Dias;Grape stalks, an inedible lignocellulosic residue from winemaking and agro-industrial grape juice production, can be valorized as a source of bioactive compounds and as feedstock for the saccharification and bioconversion of soluble sugars. Solid-state fermentation (SSF) by six white-rot fungi was applied as pretreatment. Fiber composition, free radical scavenging activity, four ligninolytic, and three hydrolytic enzyme activities were determined. Saccharification kinetics, yield, and productivity were evaluated and complemented with scanning electron microscopy (SEM), high performance liquid chromatography (HPLC) quantification of monosaccharides, and principal component analysis (PCA). After SSF, the biomass exhibited a drastic free radical scavenging activity decrease and the main enzymes produced were manganese-dependent peroxidase and xylanase. Scanning electron microscopy revealed the erosion of cell walls, and PCA exhibited a negative correlation between saccharification, and neutral detergent fiber and acid detergent lignin. Phlebia rufa pretreated biomass gave the highest sugars yield and productivity, representing a nearly three-fold increase compared to untreated samples. Also, monosaccharides quantification revealed that the 1:1 ratio of glucose to the sum of xylose plus galactose changes to the value of 2:1 after pretreatment. In this work, and for the first time, P. rufa proved to be an effective pretreatment of grape stalks for the saccharification and further bioconversion into value-added chemicals. In addition, lignocellulolytic enzymes were also produced through SSF.

Their use would address the circular economy associated with the production of low-toxicity biomass and supernatant. Studies in this field are still scarce, and substantial gaps exist in the definitions of the flocculation process parameters. In this context, the objective of this work was to evaluate TBC performance as a natural coagulant for harvesting microalgae biomass grown in sanitary effluent digested in an up flow biofilter, as well establishing a path to enable recovery and reuse of wastewater nutrients. Classical removal techniques combined with image analysis and light scattering-based equipment were used to evaluate the coagulant performance, recovery efficiency, floc strength, and floc recovery compared to aluminum sulfate (AS). The results showed that TBC was able to efficiently harvest algal biomass from the effluent, achieving color, turbidity, and optical density (OD) removal efficiencies greater than 90% with only 5 min of sedimentation. The optimal harvesting dosage was 100 mg·L-1 for TBC and 75 mg·L-1 for AS. TBC presented the advantage of harvesting biomass without changing the pH of the medium and was also able to present satisfactory removal of the analyzed parameters (color, turbidity and OD) at pH values of 5.0, 7.0, and 8.5. In addition, TBC produced stronger flocs than AS, showing a better ability to resist breakage upon sudden shear rate variations. TBC produced macronutrient-rich biomass and supernatant that was similar to that produced with AS.

The conversion of lignocellulosic biomass is a major challenge in the field of renewable energies and bio-based chemicals. The diversity of biomasses and processes leads to complex products having a wide range of polarities and molecular weights. Nowadays, the molecular description of these oxygenated matrices is still largely incomplete and new analytical strategies are required to have a better understanding of biomass products properties. The present study proposes a reliable protocol based on successive liquid-liquid extractions prior to high performance liquid chromatography hyphenated to high-resolution tandem mass spectrometry (HPLC/MSn) using a linear ion trap-Fourier transform ion cyclotron resonance mass spectrometer (LTQ/FT-ICR). The protocol allowed to fractionate an industrial sample coming from the sulfuric acid-based pretreatment of a wheat straw into four key chemical families: carbohydrates, organic acids, phenols and neutral compounds. Each fraction was separately analyzed, which limited matrix effects during mass spectrometry ionization step. Electrospray and atmospheric pressure chemical ionization sources were used in both positive and negative modes in order to ionize and detect a maximum of compounds. Thanks to HPLC/MSn, structures of heavy lignin-carbohydrate complexes (LCC) were elucidated (up to 600 g/mol) as well as carbohydrate oligomers having acid functionalities. Mono, di, tri and tetra-aromatic compounds coming from lignin were also detected. The results reported in this paper demonstrate the complexity of pretreated biomass samples and propose an analytical approach from sample simplification to data treatment in order to describe the biomass composition.

The inoculum was collected from a bioreactor performing simultaneous partial nitrification and aromatic compounds biodegradation. From day-56 onwards, 2-fluorophenol (2-FP) (12.4mgL-1) was added to the feeding wastewater and the system was bioaugmented with a 2-FP degrading bacteria (Rhodococcus sp. FP1). By the end of operation, complete 2-FP biodegradation and partial nitrification were simultaneously achieved. Aerobic granules remained stable over time. During the 2-FP loading, a shift in the community structure occurred, coinciding with the improvement of 2-FP degradation. DGGE analysis did not allow to infer on the bioaugmented strain presence but pyrosequencing analysis detected Rhodococcus genus by the end of operation. Together with other potential phenolic-degraders within granules, these microorganisms were probably responsible for 2-FP degradation.

The adsorption and later bioavailability of 137Cs from the system humic acid (HA)/humic acid like compounds (HALC) and montmorillonite was investigated. The setup of the experiments should approach as much as possible natural conditions when 137Cs is introduced into soil with HALC from decomposed biomass. The significant differences were found in the trials containing various HA/HALC and also pure montmorillonite. The 137Cs was more available when it reached soil in association with HALC originated from compost than when it was adsorbed on stable humic acids. Moreover, the long term interaction of 137Cs with HALC led to decrease of 137Cs adsorbed on montmorillonite and increase of its bioavailable fraction. UV-Vis spectrometry and infrared spectroscopy showed the clear difference between HA, fresh HALC and old HALC which could partially explain the different results.

Ecosystems show the same sublinear scaling between production and total biomass, but ecological theory cannot reconcile the existence of these nearly identical scalings at different levels of biological organization. We attempt to solve this paradox using marine phytoplankton, connecting individual and ecosystem scalings across three orders of magnitude in body size and biomass. We find that competitive interactions determined by biomass slow metabolism in a consistent fashion across species of different sizes. These effects dominate over species-specific peculiarities, explaining why community composition does not affect respiration and production patterns. The sublinear scaling of ecosystem production thus emerges from this metabolic density-dependence that operates across species, independently of the equilibrium state or resource regime. Our findings demonstrate the connection between individual and ecosystem scalings, unifying aspects of physiology and ecology to explain why growth patterns are so strikingly similar across scales.

The use of microalgae and bacteria as a strategy for the startup of bioreactors for the treatment of industrial wastewater can be a sustainable and economically viable alternative. This technology model provides satisfactory results in the nitrification and denitrification process for nitrogen removal, organic matter removal, biomass growth, sedimentation, and byproducts recovery for added-value product production. The objective of this work was to evaluate the performance of microalgae and bacteria in their symbiotic process when used in the treatment of paper pulp industry wastewater. The experiment, lasting fourteen days, utilized four bioreactors with varying concentrations in mgVSS/L of microalgae to bacteria ratio (R1-100:100, R2-100:300, R3-100:500, R4-300:100) in the startup process. Regarding the sludge volumetric index (SVI), the results show that the R1 and R2 reactors developed SVI30/SVI10 biomass in the range of 85.57 ± 7.33% and 84.72 ± 8.19%, respectively. The lipid content in the biomass of reactors R1, R2, R3 e R4 was 13%, 7%, 19%, and 22%, respectively. This high oil content at the end of the batch, may be related to the nutritional stress that the species underwent during this feeding regime. In terms of chlorophyll, the bioreactor with an initial inoculation of 100:100 showed better symbiotic growth of microalgae and bacteria, allowing exponential growth of microalgae. The total chlorophyll value for this bioreactor was 801.46 ± 196.96 μg/L. Biological removal of nitrogen from wastewater from the paper pulp industry is a challenge due to the characteristics of the effluent, but the four reactors operated in a single batch obtained good nitrogen removal. Suspended marine benthic microalgae in the water column reflect the close relationship between the benthic and pelagic components of coastal ecosystems. In this study, a 12‐year phytoplankton time‐series was used to investigate the contribution of benthic microalgae to the pelagic system at a site along the French‐Atlantic coast. Furthermore, all taxa identified were allocated into different growth forms in order to study their seasonal patterns. The highest contribution of benthic microalgae was observed during the winter period, reaching up to 60% of the carbon biomass in the water column. The haptobenthic growth form showed the highest contribution in terms of biomass, dominant in the fall‐winter period when the turbidity and the river flow were high. The epipelic growth form did not follow any seasonal pattern. The epiphytic diatom Licmophora was most commonly found during summer. As benthic microalgae were found in the water column throughout the year, the temporal variation detected in the structure of pelagic assemblages in a macrotidal ecosystem was partly derived from the differentiated contribution of several benthic growth forms.

Home respiratory care (HRC) is the provision of healthcare services at the place of residence of patients or their families, with the aim of meeting needs mainly resulting from chronic respiratory conditions, permanent disability, or terminal illness. In 2016, an innovative electronic prescription system, PEM-CRD, was fully implemented for HRC services in Portugal. To date, no study has addressed the impact of the execution of this digital innovation. For this purpose, we carried out an analysis of the prevalence and number of prescriptions for people with chronic respiratory diseases receiving HRC in the Lisbon metropolitan area, during 2014–2018, using the information obtained from the PEM-CRD database. The data analysis shows that while the number of patients receiving HRC treatment with a prescription has remained stable over the last four years, the number of prescriptions has significantly dropped since 2016 (2016–2018), with consequent paper and processes efficiency. The implementation of the digital Medical Electronic Prescription for Home Respiratory Care tool (PEM-CRD) and consequent dematerialization of these processes has increased the efficiency of prescribing in HRC. Additionally, the possibility of obtaining data through the PEM-CRD allows the monitoring of the evolving prevalence of therapies, improving the health services optimization and allowing reporting on data other than medicines.

Dias;Grape stalks, an inedible lignocellulosic residue from winemaking and agro-industrial grape juice production, can be valorized as a source of bioactive compounds and as feedstock for the saccharification and bioconversion of soluble sugars. Solid-state fermentation (SSF) by six white-rot fungi was applied as pretreatment. Fiber composition, free radical scavenging activity, four ligninolytic, and three hydrolytic enzyme activities were determined. Saccharification kinetics, yield, and productivity were evaluated and complemented with scanning electron microscopy (SEM), high performance liquid chromatography (HPLC) quantification of monosaccharides, and principal component analysis (PCA). After SSF, the biomass exhibited a drastic free radical scavenging activity decrease and the main enzymes produced were manganese-dependent peroxidase and xylanase. Scanning electron microscopy revealed the erosion of cell walls, and PCA exhibited a negative correlation between saccharification, and neutral detergent fiber and acid detergent lignin. Phlebia rufa pretreated biomass gave the highest sugars yield and productivity, representing a nearly three-fold increase compared to untreated samples. Also, monosaccharides quantification revealed that the 1:1 ratio of glucose to the sum of xylose plus galactose changes to the value of 2:1 after pretreatment. In this work, and for the first time, P. rufa proved to be an effective pretreatment of grape stalks for the saccharification and further bioconversion into value-added chemicals. In addition, lignocellulolytic enzymes were also produced through SSF.

Their use would address the circular economy associated with the production of low-toxicity biomass and supernatant. Studies in this field are still scarce, and substantial gaps exist in the definitions of the flocculation process parameters. In this context, the objective of this work was to evaluate TBC performance as a natural coagulant for harvesting microalgae biomass grown in sanitary effluent digested in an up flow biofilter, as well establishing a path to enable recovery and reuse of wastewater nutrients. Classical removal techniques combined with image analysis and light scattering-based equipment were used to evaluate the coagulant performance, recovery efficiency, floc strength, and floc recovery compared to aluminum sulfate (AS). The results showed that TBC was able to efficiently harvest algal biomass from the effluent, achieving color, turbidity, and optical density (OD) removal efficiencies greater than 90% with only 5 min of sedimentation. The optimal harvesting dosage was 100 mg·L-1 for TBC and 75 mg·L-1 for AS. TBC presented the advantage of harvesting biomass without changing the pH of the medium and was also able to present satisfactory removal of the analyzed parameters (color, turbidity and OD) at pH values of 5.0, 7.0, and 8.5. In addition, TBC produced stronger flocs than AS, showing a better ability to resist breakage upon sudden shear rate variations. TBC produced macronutrient-rich biomass and supernatant that was similar to that produced with AS.

The conversion of lignocellulosic biomass is a major challenge in the field of renewable energies and bio-based chemicals. The diversity of biomasses and processes leads to complex products having a wide range of polarities and molecular weights. Nowadays, the molecular description of these oxygenated matrices is still largely incomplete and new analytical strategies are required to have a better understanding of biomass products properties. The present study proposes a reliable protocol based on successive liquid-liquid extractions prior to high performance liquid chromatography hyphenated to high-resolution tandem mass spectrometry (HPLC/MSn) using a linear ion trap-Fourier transform ion cyclotron resonance mass spectrometer (LTQ/FT-ICR). The protocol allowed to fractionate an industrial sample coming from the sulfuric acid-based pretreatment of a wheat straw into four key chemical families: carbohydrates, organic acids, phenols and neutral compounds. Each fraction was separately analyzed, which limited matrix effects during mass spectrometry ionization step. Electrospray and atmospheric pressure chemical ionization sources were used in both positive and negative modes in order to ionize and detect a maximum of compounds. Thanks to HPLC/MSn, structures of heavy lignin-carbohydrate complexes (LCC) were elucidated (up to 600 g/mol) as well as carbohydrate oligomers having acid functionalities. Mono, di, tri and tetra-aromatic compounds coming from lignin were also detected. The results reported in this paper demonstrate the complexity of pretreated biomass samples and propose an analytical approach from sample simplification to data treatment in order to describe the biomass composition.

The inoculum was collected from a bioreactor performing simultaneous partial nitrification and aromatic compounds biodegradation. From day-56 onwards, 2-fluorophenol (2-FP) (12.4mgL-1) was added to the feeding wastewater and the system was bioaugmented with a 2-FP degrading bacteria (Rhodococcus sp. FP1). By the end of operation, complete 2-FP biodegradation and partial nitrification were simultaneously achieved. Aerobic granules remained stable over time. During the 2-FP loading, a shift in the community structure occurred, coinciding with the improvement of 2-FP degradation. DGGE analysis did not allow to infer on the bioaugmented strain presence but pyrosequencing analysis detected Rhodococcus genus by the end of operation. Together with other potential phenolic-degraders within granules, these microorganisms were probably responsible for 2-FP degradation.

The adsorption and later bioavailability of 137Cs from the system humic acid (HA)/humic acid like compounds (HALC) and montmorillonite was investigated. The setup of the experiments should approach as much as possible natural conditions when 137Cs is introduced into soil with HALC from decomposed biomass. The significant differences were found in the trials containing various HA/HALC and also pure montmorillonite. The 137Cs was more available when it reached soil in association with HALC originated from compost than when it was adsorbed on stable humic acids. Moreover, the long term interaction of 137Cs with HALC led to decrease of 137Cs adsorbed on montmorillonite and increase of its bioavailable fraction. UV-Vis spectrometry and infrared spectroscopy showed the clear difference between HA, fresh HALC and old HALC which could partially explain the different results.

Ecosystems show the same sublinear scaling between production and total biomass, but ecological theory cannot reconcile the existence of these nearly identical scalings at different levels of biological organization. We attempt to solve this paradox using marine phytoplankton, connecting individual and ecosystem scalings across three orders of magnitude in body size and biomass. We find that competitive interactions determined by biomass slow metabolism in a consistent fashion across species of different sizes. These effects dominate over species-specific peculiarities, explaining why community composition does not affect respiration and production patterns. The sublinear scaling of ecosystem production thus emerges from this metabolic density-dependence that operates across species, independently of the equilibrium state or resource regime. Our findings demonstrate the connection between individual and ecosystem scalings, unifying aspects of physiology and ecology to explain why growth patterns are so strikingly similar across scales.

The use of microalgae and bacteria as a strategy for the startup of bioreactors for the treatment of industrial wastewater can be a sustainable and economically viable alternative. This technology model provides satisfactory results in the nitrification and denitrification process for nitrogen removal, organic matter removal, biomass growth, sedimentation, and byproducts recovery for added-value product production. The objective of this work was to evaluate the performance of microalgae and bacteria in their symbiotic process when used in the treatment of paper pulp industry wastewater. The experiment, lasting fourteen days, utilized four bioreactors with varying concentrations in mgVSS/L of microalgae to bacteria ratio (R1-100:100, R2-100:300, R3-100:500, R4-300:100) in the startup process. Regarding the sludge volumetric index (SVI), the results show that the R1 and R2 reactors developed SVI30/SVI10 biomass in the range of 85.57 ± 7.33% and 84.72 ± 8.19%, respectively. The lipid content in the biomass of reactors R1, R2, R3 e R4 was 13%, 7%, 19%, and 22%, respectively. This high oil content at the end of the batch, may be related to the nutritional stress that the species underwent during this feeding regime. In terms of chlorophyll, the bioreactor with an initial inoculation of 100:100 showed better symbiotic growth of microalgae and bacteria, allowing exponential growth of microalgae. The total chlorophyll value for this bioreactor was 801.46 ± 196.96 μg/L. Biological removal of nitrogen from wastewater from the paper pulp industry is a challenge due to the characteristics of the effluent, but the four reactors operated in a single batch obtained good nitrogen removal. Ammonia nitrogen removal performances were 91.55 ± 9.99%, 72.13 ± 19.18%, 64.04 ± 21.34%, and 86.15 ± 30.10% in R1, R2, R3, and R4, respectively.

Suspended marine benthic microalgae in the water column reflect the close relationship between the benthic and pelagic components of coastal ecosystems. In this study, a 12‐year phytoplankton time‐series was used to investigate the contribution of benthic microalgae to the pelagic system at a site along the French‐Atlantic coast. Furthermore, all taxa identified were allocated into different growth forms in order to study their seasonal patterns. The highest contribution of benthic microalgae was observed during the winter period, reaching up to 60% of the carbon biomass in the water column. The haptobenthic growth form showed the highest contribution in terms of biomass, dominant in the fall‐winter period when the turbidity and the river flow were high. The epipelic growth form did not follow any seasonal pattern. The epiphytic diatom Licmophora was most commonly found during summer. As benthic microalgae were found in the water column throughout the year, the temporal variation detected in the structure of pelagic assemblages in a macrotidal ecosystem was partly derived from the differentiated contribution of several benthic growth forms.

Home respiratory care (HRC) is the provision of healthcare services at the place of residence of patients or their families, with the aim of meeting needs mainly resulting from chronic respiratory conditions, permanent disability, or terminal illness. In 2016, an innovative electronic prescription system, PEM-CRD, was fully implemented for HRC services in Portugal. To date, no study has addressed the impact of the execution of this digital innovation. For this purpose, we carried out an analysis of the prevalence and number of prescriptions for people with chronic respiratory diseases receiving HRC in the Lisbon metropolitan area, during 2014–2018, using the information obtained from the PEM-CRD database. The data analysis shows that while the number of patients receiving HRC treatment with a prescription has remained stable over the last four years, the number of prescriptions has significantly dropped since 2016 (2016–2018), with consequent paper and processes efficiency. The implementation of the digital Medical Electronic Prescription for Home Respiratory Care tool (PEM-CRD) and consequent dematerialization of these processes has increased the efficiency of prescribing in HRC. Additionally, the possibility of obtaining data through the PEM-CRD allows the monitoring of the evolving prevalence of therapies, improving the health services optimization and allowing reporting on data other than medicines.
