Elevated temperatures associated with future warmer climates could affect mosquitoes' metabolic enzyme expression and increase insecticide resistance, making vector control difficult. Understanding how mosquito rearing temperatures influence their susceptibility to insecticide and expression of metabolic enzymes could aid in the development of novel tools and strategies to control mosquitoes in a future warmer climate. This study evaluated the effects of temperature on the susceptibility of Anopheles gambiae sensu lato (s.l.) mosquitoes to pyrethroids and their expression of metabolic enzymes. Methods Anopheles gambiae s.l. eggs obtained from laboratory-established colonies were reared under eight temperature regimes (25, 28, 30, 32, 34, 36, 38, and 40 °C). Upon adult emergence, 3- to 5-day-old female non-blood-fed mosquitoes were used for susceptibility tests following the World Health Organization (WHO) bioassay protocol. Batches of 20–25 mosquitoes from each temperature regime (25–34 °C) were exposed to two pyrethroid insecticides (0.75% permethrin and 0.05% deltamethrin). In addition, the levels of four metabolic enzymes (α-esterase, β-esterase, glutathione S-transferase [GST], and mixed-function oxidase [MFO]) were examined in mosquitoes that were not exposed and those that were exposed to pyrethroids. Results Mortality in An. gambiae s.l. mosquitoes exposed to deltamethrin and permethrin decreased at temperatures above 28 °C. In addition, mosquitoes reared at higher temperatures were more resistant and had more elevated enzyme levels than those raised at low temperatures. Overall, mosquitoes that survived after being exposed to pyrethroids had higher levels of metabolic enzymes than those that were not exposed to pyrethroids. Conclusions This study provides evidence that elevated temperatures decreased An. gambiae s.l. mosquitoes' susceptibility to pyrethroids and increased the expression of metabolic enzymes. This evidence suggests that elevated temperatures projected in a future warmer climate could increase mosquitoes' resistance to insecticides and complicate malaria vector control measures. This study therefore provides vital information, and suggests useful areas of future research, on the effects of temperature variability on mosquitoes that could guide vector control measures in a future warmer climate. However, the practical complexities of localised humanitarian aid are yet to be fully discussed. This paper aims to examine the concept of localisation in humanitarian aid through a case study of a local non-governmental organisation, the Cambodian Children's Trust (CCT), in Cambodia. Drawing on the framework of localisation proposed by Baguios et al., (2021), we analyse the application of localisation of child protection programs run by CCT. We provide a holistic understanding of how localisation is conducted in practice, and the impacts it has on the wider humanitarian sector. Our study illustrates that power can be effectively developed to local entities and localisation with empowerment could be achieved despite highly challenging conditions.

Desorption kinetics characterization was carried out using the volumetric method, specifically employing a Sievert's-type apparatus to investigate solid-gas reactions. The experiments covered a temperature range from 313 K to 353 K, with an initial hydrogen pressure of 12 bar. Simultaneously, a mathematical approach was employed to numerically investigate the temperature evolution within the hydride bed. Using COMSOL Multiphysics as a simulator, a numerical simulation was conducted based on experimental data. The study examined the impact of cooling temperature on hydride temperature evolution. Results revealed that hydrogen desorption kinetics of the amorphous Mg50Ni50 alloy are more significant compared to those of Mg2Ni compounds. Moreover, the effect of the warming temperature on the equilibrium pressure can also be observed in the hydrogen desorption isotherm curves. The experimental study of the Mg50Ni50 alloy provided activation energy data, along with determination of hydride formation enthalpy and entropy. On the other hand, we showed that the hydride temperature is maximum at the hydride-hydrogen interface within the hydride center.

Oil sludge from various objects of oil fields in Kazakhstan (Mangystau region) has been studied. The possibilities of using oil (after its separation from oil sludge by bioremidiation) as a partial substitute for bitumen base in the production of modified bitumen are considered. The main physical and mechanical characteristics of modified bitumen are determined. The results confirm that the modified bitumen prepared with oil sludge and oil separated by bioremiation method meets the requirements for polymer-bitumen binder to Kazakhstan standards and is suitable for the production of modified bitumen in its physico-chemical characteristics.

Carroll; M.M. Smith;Abstract Fractures along interfaces between host rock and wellbore cement have long been identified as potential CO2 leakage pathways from subsurface CO2 storage sites. As a consequence, cement alteration due to exposure to CO2 has been studied extensively to assess wellbore integrity. Previous studies have focused on the changes to either chemical or mechanical properties of cement upon exposure to CO2-enriched brine, but not on the effects of loading conditions. This paper aims to correct this deficit by considering the combined effects of the fracture pathway and changing effective stress on chemical and mechanical degradation at conditions relevant to geologic carbon storage. Flow-through experiments on fractured cores composed of cement and tight sandstone caprock halves were conducted to study the alteration of cement due to exposure to CO2-enriched brine at 3, 7, 9, and 12 MPa effective stress. We characterized relevant reactions via solution chemistry; fracture permeability via changes to differential pressure; mechanical changes via micro-hardness testing, and pore structure changes via x-ray tomography. This study showed that the nature and the rates of the chemical reactions between cement and CO2 were not affected by the effective stress. The differences in the permeability responses of the fractures were attributed to interactions among the geometry of the flow path, the porosity increase of the reacted cement, and the mechanical deformation of reacted asperities. The suite of observed chemical reactions contributed to change in cement mechanical properties. Compared to the unreacted cement, the average hardness of the amorphous silica and depleted layers was decreased while the hardness of the calcite layer was increased. Tomographic imaging showed that preferential flow paths formed in some of the core-flood experiments, which had a significant impact on the permeability response of the fractured samples. We interpreted the observed permeability responses in terms of competition between dissolution of cement phases (leading to enhanced permeability) and mechanical deformation of reacted regions (leading to reduced permeability).

Abstract The main hindrances to the large-scale development of renewable-energy projects are the lack of bankability and the inability to align investments and investors with suitable financial instruments or robust policy measures. To illustrate a bankable project, this paper presents a research-based case study on the installation of solar photovoltaic panels on the rooftops of 195 trains of the Indian Railways. Detailed information on the annual running hours, exposure to sunlight, efficiency of solar photovoltaic generation and electrical power demands of each rail coach is considered to conduct a quantitative measure of the tentative amount of fossil fuel savings. The purpose is to provide insight into the types of renewable-energy projects that can be highly attractive to financial institutions and promoters due to their lucrative internal return on investment. As seen in this case study, there are annual savings in diesel of 12 323 088 litres and a CO2 reduction of 32 755 tonnes, with return on investment of 1.3 years. Furthermore, this study conducts a comprehensive analysis of the limitations of existing renewable-energy project financing mechanisms in India. Subsequently, three policy measures are recommended to develop a robust financial mechanism that can effectively meet the needs of investors and investors. These measures include increasing equity injection through a buy-and-hold strategy, providing direct tax benefits to promoters and financing through real-estate investment trusts. The findings are highly relevant to address the challenges associated with bridging the financial gap between access to finance and capital investment in the renewable-energy sector, especially for Asian countries.

Climate change is an emerging global health crisis, disproportionately affecting low- and middle-income countries (LMICs) where health outcomes are increasingly compromised by environmental stressors such as pollution, natural disasters, and human migration. With a focus on promoting health equity, Global Surgery advocates for expanding access to surgical care and enhancing health outcomes, particularly in resource-limited and disaster-affected areas like LMICs. The healthcare industry—and more specifically, surgical care—significantly contributes to the global carbon footprint, primarily through resource-intensive settings, i.e. operating rooms that generate greenhouse gases and substantial medical waste. Therefore, Global Surgery efforts aimed at improving surgical access through an increase in surgical volumes may inadvertently exacerbate health challenges for vulnerable populations by further contributing to environmental degradation. This predicament is particularly pronounced in LMICs, who already suffer from a disproportionate share of the global burden of disease, and where the demand for surgery is rising without corresponding resilient infrastructure. LMICs face a double jeopardy of health inequity coupled with climate vulnerability. As a movement positioned to improve health around the world, Global Surgery has an increasingly significant role in envisioning and ensuring a sustainable future. Global Surgery initiatives must prioritise sustainable infrastructure in both high-income countries (HICs) and LMICs, all while accounting for the unequal polluting contributions between HICs and LMICs and, consequently, moral responsibilities moving forward. Moreover, through targeting upstream causes of poor health at urban and perioperative levels, Global Surgery’s interventions may help to reduce the global burden of disease—avoiding preventable surgeries and their carbon footprints from the outset. Altogether, Global Surgery and climate change are two matters of social justice whose solutions must synergistically centralise the health of both the planet and its most vulnerable people.

Aggregate and limestone mining in San Antonio’s Bexar and Comal counties in Texas, USA, has caused considerable health concerns as of late. Aggregate mining actions can result in localized air quality issues in any neighborhood. Furthermore, heavy truck traffic, hauling, and transportation of the mined material contribute to pollution. In this research, PM species were sampled at four locations north of the San Antonio city limits. The data were collected using a TSI Air Quality Sampler that sampled PM1, PM2.5, PM4, PM10, wind speed, wind direction, temperature, and relative humidity. Continuous data with 1 min averages were recorded during the study period from August to September 2019. The instrument was stationed at every location for a period of 7 days each. The four locations were a ranch, an open field, a residential compound, and an elementary school. PM1 and PM2.5 concentration levels were lower compared to PM10 concentrations at all four studied sites. Our results suggest that PM concentrations are primarily impacted by mining activities. PM species were highest at the residential compound due to its proximity to an active mining area, resulting in deleterious health effects for neighbors living in the vicinity of the sampled site.

Homanics; Candice Contet;AbstractLarge conductance potassium (BK) channels are among the most sensitive molecular targets of ethanol and genetic variations in the channel-forming α subunit have been nominally associated with alcohol use disorders. However, whether the action of ethanol at BK α influences the motivation to drink alcohol remains to be determined. To address this question, we first tested the effect of systemically administered BK channel modulators on voluntary alcohol consumption in C57BL/6J males. Penitrem A (blocker) exerted dose-dependent effects on moderate alcohol intake, while paxilline (blocker) and BMS-204352 (opener) were ineffective. Because pharmacological manipulations are inherently limited by non-specific effects, we then sought to investigate the behavioral relevance of ethanol’s direct interaction with BK α by introducing in the mouse genome a point mutation known to render BK channels insensitive to ethanol while preserving their physiological function. The BK α K361N substitution prevented ethanol from reducing spike threshold in medial habenula neurons. However, it did not alter acute responses to ethanol in vivo, including ataxia, sedation, hypothermia, analgesia, and conditioned place preference. Furthermore, the mutation did not have reproducible effects on alcohol consumption in limited, continuous, or intermittent access home cage two-bottle choice paradigms conducted in both males and females. Notably, in contrast to previous observations made in mice missing BK channel auxiliary β subunits, the BK α K361N substitution had no significant impact on ethanol intake escalation induced by chronic intermittent alcohol vapor inhalation. It also did not affect the metabolic and locomotor consequences of chronic alcohol exposure. Altogether, these data suggest that the direct interaction of ethanol with BK α does not mediate the alcohol-related phenotypes examined here in mice.

Elevated temperatures associated with future warmer climates could affect mosquitoes' metabolic enzyme expression and increase insecticide resistance, making vector control difficult. Understanding how mosquito rearing temperatures influence their susceptibility to insecticide and expression of metabolic enzymes could aid in the development of novel tools and strategies to control mosquitoes in a future warmer climate. This study evaluated the effects of temperature on the susceptibility of Anopheles gambiae sensu lato (s.l.) mosquitoes to pyrethroids and their expression of metabolic enzymes. Methods Anopheles gambiae s.l. eggs obtained from laboratory-established colonies were reared under eight temperature regimes (25, 28, 30, 32, 34, 36, 38, and 40 °C). Upon adult emergence, 3- to 5-day-old female non-blood-fed mosquitoes were used for susceptibility tests following the World Health Organization (WHO) bioassay protocol. Batches of 20–25 mosquitoes from each temperature regime (25–34 °C) were exposed to two pyrethroid insecticides (0.75% permethrin and 0.05% deltamethrin). In addition, the levels of four metabolic enzymes (α-esterase, β-esterase, glutathione S-transferase [GST], and mixed-function oxidase [MFO]) were examined in mosquitoes that were not exposed and those that were exposed to pyrethroids. Results Mortality in An. gambiae s.l. mosquitoes exposed to deltamethrin and permethrin decreased at temperatures above 28 °C. In addition, mosquitoes reared at higher temperatures were more resistant and had more elevated enzyme levels than those raised at low temperatures. Overall, mosquitoes that survived after being exposed to pyrethroids had higher levels of metabolic enzymes than those that were not exposed to pyrethroids. Conclusions This study provides evidence that elevated temperatures decreased An. gambiae s.l. mosquitoes' susceptibility to pyrethroids and increased the expression of metabolic enzymes. This evidence suggests that elevated temperatures projected in a future warmer climate could increase mosquitoes' resistance to insecticides and complicate malaria vector control measures. This study therefore provides vital information, and suggests useful areas of future research, on the effects of temperature variability on mosquitoes that could guide vector control measures in a future warmer climate. However, the practical complexities of localised humanitarian aid are yet to be fully discussed. This paper aims to examine the concept of localisation in humanitarian aid through a case study of a local non-governmental organisation, the Cambodian Children's Trust (CCT), in Cambodia. Drawing on the framework of localisation proposed by Baguios et al., (2021), we analyse the application of localisation of child protection programs run by CCT. We provide a holistic understanding of how localisation is conducted in practice, and the impacts it has on the wider humanitarian sector. Our study illustrates that power can be effectively developed to local entities and localisation with empowerment could be achieved despite highly challenging conditions.

Desorption kinetics characterization was carried out using the volumetric method, specifically employing a Sievert's-type apparatus to investigate solid-gas reactions. The experiments covered a temperature range from 313 K to 353 K, with an initial hydrogen pressure of 12 bar. Simultaneously, a mathematical approach was employed to numerically investigate the temperature evolution within the hydride bed. Using COMSOL Multiphysics as a simulator, a numerical simulation was conducted based on experimental data. The study examined the impact of cooling temperature on hydride temperature evolution. Results revealed that hydrogen desorption kinetics of the amorphous Mg50Ni50 alloy are more significant compared to those of Mg2Ni compounds. Moreover, the effect of the warming temperature on the equilibrium pressure can also be observed in the hydrogen desorption isotherm curves. The experimental study of the Mg50Ni50 alloy provided activation energy data, along with determination of hydride formation enthalpy and entropy. On the other hand, we showed that the hydride temperature is maximum at the hydride-hydrogen interface within the hydride center.

Oil sludge from various objects of oil fields in Kazakhstan (Mangystau region) has been studied. The possibilities of using oil (after its separation from oil sludge by bioremidiation) as a partial substitute for bitumen base in the production of modified bitumen are considered. The main physical and mechanical characteristics of modified bitumen are determined. The results confirm that the modified bitumen prepared with oil sludge and oil separated by bioremiation method meets the requirements for polymer-bitumen binder to Kazakhstan standards and is suitable for the production of modified bitumen in its physico-chemical characteristics.

Carroll; M.M. Smith;Abstract Fractures along interfaces between host rock and wellbore cement have long been identified as potential CO2 leakage pathways from subsurface CO2 storage sites. As a consequence, cement alteration due to exposure to CO2 has been studied extensively to assess wellbore integrity. Previous studies have focused on the changes to either chemical or mechanical properties of cement upon exposure to CO2-enriched brine, but not on the effects of loading conditions. This paper aims to correct this deficit by considering the combined effects of the fracture pathway and changing effective stress on chemical and mechanical degradation at conditions relevant to geologic carbon storage. Flow-through experiments on fractured cores composed of cement and tight sandstone caprock halves were conducted to study the alteration of cement due to exposure to CO2-enriched brine at 3, 7, 9, and 12 MPa effective stress. We characterized relevant reactions via solution chemistry; fracture permeability via changes to differential pressure; mechanical changes via micro-hardness testing, and pore structure changes via x-ray tomography. This study showed that the nature and the rates of the chemical reactions between cement and CO2 were not affected by the effective stress. The differences in the permeability responses of the fractures were attributed to interactions among the geometry of the flow path, the porosity increase of the reacted cement, and the mechanical deformation of reacted asperities. The suite of observed chemical reactions contributed to change in cement mechanical properties. Compared to the unreacted cement, the average hardness of the amorphous silica and depleted layers was decreased while the hardness of the calcite layer was increased. Tomographic imaging showed that preferential flow paths formed in some of the core-flood experiments, which had a significant impact on the permeability response of the fractured samples. We interpreted the observed permeability responses in terms of competition between dissolution of cement phases (leading to enhanced permeability) and mechanical deformation of reacted regions (leading to reduced permeability).

Abstract The main hindrances to the large-scale development of renewable-energy projects are the lack of bankability and the inability to align investments and investors with suitable financial instruments or robust policy measures. To illustrate a bankable project, this paper presents a research-based case study on the installation of solar photovoltaic panels on the rooftops of 195 trains of the Indian Railways. Detailed information on the annual running hours, exposure to sunlight, efficiency of solar photovoltaic generation and electrical power demands of each rail coach is considered to conduct a quantitative measure of the tentative amount of fossil fuel savings. The purpose is to provide insight into the types of renewable-energy projects that can be highly attractive to financial institutions and promoters due to their lucrative internal return on investment. As seen in this case study, there are annual savings in diesel of 12 323 088 litres and a CO2 reduction of 32 755 tonnes, with return on investment of 1.3 years. Furthermore, this study conducts a comprehensive analysis of the limitations of existing renewable-energy project financing mechanisms in India. Subsequently, three policy measures are recommended to develop a robust financial mechanism that can effectively meet the needs of investors and investors. These measures include increasing equity injection through a buy-and-hold strategy, providing direct tax benefits to promoters and financing through real-estate investment trusts. The findings are highly relevant to address the challenges associated with bridging the financial gap between access to finance and capital investment in the renewable-energy sector, especially for Asian countries.

Climate change is an emerging global health crisis, disproportionately affecting low- and middle-income countries (LMICs) where health outcomes are increasingly compromised by environmental stressors such as pollution, natural disasters, and human migration. With a focus on promoting health equity, Global Surgery advocates for expanding access to surgical care and enhancing health outcomes, particularly in resource-limited and disaster-affected areas like LMICs. The healthcare industry—and more specifically, surgical care—significantly contributes to the global carbon footprint, primarily through resource-intensive settings, i.e. operating rooms that generate greenhouse gases and substantial medical waste. Therefore, Global Surgery efforts aimed at improving surgical access through an increase in surgical volumes may inadvertently exacerbate health challenges for vulnerable populations by further contributing to environmental degradation. This predicament is particularly pronounced in LMICs, who already suffer from a disproportionate share of the global burden of disease, and where the demand for surgery is rising without corresponding resilient infrastructure. LMICs face a double jeopardy of health inequity coupled with climate vulnerability. As a movement positioned to improve health around the world, Global Surgery has an increasingly significant role in envisioning and ensuring a sustainable future. Global Surgery initiatives must prioritise sustainable infrastructure in both high-income countries (HICs) and LMICs, all while accounting for the unequal polluting contributions between HICs and LMICs and, consequently, moral responsibilities moving forward. Moreover, through targeting upstream causes of poor health at urban and perioperative levels, Global Surgery’s interventions may help to reduce the global burden of disease—avoiding preventable surgeries and their carbon footprints from the outset. Altogether, Global Surgery and climate change are two matters of social justice whose solutions must synergistically centralise the health of both the planet and its most vulnerable people.

Aggregate and limestone mining in San Antonio’s Bexar and Comal counties in Texas, USA, has caused considerable health concerns as of late. Aggregate mining actions can result in localized air quality issues in any neighborhood. Furthermore, heavy truck traffic, hauling, and transportation of the mined material contribute to pollution. In this research, PM species were sampled at four locations north of the San Antonio city limits. The data were collected using a TSI Air Quality Sampler that sampled PM1, PM2.5, PM4, PM10, wind speed, wind direction, temperature, and relative humidity. Continuous data with 1 min averages were recorded during the study period from August to September 2019. The instrument was stationed at every location for a period of 7 days each. The four locations were a ranch, an open field, a residential compound, and an elementary school. PM1 and PM2.5 concentration levels were lower compared to PM10 concentrations at all four studied sites. Our results suggest that PM concentrations are primarily impacted by mining activities. PM species were highest at the residential compound due to its proximity to an active mining area, resulting in deleterious health effects for neighbors living in the vicinity of the sampled site.

Homanics; Candice Contet;AbstractLarge conductance potassium (BK) channels are among the most sensitive molecular targets of ethanol and genetic variations in the channel-forming α subunit have been nominally associated with alcohol use disorders. However, whether the action of ethanol at BK α influences the motivation to drink alcohol remains to be determined. To address this question, we first tested the effect of systemically administered BK channel modulators on voluntary alcohol consumption in C57BL/6J males. Penitrem A (blocker) exerted dose-dependent effects on moderate alcohol intake, while paxilline (blocker) and BMS-204352 (opener) were ineffective. Because pharmacological manipulations are inherently limited by non-specific effects, we then sought to investigate the behavioral relevance of ethanol’s direct interaction with BK α by introducing in the mouse genome a point mutation known to render BK channels insensitive to ethanol while preserving their physiological function. The BK α K361N substitution prevented ethanol from reducing spike threshold in medial habenula neurons. However, it did not alter acute responses to ethanol in vivo, including ataxia, sedation, hypothermia, analgesia, and conditioned place preference. Furthermore, the mutation did not have reproducible effects on alcohol consumption in limited, continuous, or intermittent access home cage two-bottle choice paradigms conducted in both males and females. Notably, in contrast to previous observations made in mice missing BK channel auxiliary β subunits, the BK α K361N substitution had no significant impact on ethanol intake escalation induced by chronic intermittent alcohol vapor inhalation. It also did not affect the metabolic and locomotor consequences of chronic alcohol exposure. Altogether, these data suggest that the direct interaction of ethanol with BK α does not mediate the alcohol-related phenotypes examined here in mice.
