Sampey;Scale-up to industrial production level of a fermentation process occurs after optimization at small scale, a critical transition for successful technology transfer and commercialization of a product of interest. At the large scale a number of important bioprocess engineering problems arise that should be taken into account to match the values obtained at the small scale and achieve the highest productivity and quality possible. However, the changes of the host strain's physiological and metabolic behavior in response to the scale transition are still not clear.Heterogeneity in substrate and oxygen distribution is an inherent factor at industrial scale (10,000 L) which affects the success of process up-scaling. To counteract these detrimental effects, changes in dissolved oxygen and pressure set points and addition of diluents were applied to 10,000 L scale to enable a successful process scale-up. A comprehensive semi-quantitative and time-dependent analysis of the exometabolome was performed to understand the impact of the scale-up on the metabolic/physiological behavior of the host microorganism. Intermediates from central carbon catabolism and mevalonate/ergosterol synthesis pathways were found to accumulate in both the 10 L and 10,000 L scale cultures in a time-dependent manner. Moreover, excreted metabolites analysis revealed that hypoxic conditions prevailed at the 10,000 L scale. The specific product yield increased at the 10,000 L scale, in spite of metabolic stress and catabolic-anabolic uncoupling unveiled by the decrease in biomass yield on consumed oxygen.An optimized S. cerevisiae fermentation process was successfully scaled-up to an industrial scale bioreactor. The oxygen uptake rate (OUR) and overall growth profiles were matched between scales. The major remaining differences between scales were wet cell weight and culture apparent viscosity. The metabolic and physiological behavior of the host microorganism at the 10,000 L scale was investigated with exometabolomics, indicating that reduced oxygen availability affected oxidative phosphorylation cascading into down- and up-stream pathways producing overflow metabolism. Our study revealed striking metabolic and physiological changes in response to hypoxia exerted by industrial bioprocess up-scaling.

The common assumption in potato virus epidemiology is that all daughter tubers produced by plants coming from infected mother tubers (secondary infection) will become infected via systemic translocation of the virus during growth. We hypothesize that depending on the prevalent environmental conditions, only a portion of the daughter tubers of a plant that is secondarily infected by viruses may become infected. To test this hypothesis experimental data from standardized field experiments were produced in three contrasting environments at 112, 3280, and 4000 m a.s.l. in Peru during two growing seasons. In these experiments, the percentage of infected daughter tubers produced by seed tubers that were infected with either potato potexvirus X (PVX), potato Andean mottle comovirus (APMoV), potato potyvirus Y (PVY) (jointly infected with PVX) or potato leafroll luteovirus (PLRV) was determined. Incomplete autoinfection was found in all cases, as the percentage of virus infected daughter tubers harvested from secondarily infected plants was invariably less than 100%, with the lowest percentage of infection being 30%. Changing the growing site to higher altitudes decreased autoinfection for all viruses. Therefore, the assumption of complete autoinfection of secondarily infected plants were rejected, while the hypothesis of environmentally dependent incomplete autoinfection was accepted. The findings help explain the occurrence of traditional seed management practices in the Andes and may help to develop locally adapted seed systems in environments of the world that have no steady access to healthy seed tubers coming from a formally certified seed system. The results obtained almost three decades ago are discussed in light of most recent knowledge on epigenetic regulation of host plant – virus interactions which allow for speculating about the underlying biological principles of the incomplete autoinfection. A research roadmap is proposed for achieving explicit experimental proof for the epigenetic regulation of incomplete autoinfection in the pathosystem under study. The current study utilizes annual data for the period of 1990–2014 and employs panel estimation techniques, which addresses the issues of cross-sectional dependence, fixed effect, and endogeneity. The results, across various estimates, confirm that environmental technology has a substantial negative influence on energy consumption and also plays an important role in improving energy efficiency by reducing energy intensity. These evidences suggest that environmental technology helps the OECD economies to reduce their overall energy consumption and improves overall energy efficiency in their respective countries. The comprehensive empirical outcomes document that financial development and income are the key determinants of energy demand. Given these results, the study proposes several fruitful implications regarding sustainable development goals of OECD countries.

Despite empirical literature identifying diverse drivers of CO2, the impact of national security, healthcare, entitlement programs, and total petroleum stocks has largely been overlooked. Thus, this study aims to bridge this gap by investigating, for the first time, the role of these economic variables in determining whether they contribute to CO2 reduction or escalation in the United States. To evaluate the interconnections among these variables, this study utilizes monthly data spanning from 1985 to 2022. Employing contemporary quantile approaches like Recursive CQ correlations, Cross-Quantilogram, and nonparametric quantile causality, the study effectively accommodates the nonlinear nature of the variables. These analytical techniques offer a comprehensive assessment of the relationships among the variables under scrutiny. The outcomes of the Cross-Quantilogram analysis reveal that health care, national security, and entitlement programs enhance ecological quality at different quantiles. Conversely, total petroleum stocks are associated with ecological deterioration. Based on these results, the study recommends a focus on raising awareness regarding sustainable procurement strategies, embracing environmentally friendly technologies, and improving energy efficiency in healthcare facilities.

Here, we investigated the health effects of neutraceutical oleuropein supplementation (0.59% w/w) on energy balance at the whole body and molecular level in mice fed a high fat diet (HFD). Oleuropein supplementation (HFD¿+¿O) prevented HFD-induced body weight gain resulting in a body weight that was comparable to that of normal fat diet (NFD)-fed mice. Furthermore, indirect calorimetric data, motoric movements, serum glucose and leptin levels, serum and hepatic lipid levels, adipocyte size and adipose tissue gene expression showed an improved health status compared to the control HFD-fed mice. In fact, it appeared indistinguishable between HFD¿+¿O-fed mice and NFD-fed mice. Initially, oleuropein might decrease intestinal energy uptake, while on the longer term weight maintenance could be related to an increased satiety signal. Our results indicate that oleuropein supplementation to a HFD can improve health by reducing adiposity.

Willis;Abstract Biofuels have been promoted as a renewable energy option in many countries, but have also faced extensive scrutiny over their sustainability. Food security is perhaps the most debated sustainability impact of biofuels, especially in regions such as Sub-Saharan Africa that experience high rates of malnutrition and have been a major destination for biofuel-related investments. This study assesses the local food security impacts of engagement in biofuel crop production using a consistent protocol between multiple crops and sites. We use standardized metrics of food security related to dietary diversity and perceptions of hunger, and focus on feedstock smallholders and plantation workers in four operational projects: a large-scale jatropha plantation (Mozambique), a smallholder-based jatropha project (Malawi) and two hybrid sugarcane projects (Malawi, Eswatini). Collectively these reflect the main feedstocks, modes of production and land use transitions related to biofuel projects in Sub-Sahara Africa. Inverse Probability Weighting analysis indicates that involvement in sugarcane production improved household food security for plantation workers and feedstock smallholders. Conversely, involvement in jatropha production does not have a statistically significant positive effect on household food security for both workers and smallholders. Regression models indicate that the factors driving food security indicator levels vary between study sites. Wealth indicators influence food security indicators in several sites, but the absolute level of income plays a smaller role, while income stability/regularity, access to credit and stable markets for selling sugarcane be important drivers as indicated by the strong effect of proxy variables on indicators.

The so-called Natural Resource Curse suggests that natural resource endowments are associated with lower longterm growth rates in the areas in which the resources are located. Lower growth arises because these areas tend to specialize in the development and exploitation of the natural resources at the expense of other dynamic economic activities that offer higher long-term growth potential. Empirical evidence has, however, not reached consistent conclusions. In this paper, we take advantage of the rapid growth in oil and gas development and production in Texas over the course of a decade to consider the localized effects on inter-industry county-level employment at the NAICS-2, county-level mean and median income, and key public finance measures at both the county and school district levels. Considering the effects within a single, large and economically diverse state enables us to control for important state-level variables that influence local public finances. We find little evidence of short term effects necessary to generate the circumstance of a resource curse over the longer term.

To discuss the response rates and characteristics of the study population. METHODS: Occupational cohort design leading to a retrospective cohort study of reproductive outcomes reported by 46 396 current and former employees of both sexes in the nuclear industry. Employees of nuclear establishments in the United Kingdom operated by the Atomic Energy Authority, the Atomic Weapons Establishment, and British Nuclear Fuels were surveyed with postal questionnaires ot collect information on pregnancies, children,and periods of infertility. Information on employment and monitoring for ionising radiation was supplied by the employing nuclear authority and was linked to pregnancies and periods of infertility with unique personal identification numbers. RESULTS: The design and completion of this study resulted in high quality data on a representative population of the Atomic Energy Authority, Atomic Weapons Establishment, and British Nuclear Fuels workforces. The response to the survey was extremely good (82% for male workers and 88% for female workers, excluding undelivered questionnaires), and a unique relational database has been created which will enable infertility, pregnancy, and child health outcomes to be examined with respect to the employment and radiation monitoring characteristics of parents. CONCLUSION: This is the first United Kingdom study to link detailed reproductive history data to occupational information held by employers. The methods developed for the study were found to be feasible and successful. The design can be adapted for other investigations of reproductive hazards to men and women in the workplace and is currently in use to survey over 100 000 armed forces personnel in an investigation of reproductive outcome among veterans of the Gulf war.

Despite the commitment of the United Nations (UN) to provide everyone with equal access to basic services, the construction sector still fails to reach the production capacity and quality standards which are needed to meet the fast-growing demand for affordable homes. Whilst innovation measures are urgently needed to address the existing inefficiencies, the identification and development of the most appropriate solutions require a comprehensive understanding of the barriers obstructing the design and construction phase of affordable housing. To identify such barriers, an exploratory data mining analysis was conducted in which agglomerative hierarchical clustering made it possible to gather latent knowledge from 3566 text-based research outputs sourced from the Web of Science and Scopus. The analysis captured 83 supply-side barriers which impact the efficiency of the value chain for affordable housing provision. Of these barriers, 18 affected the design and construction phase, and after grouping them by thematic area, seven key matters of concern were identified: (1) design (not) for all, (2) homogeneity of provision, (3) unhealthy living environment, (4) inadequate construction project management, (5) environmental unsustainability, (6) placemaking, and (7) inadequate technical knowledge and skillsets. The insights which resulted from the analysis were seen to support evidence-informed decision making across the affordable housing sector. The findings suggest that fixing the inefficiencies of the affordable housing provision system will require UN Member States to accelerate the transition towards a fully sustainable design and construction process. This transition should prioritize a more inclusive and socially sensitive approach to the design and construction of affordable homes, capitalizing on the benefits of greater user involvement. In addition, transformative actions which seek to deliver more resource-efficient and environmentally friendly homes should be promoted, as well as new investments in the training and upskilling of construction professionals.

Sampey;Scale-up to industrial production level of a fermentation process occurs after optimization at small scale, a critical transition for successful technology transfer and commercialization of a product of interest. At the large scale a number of important bioprocess engineering problems arise that should be taken into account to match the values obtained at the small scale and achieve the highest productivity and quality possible. However, the changes of the host strain's physiological and metabolic behavior in response to the scale transition are still not clear.Heterogeneity in substrate and oxygen distribution is an inherent factor at industrial scale (10,000 L) which affects the success of process up-scaling. To counteract these detrimental effects, changes in dissolved oxygen and pressure set points and addition of diluents were applied to 10,000 L scale to enable a successful process scale-up. A comprehensive semi-quantitative and time-dependent analysis of the exometabolome was performed to understand the impact of the scale-up on the metabolic/physiological behavior of the host microorganism. Intermediates from central carbon catabolism and mevalonate/ergosterol synthesis pathways were found to accumulate in both the 10 L and 10,000 L scale cultures in a time-dependent manner. Moreover, excreted metabolites analysis revealed that hypoxic conditions prevailed at the 10,000 L scale. The specific product yield increased at the 10,000 L scale, in spite of metabolic stress and catabolic-anabolic uncoupling unveiled by the decrease in biomass yield on consumed oxygen.An optimized S. cerevisiae fermentation process was successfully scaled-up to an industrial scale bioreactor. The oxygen uptake rate (OUR) and overall growth profiles were matched between scales. The major remaining differences between scales were wet cell weight and culture apparent viscosity. The metabolic and physiological behavior of the host microorganism at the 10,000 L scale was investigated with exometabolomics, indicating that reduced oxygen availability affected oxidative phosphorylation cascading into down- and up-stream pathways producing overflow metabolism. Our study revealed striking metabolic and physiological changes in response to hypoxia exerted by industrial bioprocess up-scaling.

The common assumption in potato virus epidemiology is that all daughter tubers produced by plants coming from infected mother tubers (secondary infection) will become infected via systemic translocation of the virus during growth. We hypothesize that depending on the prevalent environmental conditions, only a portion of the daughter tubers of a plant that is secondarily infected by viruses may become infected. To test this hypothesis experimental data from standardized field experiments were produced in three contrasting environments at 112, 3280, and 4000 m a.s.l. in Peru during two growing seasons. In these experiments, the percentage of infected daughter tubers produced by seed tubers that were infected with either potato potexvirus X (PVX), potato Andean mottle comovirus (APMoV), potato potyvirus Y (PVY) (jointly infected with PVX) or potato leafroll luteovirus (PLRV) was determined. Incomplete autoinfection was found in all cases, as the percentage of virus infected daughter tubers harvested from secondarily infected plants was invariably less than 100%, with the lowest percentage of infection being 30%. Changing the growing site to higher altitudes decreased autoinfection for all viruses. Therefore, the assumption of complete autoinfection of secondarily infected plants were rejected, while the hypothesis of environmentally dependent incomplete autoinfection was accepted. The findings help explain the occurrence of traditional seed management practices in the Andes and may help to develop locally adapted seed systems in environments of the world that have no steady access to healthy seed tubers coming from a formally certified seed system. The results obtained almost three decades ago are discussed in light of most recent knowledge on epigenetic regulation of host plant – virus interactions which allow for speculating about the underlying biological principles of the incomplete autoinfection. A research roadmap is proposed for achieving explicit experimental proof for the epigenetic regulation of incomplete autoinfection in the pathosystem under study. The current study utilizes annual data for the period of 1990–2014 and employs panel estimation techniques, which addresses the issues of cross-sectional dependence, fixed effect, and endogeneity. The results, across various estimates, confirm that environmental technology has a substantial negative influence on energy consumption and also plays an important role in improving energy efficiency by reducing energy intensity. These evidences suggest that environmental technology helps the OECD economies to reduce their overall energy consumption and improves overall energy efficiency in their respective countries. The comprehensive empirical outcomes document that financial development and income are the key determinants of energy demand. Given these results, the study proposes several fruitful implications regarding sustainable development goals of OECD countries.

Despite empirical literature identifying diverse drivers of CO2, the impact of national security, healthcare, entitlement programs, and total petroleum stocks has largely been overlooked. Thus, this study aims to bridge this gap by investigating, for the first time, the role of these economic variables in determining whether they contribute to CO2 reduction or escalation in the United States. To evaluate the interconnections among these variables, this study utilizes monthly data spanning from 1985 to 2022. Employing contemporary quantile approaches like Recursive CQ correlations, Cross-Quantilogram, and nonparametric quantile causality, the study effectively accommodates the nonlinear nature of the variables. These analytical techniques offer a comprehensive assessment of the relationships among the variables under scrutiny. The outcomes of the Cross-Quantilogram analysis reveal that health care, national security, and entitlement programs enhance ecological quality at different quantiles. Conversely, total petroleum stocks are associated with ecological deterioration. Based on these results, the study recommends a focus on raising awareness regarding sustainable procurement strategies, embracing environmentally friendly technologies, and improving energy efficiency in healthcare facilities.

Here, we investigated the health effects of neutraceutical oleuropein supplementation (0.59% w/w) on energy balance at the whole body and molecular level in mice fed a high fat diet (HFD). Oleuropein supplementation (HFD¿+¿O) prevented HFD-induced body weight gain resulting in a body weight that was comparable to that of normal fat diet (NFD)-fed mice. Furthermore, indirect calorimetric data, motoric movements, serum glucose and leptin levels, serum and hepatic lipid levels, adipocyte size and adipose tissue gene expression showed an improved health status compared to the control HFD-fed mice. In fact, it appeared indistinguishable between HFD¿+¿O-fed mice and NFD-fed mice. Initially, oleuropein might decrease intestinal energy uptake, while on the longer term weight maintenance could be related to an increased satiety signal. Our results indicate that oleuropein supplementation to a HFD can improve health by reducing adiposity.

Willis;Abstract Biofuels have been promoted as a renewable energy option in many countries, but have also faced extensive scrutiny over their sustainability. Food security is perhaps the most debated sustainability impact of biofuels, especially in regions such as Sub-Saharan Africa that experience high rates of malnutrition and have been a major destination for biofuel-related investments. This study assesses the local food security impacts of engagement in biofuel crop production using a consistent protocol between multiple crops and sites. We use standardized metrics of food security related to dietary diversity and perceptions of hunger, and focus on feedstock smallholders and plantation workers in four operational projects: a large-scale jatropha plantation (Mozambique), a smallholder-based jatropha project (Malawi) and two hybrid sugarcane projects (Malawi, Eswatini). Collectively these reflect the main feedstocks, modes of production and land use transitions related to biofuel projects in Sub-Sahara Africa. Inverse Probability Weighting analysis indicates that involvement in sugarcane production improved household food security for plantation workers and feedstock smallholders. Conversely, involvement in jatropha production does not have a statistically significant positive effect on household food security for both workers and smallholders. Regression models indicate that the factors driving food security indicator levels vary between study sites. Wealth indicators influence food security indicators in several sites, but the absolute level of income plays a smaller role, while income stability/regularity, access to credit and stable markets for selling sugarcane be important drivers as indicated by the strong effect of proxy variables on indicators.

The so-called Natural Resource Curse suggests that natural resource endowments are associated with lower longterm growth rates in the areas in which the resources are located. Lower growth arises because these areas tend to specialize in the development and exploitation of the natural resources at the expense of other dynamic economic activities that offer higher long-term growth potential. Empirical evidence has, however, not reached consistent conclusions. In this paper, we take advantage of the rapid growth in oil and gas development and production in Texas over the course of a decade to consider the localized effects on inter-industry county-level employment at the NAICS-2, county-level mean and median income, and key public finance measures at both the county and school district levels. Considering the effects within a single, large and economically diverse state enables us to control for important state-level variables that influence local public finances. We find little evidence of short term effects necessary to generate the circumstance of a resource curse over the longer term.

To discuss the response rates and characteristics of the study population. METHODS: Occupational cohort design leading to a retrospective cohort study of reproductive outcomes reported by 46 396 current and former employees of both sexes in the nuclear industry. Employees of nuclear establishments in the United Kingdom operated by the Atomic Energy Authority, the Atomic Weapons Establishment, and British Nuclear Fuels were surveyed with postal questionnaires ot collect information on pregnancies, children,and periods of infertility. Information on employment and monitoring for ionising radiation was supplied by the employing nuclear authority and was linked to pregnancies and periods of infertility with unique personal identification numbers. RESULTS: The design and completion of this study resulted in high quality data on a representative population of the Atomic Energy Authority, Atomic Weapons Establishment, and British Nuclear Fuels workforces. The response to the survey was extremely good (82% for male workers and 88% for female workers, excluding undelivered questionnaires), and a unique relational database has been created which will enable infertility, pregnancy, and child health outcomes to be examined with respect to the employment and radiation monitoring characteristics of parents. CONCLUSION: This is the first United Kingdom study to link detailed reproductive history data to occupational information held by employers. The methods developed for the study were found to be feasible and successful. The design can be adapted for other investigations of reproductive hazards to men and women in the workplace and is currently in use to survey over 100 000 armed forces personnel in an investigation of reproductive outcome among veterans of the Gulf war.

Despite the commitment of the United Nations (UN) to provide everyone with equal access to basic services, the construction sector still fails to reach the production capacity and quality standards which are needed to meet the fast-growing demand for affordable homes. Whilst innovation measures are urgently needed to address the existing inefficiencies, the identification and development of the most appropriate solutions require a comprehensive understanding of the barriers obstructing the design and construction phase of affordable housing. To identify such barriers, an exploratory data mining analysis was conducted in which agglomerative hierarchical clustering made it possible to gather latent knowledge from 3566 text-based research outputs sourced from the Web of Science and Scopus. The analysis captured 83 supply-side barriers which impact the efficiency of the value chain for affordable housing provision. Of these barriers, 18 affected the design and construction phase, and after grouping them by thematic area, seven key matters of concern were identified: (1) design (not) for all, (2) homogeneity of provision, (3) unhealthy living environment, (4) inadequate construction project management, (5) environmental unsustainability, (6) placemaking, and (7) inadequate technical knowledge and skillsets. The insights which resulted from the analysis were seen to support evidence-informed decision making across the affordable housing sector. The findings suggest that fixing the inefficiencies of the affordable housing provision system will require UN Member States to accelerate the transition towards a fully sustainable design and construction process. This transition should prioritize a more inclusive and socially sensitive approach to the design and construction of affordable homes, capitalizing on the benefits of greater user involvement. In addition, transformative actions which seek to deliver more resource-efficient and environmentally friendly homes should be promoted, as well as new investments in the training and upskilling of construction professionals.
