    The continuous increase in the world energy and chemicals demand requires the development of sustainable alternatives to non-renewable sources of energy. Biomass facilities and biorefineries represent interesting options to gradually replace the present industry based on fossil fuels. Lignocellulose is the most promising feedstock to be used in biorefineries. From a sugar platform perspective, a wide range of fuels and chemicals can be obtained via microbial fermentation processes, being ethanol the most significant lignocellulose-derived fuel. Before fermentation, lignocellulose must be pretreated to overcome its inherent recalcitrant structure and obtain the fermentable sugars. Usually, harsh conditions are required for pretreatment of lignocellulose, producing biomass degradation and releasing different compounds that are inhibitors of the hydrolytic enzymes and fermenting microorganisms. Moreover, the lignin polymer that remains in pretreated materials also affects biomass conversion by limiting the enzymatic hydrolysis. The use of laccases has been considered as a very powerful tool for delignification and detoxification of pretreated lignocellulosic materials, boosting subsequent saccharification and fermentation processes. This review compiles the latest studies about the application of laccases as useful and environmentally friendly delignification and detoxification technology, highlighting the main challenges and possible ways to make possible the integration of these enzymes in future lignocellulose-based industries.

    AbstractOperating the saccharification and fermentation processes at high‐substrate loadings is a key factor for making ethanol production from lignocellulosic biomass economically viable. However, increasing the substrate loading presents some disadvantages, including a higher concentration of inhibitors (furan derivatives, weak acids, and phenolic compounds) in the media, which negatively affect the fermentation performance. One strategy to eliminate soluble inhibitors is filtering and washing the pretreated material. In this study, it was observed that even if the material was previously washed, inhibitory compounds were released during the enzymatic hydrolysis step. Laccase enzymatic treatment was evaluated as a method to reduce these inhibitory effects. The laccase efficiency was analyzed in a presaccharification and simultaneous saccharification and fermentation process at high‐substrate loadings. Water‐insoluble solids fraction from steam‐exploded wheat straw was used as substrate and Saccharomyces cerevisiae as fermenting microorganism. Laccase supplementation reduced strongly the phenolic content in the media, without affecting weak acids and furan derivatives. This strategy resulted in an improved yeast performance during simultaneous saccharification and fermentation process, increasing significantly ethanol productivity. © 2012 American Institute of Chemical Engineers Biotechnol.     The present work reviews the role of biogas as advanced biofuel in the renewable energy system, summarizing the main raw materials used for biogas production and the most common technologies for biogas upgrading and delving into emerging biological methanation processes. In addition, it provides a description of current European legislative framework and the potential biomethane business models as well as the main biogas production issues to be addressed to fully deploy these upgrading technologies. Biomethane could be competitive due to negative or zero waste feedstock prices, and competitive to fossil fuels in the transport sector and power generation if upgrading technologies become cheaper and environmentally sustainable.

    Nowadays, the transport sector is one of the main sources of greenhouse gas (GHG) emissions and air pollution in cities. The use of renewable energies is therefore imperative to improve the environmental sustainability of this sector. In this regard, biofuels play an important role as they can be blended directly with fossil fuels and used in traditional vehicles’ engines. Bioethanol is the most used biofuel worldwide and can replace gasoline or form different gasoline-ethanol blends. Additionally, it is an important building block to obtain different high added-value compounds (e.g., acetaldehyde, ethylene, 1,3-butadiene, ethyl acetate). Today, bioethanol is mainly produced from food crops (first-generation (1G) biofuels), and a transition to the production of the so-called advanced ethanol (obtained from lignocellulosic feedstocks, non-food crops, or industrial waste and residue streams) is needed to meet sustainability criteria and to have a better GHG balance. This work gives an overview of the current production, use, and regulation rules of bioethanol as a fuel, as well as the advanced processes and the co-products that can be produced together with bioethanol in a biorefinery context. Special attention is given to the opportunities for making a sustainable transition from bioethanol 1G to advanced bioethanol.

    AbstractLignocellulosic ethanol production requires high substrate concentrations for its cost-competitiveness. This implies the presence of high concentrations of insoluble solids (IS) at the initial stages of the process, which may limit the fermentation performance of the corresponding microorganism. The presence of 40–60% IS (w/w) resulted in lower glucose consumption rates and reduced ethanol volumetric productivities of Saccharomyces cerevisiae F12. Yeast cells exposed to IS exhibited a wrinkled cell surface and a reduced mean cell size due to cavity formation. In addition, the intracellular levels of reactive oxygen species (ROS) increased up to 40%. These ROS levels increased up to 70% when both lignocellulose-derived inhibitors and IS were simultaneously present. The general stress response mechanisms (e.g. DDR2, TPS1 or ZWF1 genes, trehalose and glycogen biosynthesis, and DNA repair mechanisms) were found repressed, and ROS formation could not be counteracted by the induction of the genes involved in repairing the oxidative damage such as glutathione, thioredoxin and methionine scavenging systems (e.g. CTA1, GRX4, MXR1, and TSA1; and the repression of cell cycle progression, CLN3). Overall, these results clearly show the role of IS as an important microbial stress factor that affect yeast cells at physical, physiological, and molecular levels.

    The present work evaluates a two-step pretreatment process based on steam explosion and extrusion technologies for the optimal fractionation of lignocellulosic biomass. Two-step pretreatment of barley straw resulted in overall glucan, hemicellulose and lignin recovery yields of 84%, 91% and 87%, respectively. Precipitation of the collected lignin-rich liquid fraction yielded a solid residue with high lignin content, offering possibilities for subsequent applications. Moreover, hydrolysability tests showed almost complete saccharification of the pretreated solid residue, which when combined with the low concentration of the generated inhibitory compounds, is representative of a good pretreatment approach. Scheffersomyces stipitis was capable of fermenting all of the glucose and xylose from the non-diluted hemicellulose fraction, resulting in an ethanol concentration of 17.5 g/L with 0.34 g/g yields. Similarly, Saccharomyces cerevisiae produced about 4% (v/v) ethanol concentration with 0.40 g/g yields, during simultaneous saccharification and fermentation (SSF) of the two-step pretreated solid residue at 10% (w/w) consistency. These results increased the overall conversion yields from a one-step steam explosion pretreatment by 1.4-fold, showing the effectiveness of including an extrusion step to enhance overall biomass fractionation and carbohydrates conversion via microbial fermentation processes.

    In this work, laccase enzymes were evaluated to detoxify the whole slurry from steam-exploded wheat straw. For it, two different strategies, laccase treatment before or after enzymatic hydrolysis, were employed. The detoxification efficiency was analyzed on enzymatic hydrolysis and fermentation levels by the thermotolerant yeast Kluyveromyces marxianus. Laccases reduced phenolic compounds without affecting weak acids and furan derivates. A lower glucose recovery was observed when laccase treatments were carried out before enzymatic hydrolysis, phenomenon that was not showed after enzymatic hydrolysis. In contrast, both laccase treatment strategies enhanced ethanol concentrations, reducing significantly the lag phase of the yeast and allowing substrate loading increments of saccharification and fermentation broths.

    AbstractBiodiesel, unlike to its fossil-based homologue (diesel), is renewable. Its use contributes to greater sustainability in the energy sector, mainly by reducing greenhouse gas emissions. Current biodiesel production relies on plant- and animal-related feedstocks, resulting in high final costs to the prices of those raw materials. In addition, the production of those materials competes for arable land and has provoked a heated debate involving their use food vs. fuel. As an alternative, single-cell oils (SCOs) obtained from oleaginous microorganisms are attractive sources as a biofuel precursor due to their high lipid content, and composition similar to vegetable oils and animal fats. To make SCOs competitive from an economic point of view, the use of readily available low-cost substrates becomes essential. This work reviews the most recent advances in microbial oil production from non-synthetic sugar-rich media, particularly sugars from lignocellulosic wastes, highlighting the main challenges and prospects for deploying this technology fully in the framework of a Biorefinery concept.

    The complete deployment of a bio-based economy is essential to meet the United Nations' Sustainable Development Goals from the 2030 Agenda. In this context, food waste and lignocellulosic residues are considered low-cost feedstocks for obtaining industrially attractive products through biological processes. The effective conversion of these raw materials is, however, still challenging, since they are recalcitrant to bioprocessing and must be first treated to alter their physicochemical properties and ease the accessibility to their structural components. Among the full pallet of pretreatments, physical methods are recognised to have a high potential to transform food waste and lignocellulosic residues. This review provides a critical discussion about the recent advances on milling, extrusion, ultrasound, and microwave pretreatments. Their mechanisms and modes of application are analysed and the main drawbacks and limitations for their use at an industrial scale are discussed.

    Producción Científica The URBIOFIN demonstration-scale plant constitutes the first integrated biorefinery for the transformation of municipal solid waste (MSW) into new biobased products and a pioneer concept for converting the current linear economy to a more circular economy in the European Union (EU). URBIOFIN, with a total budget of 15 million €, is based on the synergistic action of 16 European companies, universities and research centers, and represents nowadays the most ambitious joint public-private research initiative to create a new model for the management of the organic fraction of MSW (OFMSW). The URBIOFIN biorefinery constitutes a demo-scale plant for the transformation of OFMSW into bioblocks (bioethanol, volatile fatty acids (VFA), and biogas), biopolymers (short-chain (scl-PHA) and medium-chain polyhydroxyalkanoates (mcl-PHA)) and additives (bioethylene and biofertilizers) using a battery of innovative physical-chemical and biological processes configured in a three step approach. In the pretreatment step, MSW must be sorted into OFMSW and non-organics that will be recycled. Subsequently, a fraction of OFMSW is pretreated, hydrolyzed and fermented into bioethanol, which will be catalytically converted into bioethylene (to be used for fruit ripening applications). Alternatively, the other fraction of OFMSW can be partially converted into VFAs, which will be biologically converted into scl-PHAs or elongated with bioethanol and further bioconverted into mcl-PHAs (to be used for packaging and agriculture). Finally, the VFA-free digestate is mixed with the vinasse originated from OFMSW fermentation into bioethanol and anaerobically biotransformed into biogas. Finally, biogas is photosynthetically upgraded to biomethane (to be injected into natural gas grids) or desulfurized and bioconverted into PHAs (for packaging applications) in biogas fermenters based on type II methanotrophs under nutrient-limiting conditions. The microalgae produced during photosynthetic biogas upgrading are further converted into biofertilizer.     The continuous increase in the world energy and chemicals demand requires the development of sustainable alternatives to non-renewable sources of energy. Biomass facilities and biorefineries represent interesting options to gradually replace the present industry based on fossil fuels. Lignocellulose is the most promising feedstock to be used in biorefineries. From a sugar platform perspective, a wide range of fuels and chemicals can be obtained via microbial fermentation processes, being ethanol the most significant lignocellulose-derived fuel. Before fermentation, lignocellulose must be pretreated to overcome its inherent recalcitrant structure and obtain the fermentable sugars. Usually, harsh conditions are required for pretreatment of lignocellulose, producing biomass degradation and releasing different compounds that are inhibitors of the hydrolytic enzymes and fermenting microorganisms. Moreover, the lignin polymer that remains in pretreated materials also affects biomass conversion by limiting the enzymatic hydrolysis. The use of laccases has been considered as a very powerful tool for delignification and detoxification of pretreated lignocellulosic materials, boosting subsequent saccharification and fermentation processes. This review compiles the latest studies about the application of laccases as useful and environmentally friendly delignification and detoxification technology, highlighting the main challenges and possible ways to make possible the integration of these enzymes in future lignocellulose-based industries.

    AbstractOperating the saccharification and fermentation processes at high‐substrate loadings is a key factor for making ethanol production from lignocellulosic biomass economically viable. However, increasing the substrate loading presents some disadvantages, including a higher concentration of inhibitors (furan derivatives, weak acids, and phenolic compounds) in the media, which negatively affect the fermentation performance. One strategy to eliminate soluble inhibitors is filtering and washing the pretreated material. In this study, it was observed that even if the material was previously washed, inhibitory compounds were released during the enzymatic hydrolysis step. Laccase enzymatic treatment was evaluated as a method to reduce these inhibitory effects. The laccase efficiency was analyzed in a presaccharification and simultaneous saccharification and fermentation process at high‐substrate loadings. Water‐insoluble solids fraction from steam‐exploded wheat straw was used as substrate and Saccharomyces cerevisiae as fermenting microorganism. Laccase supplementation reduced strongly the phenolic content in the media, without affecting weak acids and furan derivatives. This strategy resulted in an improved yeast performance during simultaneous saccharification and fermentation process, increasing significantly ethanol productivity. © 2012 American Institute of Chemical Engineers Biotechnol.     The present work reviews the role of biogas as advanced biofuel in the renewable energy system, summarizing the main raw materials used for biogas production and the most common technologies for biogas upgrading and delving into emerging biological methanation processes. In addition, it provides a description of current European legislative framework and the potential biomethane business models as well as the main biogas production issues to be addressed to fully deploy these upgrading technologies. Biomethane could be competitive due to negative or zero waste feedstock prices, and competitive to fossil fuels in the transport sector and power generation if upgrading technologies become cheaper and environmentally sustainable.

    Nowadays, the transport sector is one of the main sources of greenhouse gas (GHG) emissions and air pollution in cities. The use of renewable energies is therefore imperative to improve the environmental sustainability of this sector. In this regard, biofuels play an important role as they can be blended directly with fossil fuels and used in traditional vehicles’ engines. Bioethanol is the most used biofuel worldwide and can replace gasoline or form different gasoline-ethanol blends. Additionally, it is an important building block to obtain different high added-value compounds (e.g., acetaldehyde, ethylene, 1,3-butadiene, ethyl acetate). Today, bioethanol is mainly produced from food crops (first-generation (1G) biofuels), and a transition to the production of the so-called advanced ethanol (obtained from lignocellulosic feedstocks, non-food crops, or industrial waste and residue streams) is needed to meet sustainability criteria and to have a better GHG balance. This work gives an overview of the current production, use, and regulation rules of bioethanol as a fuel, as well as the advanced processes and the co-products that can be produced together with bioethanol in a biorefinery context. Special attention is given to the opportunities for making a sustainable transition from bioethanol 1G to advanced bioethanol.

    AbstractLignocellulosic ethanol production requires high substrate concentrations for its cost-competitiveness. This implies the presence of high concentrations of insoluble solids (IS) at the initial stages of the process, which may limit the fermentation performance of the corresponding microorganism. The presence of 40–60% IS (w/w) resulted in lower glucose consumption rates and reduced ethanol volumetric productivities of Saccharomyces cerevisiae F12. Yeast cells exposed to IS exhibited a wrinkled cell surface and a reduced mean cell size due to cavity formation. In addition, the intracellular levels of reactive oxygen species (ROS) increased up to 40%. These ROS levels increased up to 70% when both lignocellulose-derived inhibitors and IS were simultaneously present. The general stress response mechanisms (e.g. DDR2, TPS1 or ZWF1 genes, trehalose and glycogen biosynthesis, and DNA repair mechanisms) were found repressed, and ROS formation could not be counteracted by the induction of the genes involved in repairing the oxidative damage such as glutathione, thioredoxin and methionine scavenging systems (e.g. CTA1, GRX4, MXR1, and TSA1; and the repression of cell cycle progression, CLN3). Overall, these results clearly show the role of IS as an important microbial stress factor that affect yeast cells at physical, physiological, and molecular levels.

    The present work evaluates a two-step pretreatment process based on steam explosion and extrusion technologies for the optimal fractionation of lignocellulosic biomass. Two-step pretreatment of barley straw resulted in overall glucan, hemicellulose and lignin recovery yields of 84%, 91% and 87%, respectively. Precipitation of the collected lignin-rich liquid fraction yielded a solid residue with high lignin content, offering possibilities for subsequent applications. Moreover, hydrolysability tests showed almost complete saccharification of the pretreated solid residue, which when combined with the low concentration of the generated inhibitory compounds, is representative of a good pretreatment approach. Scheffersomyces stipitis was capable of fermenting all of the glucose and xylose from the non-diluted hemicellulose fraction, resulting in an ethanol concentration of 17.5 g/L with 0.34 g/g yields. Similarly, Saccharomyces cerevisiae produced about 4% (v/v) ethanol concentration with 0.40 g/g yields, during simultaneous saccharification and fermentation (SSF) of the two-step pretreated solid residue at 10% (w/w) consistency. These results increased the overall conversion yields from a one-step steam explosion pretreatment by 1.4-fold, showing the effectiveness of including an extrusion step to enhance overall biomass fractionation and carbohydrates conversion via microbial fermentation processes.

    In this work, laccase enzymes were evaluated to detoxify the whole slurry from steam-exploded wheat straw. For it, two different strategies, laccase treatment before or after enzymatic hydrolysis, were employed. The detoxification efficiency was analyzed on enzymatic hydrolysis and fermentation levels by the thermotolerant yeast Kluyveromyces marxianus. Laccases reduced phenolic compounds without affecting weak acids and furan derivates. A lower glucose recovery was observed when laccase treatments were carried out before enzymatic hydrolysis, phenomenon that was not showed after enzymatic hydrolysis. In contrast, both laccase treatment strategies enhanced ethanol concentrations, reducing significantly the lag phase of the yeast and allowing substrate loading increments of saccharification and fermentation broths.

    AbstractBiodiesel, unlike to its fossil-based homologue (diesel), is renewable. Its use contributes to greater sustainability in the energy sector, mainly by reducing greenhouse gas emissions. Current biodiesel production relies on plant- and animal-related feedstocks, resulting in high final costs to the prices of those raw materials. In addition, the production of those materials competes for arable land and has provoked a heated debate involving their use food vs. fuel. As an alternative, single-cell oils (SCOs) obtained from oleaginous microorganisms are attractive sources as a biofuel precursor due to their high lipid content, and composition similar to vegetable oils and animal fats. To make SCOs competitive from an economic point of view, the use of readily available low-cost substrates becomes essential. This work reviews the most recent advances in microbial oil production from non-synthetic sugar-rich media, particularly sugars from lignocellulosic wastes, highlighting the main challenges and prospects for deploying this technology fully in the framework of a Biorefinery concept.

    The complete deployment of a bio-based economy is essential to meet the United Nations' Sustainable Development Goals from the 2030 Agenda. In this context, food waste and lignocellulosic residues are considered low-cost feedstocks for obtaining industrially attractive products through biological processes. The effective conversion of these raw materials is, however, still challenging, since they are recalcitrant to bioprocessing and must be first treated to alter their physicochemical properties and ease the accessibility to their structural components. Among the full pallet of pretreatments, physical methods are recognised to have a high potential to transform food waste and lignocellulosic residues. This review provides a critical discussion about the recent advances on milling, extrusion, ultrasound, and microwave pretreatments. Their mechanisms and modes of application are analysed and the main drawbacks and limitations for their use at an industrial scale are discussed.

    Producción Científica The URBIOFIN demonstration-scale plant constitutes the first integrated biorefinery for the transformation of municipal solid waste (MSW) into new biobased products and a pioneer concept for converting the current linear economy to a more circular economy in the European Union (EU). URBIOFIN, with a total budget of 15 million €, is based on the synergistic action of 16 European companies, universities and research centers, and represents nowadays the most ambitious joint public-private research initiative to create a new model for the management of the organic fraction of MSW (OFMSW). The URBIOFIN biorefinery constitutes a demo-scale plant for the transformation of OFMSW into bioblocks (bioethanol, volatile fatty acids (VFA), and biogas), biopolymers (short-chain (scl-PHA) and medium-chain polyhydroxyalkanoates (mcl-PHA)) and additives (bioethylene and biofertilizers) using a battery of innovative physical-chemical and biological processes configured in a three step approach. In the pretreatment step, MSW must be sorted into OFMSW and non-organics that will be recycled. Subsequently, a fraction of OFMSW is pretreated, hydrolyzed and fermented into bioethanol, which will be catalytically converted into bioethylene (to be used for fruit ripening applications). Alternatively, the other fraction of OFMSW can be partially converted into VFAs, which will be biologically converted into scl-PHAs or elongated with bioethanol and further bioconverted into mcl-PHAs (to be used for packaging and agriculture). Finally, the VFA-free digestate is mixed with the vinasse originated from OFMSW fermentation into bioethanol and anaerobically biotransformed into biogas. Finally, biogas is photosynthetically upgraded to biomethane (to be injected into natural gas grids) or desulfurized and bioconverted into PHAs (for packaging applications) in biogas fermenters based on type II methanotrophs under nutrient-limiting conditions. The microalgae produced during photosynthetic biogas upgrading are further converted into biofertilizer.
