    Young and aged rats were treated chronically with ethanol or scopolamine. Muscarinic receptors were measured in cerebral cortex, hippocampus and striatum. Following scopolamine treatment muscarinic receptor density in cerebral cortex, hippocampus and striatum of young rats increased by 34, 57 and 27%, respectively; in brains of aged rats the increase was 41% in cerebral cortex, 43% in hippocampus and nil in striatum. Affinity of muscarinic receptors was not changed by scopolamine treatment. Following chronic ethanol administration there was a 48% increase in cortical muscarinic receptor density in young, but not aged rats. The density of muscarinic receptors in hippocampus and striatum of both young and aged rats was not affected by ethanol treatment. Affinity of receptors in hippocampus of aged, ethanol-treated rats was increased compared to age-matched controls. Adaptative responses of the muscarinic receptor/transducer system to neurotransmitter availability are present in both young and aged rats, both the ethanol-induced response is present only in young animals. This suggests differences in the mechanism of action of ethanol and receptor agonists and antagonists in modulating receptor plasticity.

    The uptake of [14C]deoxyglucose by brains of rats that were given alcohol in drinking water for 7 months was investigated. There was a general, approximately 50%, increase in deoxyglucose uptake in brains of ethanol-treated rats with areas of the limbic system being particularly affected.

    AbstractAlcohol use disorder (AUD) is characterized by cycles of abuse, withdrawal, and relapse. Neuroadaptations in the basal ganglia are observed in AUD; specifically in the putamen, globus pallidus (GP), and ventral pallidum (VP). These regions are associated with habit formation, drug‐seeking behaviors, and reward processing. While previous studies have shown the crucial role of glial cells in drug seeking, it remains unknown whether glial cells in the basal ganglia are altered in AUD. Glial cells in the putamen, GP, and VP were examined in human post‐mortem tissue of AUD and alcohol remission cases. Immunohistochemistry was performed to analyze cell count, staining intensity, and morphology of microglia and astrocytes, using markers Iba‐1 and GFAP. Morphological analysis revealed a significant decrease in microglia cell size and process retraction, indicating activation or a dystrophic microglia phenotype in individuals with AUD compared to controls. Microglia staining intensity was also higher in the GP and VP in AUD cases, whereas microglia staining intensity and cell size in remission cases were not different to control cases. In contrast, no astrocyte changes were observed in examined brain regions for both AUD and remission cases compared to controls. These results suggest alcohol exposure alters microglia, potentially contributing to dysfunctions in the basal ganglia that maintain addiction, and abstinence from alcohol may reverse microglia changes and associated dysfunctions. Overall, this study further characterizes AUD neuropathology and implicates microglia in the putamen, GP, and VP as a potential target for therapy.

    AbstractAdolescent intermittent ethanol (AIE) exposure diminishes neurogenesis and dendritic spine density in the dentate gyrus. The cholinesterase inhibitor, donepezil (Aricept), reverses AIE effects on dendritic spines, possibly by interacting with inflammatory and/or epigenetic mediators after AIE exposure. This study tests the hypothesis that donepezil reverses AIE-induced neuroimmune, and epigenetic changes in the adult dentate gyrus. Adolescent Sprague-Dawley male rats (PD30-43) were given 10 intermittent, intragastric doses of ethanol (5.0 g/kg) or isovolumetric water (AIW). Twenty-one days later half of the animals from each group were treated with either donepezil or isovolumetric water (i.g.) once daily for four days. Two hours after the last donepezil or water dose animals were sacrificed and brains prepared for immunohistochemical analyses. AIE reduced immunoreactivity for doublecortin (DCX) and increased immunoreactivity for activated caspase-3 and death receptor-3 in adulthood, suggesting an enduring attenuation of neurogenesis and an increase in progenitor death. These effects were reversed by donepezil treatment in adulthood. AIE also increased immunoreactivity for the inflammatory signaling molecules HMGB1 and RAGE, as well as the activated phosphorylated transcription factor pNFκB p65, and the gene silencing marker dimethylated histone H3K9. All of these AIE effects were also reversed by donepezil, with the exception of HMGB1.

    This study focused on the DNA-binding activity and protein expression of the transcription factors Egr-1 and Egr-3 in the rat brain cortex and hippocampus after chronic or acute ethanol exposure. DNA-binding activity was reduced in both regions after chronic ethanol exposure and was restored to the level of the pair-fed group at 16 h of withdrawal. Cortical Egr-1 protein levels were not altered by chronic ethanol exposure but increased 16 h after withdrawal, thus mirroring DNA-binding activity. In contrast, Egr-3 protein levels did not undergo any change. There was no change in the level of either protein in the hippocampus. Immunohistochemistry revealed a region-selective change in immunopositive cells in the cortex and hippocampus. Finally, an acute bolus dose of ethanol did not affect Egr DNA-binding activity and ethanol treatment did not alter the DNA-binding activity or protein levels of the transcription factor Sp1. These observations suggest that chronic exposure to ethanol has region-selective effects on the DNA-binding activity and protein expression of Egr-1 and Egr-3 transcription factors in the rat brain. These changes occur after prolonged ethanol exposure and may thus reflect neuroadaptive changes associated with physical dependency and withdrawal. These effects are also transcription factor-selective. Clearly, protein expression is not the sole mediator of the changes in DNA-binding activity and chronic ethanol exposure must have effects on modulatory agents of Egr DNA-binding activity.

    Anthropogenic emissions of carbon dioxide in the atmosphere have generated rapid variations in atmospheric composition which drives major climate changes. Climate change related effects include changes in physico-chemical proprieties of sea and freshwater, such as variations in water temperature, salinity, pH/pCO2 and oxygen content, which can impact fish critical physiological functions including reproduction. In this context, the main aim of the present review is to discuss how climate change related effects (variation in water temperature and salinity, increases in duration and frequency of hypoxia events, water acidification) would impact reproduction by affecting the neuroendocrine axis (brain-pituitary-gonad axis). Variations in temperature and photoperiod regimes are known to strongly affect sex differentiation and the timing and phenology of spawning period in several fish species. Temperature mainly acts at the level of gonad by interfering with steroidogenesis, (notably on gonadal aromatase activity) and gametogenesis. Temperature is also directly involved in the quality of released gametes and embryos development. Changes in salinity or water acidification are especially associated with reduction of sperm quality and reproductive output. Hypoxia events are able to interact with gonad steroidogenesis by acting on the steroids precursor cholesterol availability or directly on aromatase action, with an impact on the quality of gametes and reproductive success. Climate change related effects on water parameters likely influence also the reproductive behavior of fish. Although the precise mechanisms underlying the regulation of these effects are not always understood, in this review we discuss different hypothesis and propose future research perspectives.

    1. The activity of manganese-superoxide dismutase (EC1.15.1; SOD) was increased in the livers and kidneys of adult rats after exposure to aqueous ethanol (200 ml/l) for 32 weeks.2. The concentration of Mn in the livers and kidneys was significantly higher after 24 weeks, and by 32 weeks liver copper and zinc levels were lower.3. The activity of foetal (day 19) liver superoxide dismutase was appreciably higher in offspring from dams receiving ethanol during pregnancy. Quantitatively the response appeared to be almost entirely due to the Mn-SOD form of the enzyme.4. Maternal alcoholism during pregnancy had no effect on the levels of Cu, Mn or Zn in foetal (day 19) livers.

    Background:  Neuropathological damage as a result of chronic alcohol abuse often results in the impairment of cognitive function. The damage is particularly marked in the frontal cortex. The 14‐3‐3 protein family consists of 7 proteins, β, γ, ε, ζ, η, θ, and σ, encoded by 7 distinct genes. They are highly conserved molecular chaperones with roles in the regulation of metabolism, signal transduction, cell‐cycle control, protein trafficking, and apoptosis. They may also play an important role in neurodegeneration in chronic alcoholism.Methods:  We used real‐time PCR to measure the expression of 14‐3‐3 mRNA transcripts in both the dorsolateral prefrontal cortex and motor cortex of human brains obtained at autopsy.Results:  We found significantly lower 14‐3‐3β, γ, and θ expression in both cortical areas of alcoholics, but no difference in 14‐3‐3η expression, and higher expression of 14‐3‐3σ in both areas. Levels of 14‐3‐3ζ and ε transcripts were significantly lower only in alcoholic motor cortex.Conclusions:  Altered 14‐3‐3 expression could contribute to synaptic dysfunction and altered neurotransmission in chronic alcohol misuse by human subjects.

    Abstract Administration of p-chlorophenylalanine, a tryptophan hydroxylase inhibitor that selectively depletes brain serotonin, to rats conditioned to discriminate between ethanol and saline, produced complete inhibition of discriminative behaviour. These findings provide evidence for a serotonergic mediation in the discrimination of interoceptive cues produced by ethanol in the central nervous system.

    Young and aged rats were treated chronically with ethanol or scopolamine. Muscarinic receptors were measured in cerebral cortex, hippocampus and striatum. Following scopolamine treatment muscarinic receptor density in cerebral cortex, hippocampus and striatum of young rats increased by 34, 57 and 27%, respectively; in brains of aged rats the increase was 41% in cerebral cortex, 43% in hippocampus and nil in striatum. Affinity of muscarinic receptors was not changed by scopolamine treatment. Following chronic ethanol administration there was a 48% increase in cortical muscarinic receptor density in young, but not aged rats. The density of muscarinic receptors in hippocampus and striatum of both young and aged rats was not affected by ethanol treatment. Affinity of receptors in hippocampus of aged, ethanol-treated rats was increased compared to age-matched controls. Adaptative responses of the muscarinic receptor/transducer system to neurotransmitter availability are present in both young and aged rats, both the ethanol-induced response is present only in young animals. This suggests differences in the mechanism of action of ethanol and receptor agonists and antagonists in modulating receptor plasticity.

    The uptake of [14C]deoxyglucose by brains of rats that were given alcohol in drinking water for 7 months was investigated. There was a general, approximately 50%, increase in deoxyglucose uptake in brains of ethanol-treated rats with areas of the limbic system being particularly affected.

    AbstractAlcohol use disorder (AUD) is characterized by cycles of abuse, withdrawal, and relapse. Neuroadaptations in the basal ganglia are observed in AUD; specifically in the putamen, globus pallidus (GP), and ventral pallidum (VP). These regions are associated with habit formation, drug‐seeking behaviors, and reward processing. While previous studies have shown the crucial role of glial cells in drug seeking, it remains unknown whether glial cells in the basal ganglia are altered in AUD. Glial cells in the putamen, GP, and VP were examined in human post‐mortem tissue of AUD and alcohol remission cases. Immunohistochemistry was performed to analyze cell count, staining intensity, and morphology of microglia and astrocytes, using markers Iba‐1 and GFAP. Morphological analysis revealed a significant decrease in microglia cell size and process retraction, indicating activation or a dystrophic microglia phenotype in individuals with AUD compared to controls. Microglia staining intensity was also higher in the GP and VP in AUD cases, whereas microglia staining intensity and cell size in remission cases were not different to control cases. In contrast, no astrocyte changes were observed in examined brain regions for both AUD and remission cases compared to controls. These results suggest alcohol exposure alters microglia, potentially contributing to dysfunctions in the basal ganglia that maintain addiction, and abstinence from alcohol may reverse microglia changes and associated dysfunctions. Overall, this study further characterizes AUD neuropathology and implicates microglia in the putamen, GP, and VP as a potential target for therapy.

    AbstractAdolescent intermittent ethanol (AIE) exposure diminishes neurogenesis and dendritic spine density in the dentate gyrus. The cholinesterase inhibitor, donepezil (Aricept), reverses AIE effects on dendritic spines, possibly by interacting with inflammatory and/or epigenetic mediators after AIE exposure. This study tests the hypothesis that donepezil reverses AIE-induced neuroimmune, and epigenetic changes in the adult dentate gyrus. Adolescent Sprague-Dawley male rats (PD30-43) were given 10 intermittent, intragastric doses of ethanol (5.0 g/kg) or isovolumetric water (AIW). Twenty-one days later half of the animals from each group were treated with either donepezil or isovolumetric water (i.g.) once daily for four days. Two hours after the last donepezil or water dose animals were sacrificed and brains prepared for immunohistochemical analyses. AIE reduced immunoreactivity for doublecortin (DCX) and increased immunoreactivity for activated caspase-3 and death receptor-3 in adulthood, suggesting an enduring attenuation of neurogenesis and an increase in progenitor death. These effects were reversed by donepezil treatment in adulthood. AIE also increased immunoreactivity for the inflammatory signaling molecules HMGB1 and RAGE, as well as the activated phosphorylated transcription factor pNFκB p65, and the gene silencing marker dimethylated histone H3K9. All of these AIE effects were also reversed by donepezil, with the exception of HMGB1.

    This study focused on the DNA-binding activity and protein expression of the transcription factors Egr-1 and Egr-3 in the rat brain cortex and hippocampus after chronic or acute ethanol exposure. DNA-binding activity was reduced in both regions after chronic ethanol exposure and was restored to the level of the pair-fed group at 16 h of withdrawal. Cortical Egr-1 protein levels were not altered by chronic ethanol exposure but increased 16 h after withdrawal, thus mirroring DNA-binding activity. In contrast, Egr-3 protein levels did not undergo any change. There was no change in the level of either protein in the hippocampus. Immunohistochemistry revealed a region-selective change in immunopositive cells in the cortex and hippocampus. Finally, an acute bolus dose of ethanol did not affect Egr DNA-binding activity and ethanol treatment did not alter the DNA-binding activity or protein levels of the transcription factor Sp1. These observations suggest that chronic exposure to ethanol has region-selective effects on the DNA-binding activity and protein expression of Egr-1 and Egr-3 transcription factors in the rat brain. These changes occur after prolonged ethanol exposure and may thus reflect neuroadaptive changes associated with physical dependency and withdrawal. These effects are also transcription factor-selective. Clearly, protein expression is not the sole mediator of the changes in DNA-binding activity and chronic ethanol exposure must have effects on modulatory agents of Egr DNA-binding activity.

    Anthropogenic emissions of carbon dioxide in the atmosphere have generated rapid variations in atmospheric composition which drives major climate changes. Climate change related effects include changes in physico-chemical proprieties of sea and freshwater, such as variations in water temperature, salinity, pH/pCO2 and oxygen content, which can impact fish critical physiological functions including reproduction. In this context, the main aim of the present review is to discuss how climate change related effects (variation in water temperature and salinity, increases in duration and frequency of hypoxia events, water acidification) would impact reproduction by affecting the neuroendocrine axis (brain-pituitary-gonad axis). Variations in temperature and photoperiod regimes are known to strongly affect sex differentiation and the timing and phenology of spawning period in several fish species. Temperature mainly acts at the level of gonad by interfering with steroidogenesis, (notably on gonadal aromatase activity) and gametogenesis. Temperature is also directly involved in the quality of released gametes and embryos development. Changes in salinity or water acidification are especially associated with reduction of sperm quality and reproductive output. Hypoxia events are able to interact with gonad steroidogenesis by acting on the steroids precursor cholesterol availability or directly on aromatase action, with an impact on the quality of gametes and reproductive success. Climate change related effects on water parameters likely influence also the reproductive behavior of fish. Although the precise mechanisms underlying the regulation of these effects are not always understood, in this review we discuss different hypothesis and propose future research perspectives.

    1. The activity of manganese-superoxide dismutase (EC1.15.1; SOD) was increased in the livers and kidneys of adult rats after exposure to aqueous ethanol (200 ml/l) for 32 weeks.2. The concentration of Mn in the livers and kidneys was significantly higher after 24 weeks, and by 32 weeks liver copper and zinc levels were lower.3. The activity of foetal (day 19) liver superoxide dismutase was appreciably higher in offspring from dams receiving ethanol during pregnancy. Quantitatively the response appeared to be almost entirely due to the Mn-SOD form of the enzyme.4. Maternal alcoholism during pregnancy had no effect on the levels of Cu, Mn or Zn in foetal (day 19) livers.

    Background:  Neuropathological damage as a result of chronic alcohol abuse often results in the impairment of cognitive function. The damage is particularly marked in the frontal cortex. The 14‐3‐3 protein family consists of 7 proteins, β, γ, ε, ζ, η, θ, and σ, encoded by 7 distinct genes. They are highly conserved molecular chaperones with roles in the regulation of metabolism, signal transduction, cell‐cycle control, protein trafficking, and apoptosis. They may also play an important role in neurodegeneration in chronic alcoholism.Methods:  We used real‐time PCR to measure the expression of 14‐3‐3 mRNA transcripts in both the dorsolateral prefrontal cortex and motor cortex of human brains obtained at autopsy.Results:  We found significantly lower 14‐3‐3β, γ, and θ expression in both cortical areas of alcoholics, but no difference in 14‐3‐3η expression, and higher expression of 14‐3‐3σ in both areas. Levels of 14‐3‐3ζ and ε transcripts were significantly lower only in alcoholic motor cortex.Conclusions:  Altered 14‐3‐3 expression could contribute to synaptic dysfunction and altered neurotransmission in chronic alcohol misuse by human subjects.

    Abstract Administration of p-chlorophenylalanine, a tryptophan hydroxylase inhibitor that selectively depletes brain serotonin, to rats conditioned to discriminate between ethanol and saline, produced complete inhibition of discriminative behaviour. These findings provide evidence for a serotonergic mediation in the discrimination of interoceptive cues produced by ethanol in the central nervous system.
