacy in CAD during the 6-month treatment period: mean difference +43.1 days, 95% confidence interval (17.6–68.5; p = 0.001). Since significant heterogeneity of effect across sites and unequal sample sizes among sites ( n = 3–66) were identified, a site-level random meta-analysis was performed with results supporting the pre-specified analysis: mean difference +32.4 days, p = 0.014. The SMO effect was sustained during the medication-free follow-up period. SMO was well-tolerated. Conclusions: Results of this large RCT in alcohol-dependent patients demonstrated a significant and clinically relevant sustained effect of SMO on CAD. Regional land carbon budgets provide insights on the spatial distribution of the land uptake of atmospheric carbon dioxide, and can be used to evaluate carbon cycle models and to define baselines for land-based additional mitigation efforts. The scientific community has been involved in providing observation-based estimates of regional carbon budgets either by downscaling atmospheric CO2 observations into surface fluxes with atmospheric inversions, by using inventories of carbon stock changes in terrestrial ecosystems, by upscaling local field observations such as flux towers with gridded climate and remote sensing fields or by integrating data-driven or process-oriented terrestrial carbon cycle models. The first coordinated attempt to collect regional carbon budgets for nine regions covering the entire globe in the RECCAP-1 project has delivered estimates for the decade 2000–2009, but these budgets were not comparable between regions, due to different definitions and component fluxes reported or omitted. The recent recognition of lateral fluxes of carbon by human activities and rivers, that connect CO2 uptake in one area with its release in another also requires better definition and protocols to reach harmonized regional budgets that can be summed up to the globe and compared with the atmospheric CO2 growth rate and inversion results. In this study, for the international initiative RECCAP-2 coordinated by the Global Carbon Project, which aims as an update of regional carbon budgets over the last two decades based on observations, for 10 regions covering the globe, with a better harmonization that the precursor project, we provide recommendations for using atmospheric inversions results to match bottom-up carbon accounting and models, and we define the different component fluxes of the net land atmosphere carbon exchange that should be reported by each research group in charge of each region. Special attention is given to lateral fluxes, inland water fluxes and land use fluxes.

Kibiki, G. in OpenAIREThe Pan-African Consortium for the Evaluation of Anti-Tuberculosis Antibiotics (PanACEA) was designed to build tuberculosis (TB) trial capacity whilst conducting clinical trials on novel and existing agents to shorten and simplify TB treatment. PanACEA has now established a dynamic network of 11 sub-Saharan clinical trial sites and four European research institutions.In 2011, a capacity development program, funded by the European & Developing Countries Clinical Trials Partnership (EDCTP), was launched with four objectives, aiming at strengthening collaborating TB research sites to reach the ultimate goal of becoming self-sustainable institutions: networking; training; conducting clinical trials; and infrastructure scaling-up of sites.Assessment in six sub-Saharan TB-endemic countries (Gabon, Kenya, South Africa, Tanzania, Uganda and Zambia) were performed through a structured questionnaire, site visits, discussion with the PanACEA consortium, setting of milestones and identification of priorities and followed-up with evaluations of each site. The results of this needs-based assessment was then translated into capacity development measures.In the initial phase, over a four-year period (March 2011 - June 2014), the programme scaled-up six sites; conducted a monitoring training program for 11 participants; funded five MSc and four PhD students, fostering gender balance; conducted four epidemiological studies; supported sites to conduct five Phase II studies and formed a sustainable platform for TB research (panacea-tb.net).Our experience of conducting TB clinical trials within the PanACEA programme environment of mentoring, networking and training has provided a sound platform for establishing future sustainable research centres. Our goal of facilitating emergent clinical TB trial sites to better initiate and lead research activities has been mostly successful.

Therefore, the effects of alcohol-based skin disinfection on oxygen metabolism in neutrophils were tested using 70% ethanol or an ethanol–isopropanol– n -propanol mixture. Neutrophil respiratory burst activity as assessed fluorometrically by oxidation of 2′,7′-dichlorofluorescein diacetate increased at 10 min after disinfection with 70% ethanol compared to the activity at 30 s. The increase was significant for triggering oxidative burst with formylpeptide but not with phorbol myristate acetate.

Verburg, Peter H. in OpenAIREAmbitious international targets are being developed to protect and restore biodiversity under the Convention on Biological Diversity's post-2020 Global Biodiversity Framework and the European Union's Green Deal. Yet, the land system consequences of meeting such targets are unclear, as multiple pathways may be able to deliver on the set targets. This paper introduces a novel scenario approach assessing the plural implementations of these targets. The Nature Futures Framework (NFF) developed by the Intergovernmental Science-Policy Platform on Biodiversity and Ecosystem Services aims to illustrate the different, positive ways in which society can value nature. It therefore offers a lens through which the spatial implementation of sustainability targets may be envisioned. We used CLUMondo, a spatially explicit model, to simulate plural land system scenarios for Europe for 2050. The model builds on current land system representations of Europe and explores how and where sustainability targets can be implemented under projected population trends and commodity demands. We created three different scenarios in which the sustainability targets are met, each representing an alternative, normative view on nature as represented by the NFF, favoring land systems providing strong climate regulation (Nature for Society), species conservation (Nature for Nature), or agricultural heritage features (Nature as Culture). Our results show that, irrespective of the NFF view, meeting sustainability targets will require European land systems to drastically change, as natural grasslands and forests are forecast to expand while productive areas are projected to undergo a dual intensification and diversification trajectory. Despite each NFF perspective showcasing a similar direction of change, 20% of Europe's land area will differ based on the adopted NFF perspective, with hotspots of disagreement identified in eastern and western Europe. These simulations go beyond existing scenario approaches by not only depicting broad societal developments for Europe, but also by quantifying the land system synergies and trade-offs associated with alternative, archetypal, interpretations and values of how nature may be managed for sustainability. This quantification exemplifies a means towards constructive dialogue, on the one hand by acknowledging areas of contention, and bringing such issues to the fore, and on the other by highlighting points of convergence in a vision for a sustainable Europe.

Large waste water treatment plants (WWTP) often operate nitrification in two different process environments: the cold-dilute sewage is treated in the mainstream nitrification/denitrification system, while the high strength ammonia liquors from sludge dewatering are treated in a separate high temperature reactor (SBR). This study investigates transfer from nitrifier biomass into a two-stage WWTP, commonly referred to as bioaugmentation. Besides the quantitation of ammonia oxidising bacteria (AOB), community differences were analysed with two techniques, denaturing gradient gel electrophoresis and real-time PCR melt curve analysis. It was shown that, without bioaugmentation, two distinct AOB communities establish in the mainstream and in the SBR, respectively. A gradual shift of the two AOB communities with increasing pump rates between the systems could be demonstrated. These molecular findings support process engineering experience, that cycling of waste activated sludge improves the ability of AOB to adapt to different process environments.

Motiviert durch ein Anwendungsbeispiel wird die Komfortzone für mobile, kinematisch redundante Manipulatoren definiert. Die Lage und Orientierung der Basis des Manipulators und die Gelenkwinkel werden so gewählt, dass ausgesuchte Zielfunktionen minimal werden. Diese Zielfunktionen werden aus bekannten Maßen wie kartesische Geschwindigkeit, Kraft und Steifigkeit abgeleitet. Sie werden abhängig von den Parametern der Basis und den Gelenkwinkeln des Manipulators bestimmt, wobei nur ein Unterraum an möglichen Konfigurationen aufgrund der vorgegebenen Lage und Orientierung des Endeffektors (EE) für die Optimierung frei wählbar ist. Es werden im Sinne der multikriteriellen Optimierung Pareto-Fronten errechnet, welche durch vordefinierte Grenzen (z.B. minimale erreichbare Kraft oder minimale erreichbare Geschwindigkeit) begrenzt werden. Motivated by an application example, the comfort zone for mobile kinematically redundant manipulators is defined. The position and orientation of the mobile platform (=base of the manipulator) and the joint angles are selected in such a way that selected target functions are minimized. These objective functions are derived from known measures such as Cartesian velocity, force and stiffness. The objective functions are evaluated depending on the parameters of the base and the joint angles of the manipulator, whereby only a subspace of possible configurations is freely selectable for optimization due to the given position and orientation of the end effector (EE). Pareto-fronts are calculated in the sense of multi-criteria optimization, which are limited by, e.g., minimum achievable force or minimum achievable speed. Breath tests allow a non-invasive and fast diagnostic of different specific enzymes' phenotypic functionality. Over the last decade several 13C-breath tests were successfully tested, with the (13)C-urea breath test being approved by the US Food and Drug Administration (FDA) and the European Medicines Agency (EMA). The use of other targets than labeled (13)CO2 in exhaled breath offers additional possibilities. High sensitivity analytical technologies, such as proton-transfer reaction time-of-flight mass spectrometry, enable the detection of different volatile targets in the low ppb (parts per billion) range in real-time.In the current study volunteers received 0.8 mg deuterated 2-propanol, which was converted to d3-acetone (m/z 62.08) by alcohol dehydrogenase. D3-acetone (m/z 62.08) appeared in exhaled breath concentrations up to 30 ppb (at maximum). Parallel consumption of ethanol seems to reduce the activity of the enzyme, resulting in approximately 15-30% reduction of the produced d3-acetone. Phenotypic determination of enzyme activities is important, since the functionality of enzymes is influenced by factors such as age, sex, life-style, diet, organ function, metabolism, etc, which cannot be entirely accounted for by genetic factors.

Small diffusible redox proteins play a ubiquitous role in bioenergetic systems, facilitating electron transfer (ET) between membrane bound complexes. Sustaining high ET turnover rates requires that the association between extrinsic and membrane-bound partners is highly specific, yet also sufficiently weak to promote rapid post-ET separation. In oxygenic photosynthesis the small soluble electron carrier protein plastocyanin (Pc) shuttles electrons between the membrane integral cytochrome b6f (cytb6f) and photosystem I (PSI) complexes. Here we use peak-force quantitative nanomechanical mapping (PF-QNM) atomic force microscopy (AFM) to quantify the dynamic forces involved in transient interactions between cognate ET partners. An AFM probe functionalised with Pc molecules is brought into contact with cytb6f complexes, immobilised on a planar silicon surface. PF-QNM interrogates the unbinding force of the cytb6f-Pc interactions at the single molecule level with picoNewton force resolution and on a time scale comparable to the ET time in vivo (ca. 120 μs). Using this approach, we show that although the unbinding force remains unchanged the interaction frequency increases over five-fold when Pc and cytb6f are in opposite redox states, so complementary charges on the cytb6f and Pc cofactors likely contribute to the electrostatic forces that initiate formation of the ET complex. These results suggest that formation of the docking interface is under redox state control, which lowers the probability of unproductive encounters between Pc and cytb6f molecules in the same redox state, ensuring the efficiency and directionality of this central reaction in the 'Z-scheme' of photosynthetic ET.

Mavroulis, S. in OpenAIREAbstractRecent wildfire events (e.g. Mediterranean region, USA, and Australia) showed that this hazard poses a serious threat for wildland–urban interface (WUI) areas around the globe. Furthermore, recent events in regions where wildfire does not constitute a frequent hazard (e.g. Siberia, Scandinavia) indicated that the spatial pattern of wildfire risk might have significantly changed. To prepare for upcoming extreme events, it is critical for decision-makers to have a thorough understanding of the vulnerability of the built environment to wildfire. Building quality and design standards are important not only because building loss is costly but also because robust buildings may offer shelter when evacuation is not possible. However, studies aiming at the analysis of wildfire vulnerability for the built environment are limited. This paper presents an innovative solution for the vulnerability assessment to wildfires, making use of an all-relevant feature selection algorithm established on statistical relationships to develop a physical vulnerability index for buildings subject to wildfire. Data from a recent and systematically documented wildfire event in Greece (Mati, 2018) are used to select and weight the relevant indicators using a permutation-based automated feature selection based on random forests. Building characteristics including the structural type, the roof type, material and shape, the inclination of the ground, the surrounding vegetation, the material of the shutters and the ground covering were selected and formed into the index. The index may be used in other places in Europe and beyond, especially where no empirical data are available supporting decision-making and risk reduction of an emerging hazard amplified by climate change.
