Potato plants (Solanum tuberosum L., cv. Désirée) were transformed with the polyphosphate kinase gene from Escherichia coli fused to the leader sequence of the ferredoxin oxidoreductase gene (FNR) from Spinacea oleracea under the control of the leaf specific St-LS1 promoter to introduce a novel phosphate pool in the chloroplasts of green tissues. Transgenic plants (cpPPK) in tissue culture developed necrotic lesions in older leaves and showed earlier leaf senescence while greenhouse plants showed no noticeable phenotype. Leaves of cpPPK plants contained less starch but higher concentrations of soluble sugars. The presence of polyphosphate in cpPPK leaves was demonstrated by toluidine blue staining and unambiguously verified and quantified by in vitro 31P-NMR of extracts. Polyphosphate accumulated during leaf development from 0.06 in juvenile leaves to 0.83 mg P g-1 DW in old leaves and had an average chain length of 18 residues in mature leaves. In situ 31P-NMR on small leaf pieces perfused with well-oxygenated medium showed only 0.036 mg P g-1 DW polyphosphate that was, however, greatly increased upon treatment with 50 mM ammonium sulfate at pH 7.3. This phenomenon along with a yield of 0.47 mg P g-1 DW polyphosphate from an extract of the same leaf material suggests that 93% of the polyphosphate pool is immobile. This conclusion is substantiated by the observation that no differences in polyphosphate pool sizes could be discerned between darkened and illuminated leaves, leaves treated with methylviologen or anaerobis and control leaves, treatments causing a change in the pool of ATP available for polyPi synthesis. Results are discussed in the context of the chelating properties of polyphosphates for cations and its consequences for the partitioning of photoassimilate between starch and soluble sugars.

Lapola;Large uncertainties still dominate the hypothesis of an abrupt large-scale shift of the Amazon forest caused by climate change [Amazonian forest dieback (AFD)] even though observational evidence shows the forest and regional climate changing. Here, we assess whether mitigation or adaptation action should be taken now, later, or not at all in light of such uncertainties. No action/later action would result in major social impacts that may influence migration to large Amazonian cities through a causal chain of climate change and forest degradation leading to lower river-water levels that affect transportation, food security, and health. Net-present value socioeconomic damage over a 30-year period after AFD is estimated between US dollar (USD) $957 billion (×10 9 ) and $3,589 billion (compared with Gross Brazilian Amazon Product of USD $150 billion per year), arising primarily from changes in the provision of ecosystem services. Costs of acting now would be one to two orders of magnitude lower than economic damages. However, while AFD mitigation alternatives—e.g., curbing deforestation—are attainable (USD $64 billion), their efficacy in achieving a forest resilience that prevents AFD is uncertain. Concurrently, a proposed set of 20 adaptation measures is also attainable (USD $122 billion) and could bring benefits even if AFD never occurs. An interdisciplinary research agenda to fill lingering knowledge gaps and constrain the risk of AFD should focus on developing sound experimental and modeling evidence regarding its likelihood, integrated with socioeconomic assessments to anticipate its impacts and evaluate the feasibility and efficacy of mitigation/adaptation options.

Tol; Samuel Frankhauser;Climate change is unique among the consequences of fossil fuel burning in its far reaching impact, both spatially and temporally. Earlier studies estimate the aggregated monetized damage due to climate change at 1.5 to 2.0% of world GDP (for 2 × CO2); the OECD would lose 1.0 to 1.5% of GDP; the developing countries 2.0 to 9.0%, according to these estimates. These figures are not comprehensive and highly uncertain. Newer studies increasingly emphasize adaptation, variability, extreme events, other (non-climate change) stress factors and the need for integrated assessment of damages. As a result, differences in impacts between regions and sectors have increased, the market impacts in developed countries tended to fall, and non-market impacts have become increasingly important. Marginal damages are more interesting from a policy point of view. Earlier estimates range from about US$5 to US$125 per tonne of carbon, with most estimates at the lower end of this range. These figures are based on polynomial functions in the level of climate change, but the rate of change may be equally important, as are the speed of adaptation, restoration and value adjustment. Furthermore, future vulnerability to climate change will be different from current vulnerability. On the whole, the market impacts fall (relatively) with economic growth while the non-market impacts rise (relatively) with growth.

Dietary shifts are key for enhancing the sustainability of current food systems but need to account for potential economic, social and environmental indirect effects as well. By tracing physical quantities of biomass along supply chains in a global economic model, we investigate the benefits of adopting the EAT-Lancet diet and other social, economic and environmental spillovers in the wider economy. We find that decreased global food demand reduces global biomass production, food prices, trade, land use and food loss and waste but also reduces food affordability for low-income agricultural households. In sub-Saharan Africa, increased food demand and higher prices decrease food affordability also for non-agricultural households. Economic spillovers into non-food sectors limit agricultural land and greenhouse gas reductions as cheaper biomass is demanded more for non-food use. From an environmental perspective, economy-wide greenhouse gas emissions increase as lower global food demand at lower prices frees income subsequently spent on non-food items.

Possible explanations for this gap are construction mistakes, improper adjusting of equipment, excessive simplification in simulation models and occupant behaviour. Many researchers and governmental institutions think the occupant is the main cause of this gap. However, only limited evidence exists for this. Therefore, an analysis is presented of actual and theoretical energy consumption based on specific household types and building characteristics. Using a large dataset (1.4 million social housing households), the average actual and theoretical energy consumptions (gas and electricity) of different household types and characteristics (income level, type of income, number of occupants and their age) were compared for each energy label. Additionally, the 10% highest and lowest energy-consuming groups were analysed. The use of combinations of occupant characteristics instead of individual occupant characteristics provides new insights into the influence of the occupant on energy demand. For example, in contrast to previous studies, low-income households consume more gas per m2 (space heating and hot water) than households with a high income for all types of housing. Furthermore, the performance gap is caused not only by the occupant but also by the assumed building characteristics.

With the aim of reducing the environmental burden of decentralized wastewater treatment plants in India, this project investigated five primary materials (stainless steel (SS), mild steel (MS), glass fibre reinforced polymer (GFRP), high density polyethylene (HDPE), and reinforced concrete cement (RCC)) in terms of the relative environmental impact that each would incur across 13 midpoint and 4 endpoint impact categories during the early life stages. The results showed that SS demonstrated substantially higher impact in total (5.47 Pt) and across each of the endpoint categories, most notably human health (3.12 Pt). Further investigations demonstrated that this was largely fed by the respiratory inorganics midpoint category that accounted for 50 % of the total impact (2.75 Pt), while global warming (0.93 Pt), non-renewable energy (0.70 Pt) and terrestrial ecotoxicity (0.62 Pt) were the only other considerable impacts. GFRP incurred the second greatest impact overall (2.32 Pt), while MS, RCC and HDPE followed with 1.82 Pt, 0.78 Pt, and 0.39 Pt respectively. HDPE afforded the greatest efficiency in all midpoint categories except carcinogens where RCC incurred the least environmental cost. Results were then compared with previous work and likely causal factors highlighted. Further study is recommended to investigate the longevity of the alternative materials in a wastewater containment role to support these results.

In Lebanon, fragility and hybridity — manifested in violent conflict and sectarian oligopolies — undermine entrepreneurial activities, among others through its impact on electricity supply. It is found here that hybrid governance results in inflated electricity prices and reduced electricity reliability, undermining business competitiveness and limiting innovative entrepreneurship. The article's contributions lie in providing a deeper understanding of entrepreneurship in fragile states and showing not merely that, but precisely how, governance matters for developmental entrepreneurship.

It can support sustainable development by aiming to safeguard the resources to mitigate negative impacts on the climate and the environment and to sustain our current and future generations. Yet, despite progress with circular economy initiatives, there is a risk of focusing on incremental innovations with little real impact, and possibly even creating serious negative rebound effects. This study suggests that the concept of “sufficiency” is inadequately represented in the current circular economy discourse and innovations, and this may be undermining real progress. In this paper, the Sufficiency-based Circular Economy paradigm is introduced. We investigate the following questions: What is the role of business in the sufficiency-based circular economy? What are the institutional limitations to the role of business as a driver for the transition and how might these be overcome? We conduct a “practice research” by analyzing company cases of sufficiency practices in a business context. We analyse 150 business cases to identify how their organizational strategies support sufficiency and what type of innovations they exemplify within this transition. We investigate seven core business elements for economic transformation (purpose, ownership, governance, finance, networks, scale-up and impact) of these businesses to understand how they drive the value propositions and their impact on the wider transition. This is followed by a discussion on a broader business and policy perspective of the Sufficiency-based Circular Economy.

AbstractIntroductionFoetal alcohol spectrum disorders (FASD) are a set of preventable conditions where the foetus is exposed to alcohol in utero and as a result suffers adverse consequences. To develop a public health strategy related to FASD, it is important to first establish what is known by the public about this condition. This study aimed to assess the current level of knowledge about FASD in the UK general population.MethodsA mixed methodology study was conducted using a 17‐item questionnaire and focus group sessions. Four focus groups were held with an average of 10 people in each group. Semi‐structured questions and thematic analysis of interviews alongside quantitative analysis of the questionnaire data was completed. The research was approved by an National Health service (NHS) research ethical committee.ResultsA total of 674 people responded to the questionnaire and a majority (86.7%) had heard about FASD, with most receiving their information from the media (26.2%) or from their work (27.7%). Four broad themes emerged. Overall these were: a general lack of knowledge about the subject; information about the subject needed to be personally relevant; there was a need for further education; and there was a lack of clarity in the current guidance on alcohol use in pregnancy.Discussion and conclusionsCurrently there appears to be a superficial level of knowledge about FASD in the UK general public. More detailed work in subgroups, such as young women, to identify their specific needs may be necessary before targeted public health and educational interventions can be developed to meet the needs of the general public.

Reducing greenhouse gas (GHG) emissions from agriculture has become a critical target in national climate change policies. More than 80% of the countries in Sub-Saharan Africa (SSA) refer to the reduction of agricultural emissions, including livestock, in their nationally determined contribution (NDC) to mitigate climate change. The livestock sector in Kenya contributes largely to the gross domestic product and to GHG emissions from the land use sector. The government has recently pledged in its NDC to curb total GHG emissions by 30% by 2030. Quantifying and linking the mitigation potential of farm practices to national targets is required to support realistically the implementation of NDCs. Improvements in feed and manure management represent promising mitigation options for dairy production. This study aimed (i) to assess mitigation and food production benefits of feed and manure management scenarios, including land use changes covering Kenya's entire dairy production region and (ii) to analyse the contribution of these practices to national targets on milk production and mitigation, and their biophysical feasibility given the availability of arable land. The results indicate that improving forage quality by increasing the use of Napier grass and supplementing dairy concentrates supports Kenya's NDC target, reduces emission intensities by 26%–31%, partially achieves the national milk productivity target for 2030 by 38%–41%, and shows high feasibility given the availability of arable land. Covering manure heaps may reduce emissions from manure management by 68%. In contrast, including maize silage in cattle diets would not reduce emission intensities due to the risk of ten-fold higher emissions from the conversion of land required to grow additional maize. The shortage of arable land may render the implementation of these improved feed practices largely infeasible. This assessment provides the first quantitative estimates of the potential of feed intensification and manure management to mitigate GHG emissions and to increase milk yields at sectoral-level and at a high spatial resolution for an SSA country. The scientific evidence is tailored to support actual policy and decision-making processes at the national level, such as 'Nationally Appropriate Mitigation Actions'. Linking feed intensification and manure management strategies with spatially-explicit estimates of mitigation and food production to national targets may help the sector to access climate financing while contributing to food security.

Potato plants (Solanum tuberosum L., cv. Désirée) were transformed with the polyphosphate kinase gene from Escherichia coli fused to the leader sequence of the ferredoxin oxidoreductase gene (FNR) from Spinacea oleracea under the control of the leaf specific St-LS1 promoter to introduce a novel phosphate pool in the chloroplasts of green tissues. Transgenic plants (cpPPK) in tissue culture developed necrotic lesions in older leaves and showed earlier leaf senescence while greenhouse plants showed no noticeable phenotype. Leaves of cpPPK plants contained less starch but higher concentrations of soluble sugars. The presence of polyphosphate in cpPPK leaves was demonstrated by toluidine blue staining and unambiguously verified and quantified by in vitro 31P-NMR of extracts. Polyphosphate accumulated during leaf development from 0.06 in juvenile leaves to 0.83 mg P g-1 DW in old leaves and had an average chain length of 18 residues in mature leaves. In situ 31P-NMR on small leaf pieces perfused with well-oxygenated medium showed only 0.036 mg P g-1 DW polyphosphate that was, however, greatly increased upon treatment with 50 mM ammonium sulfate at pH 7.3. This phenomenon along with a yield of 0.47 mg P g-1 DW polyphosphate from an extract of the same leaf material suggests that 93% of the polyphosphate pool is immobile. This conclusion is substantiated by the observation that no differences in polyphosphate pool sizes could be discerned between darkened and illuminated leaves, leaves treated with methylviologen or anaerobis and control leaves, treatments causing a change in the pool of ATP available for polyPi synthesis. Results are discussed in the context of the chelating properties of polyphosphates for cations and its consequences for the partitioning of photoassimilate between starch and soluble sugars.

Lapola;Large uncertainties still dominate the hypothesis of an abrupt large-scale shift of the Amazon forest caused by climate change [Amazonian forest dieback (AFD)] even though observational evidence shows the forest and regional climate changing. Here, we assess whether mitigation or adaptation action should be taken now, later, or not at all in light of such uncertainties. No action/later action would result in major social impacts that may influence migration to large Amazonian cities through a causal chain of climate change and forest degradation leading to lower river-water levels that affect transportation, food security, and health. Net-present value socioeconomic damage over a 30-year period after AFD is estimated between US dollar (USD) $957 billion (×10 9 ) and $3,589 billion (compared with Gross Brazilian Amazon Product of USD $150 billion per year), arising primarily from changes in the provision of ecosystem services. Costs of acting now would be one to two orders of magnitude lower than economic damages. However, while AFD mitigation alternatives—e.g., curbing deforestation—are attainable (USD $64 billion), their efficacy in achieving a forest resilience that prevents AFD is uncertain. Concurrently, a proposed set of 20 adaptation measures is also attainable (USD $122 billion) and could bring benefits even if AFD never occurs. An interdisciplinary research agenda to fill lingering knowledge gaps and constrain the risk of AFD should focus on developing sound experimental and modeling evidence regarding its likelihood, integrated with socioeconomic assessments to anticipate its impacts and evaluate the feasibility and efficacy of mitigation/adaptation options.

Earlier studies estimate the aggregated monetized damage due to climate change at 1.5 to 2.0% of world GDP (for 2 × CO2); the OECD would lose 1.0 to 1.5% of GDP; the developing countries 2.0 to 9.0%, according to these estimates. These figures are not comprehensive and highly uncertain. Newer studies increasingly emphasize adaptation, variability, extreme events, other (non-climate change) stress factors and the need for integrated assessment of damages. As a result, differences in impacts between regions and sectors have increased, the market impacts in developed countries tended to fall, and non-market impacts have become increasingly important. Marginal damages are more interesting from a policy point of view. Earlier estimates range from about US$5 to US$125 per tonne of carbon, with most estimates at the lower end of this range. These figures are based on polynomial functions in the level of climate change, but the rate of change may be equally important, as are the speed of adaptation, restoration and value adjustment. Furthermore, future vulnerability to climate change will be different from current vulnerability. On the whole, the market impacts fall (relatively) with economic growth while the non-market impacts rise (relatively) with growth.

Dietary shifts are key for enhancing the sustainability of current food systems but need to account for potential economic, social and environmental indirect effects as well. By tracing physical quantities of biomass along supply chains in a global economic model, we investigate the benefits of adopting the EAT-Lancet diet and other social, economic and environmental spillovers in the wider economy. We find that decreased global food demand reduces global biomass production, food prices, trade, land use and food loss and waste but also reduces food affordability for low-income agricultural households. In sub-Saharan Africa, increased food demand and higher prices decrease food affordability also for non-agricultural households. Economic spillovers into non-food sectors limit agricultural land and greenhouse gas reductions as cheaper biomass is demanded more for non-food use. From an environmental perspective, economy-wide greenhouse gas emissions increase as lower global food demand at lower prices frees income subsequently spent on non-food items.

Possible explanations for this gap are construction mistakes, improper adjusting of equipment, excessive simplification in simulation models and occupant behaviour. Many researchers and governmental institutions think the occupant is the main cause of this gap. However, only limited evidence exists for this. Therefore, an analysis is presented of actual and theoretical energy consumption based on specific household types and building characteristics. Using a large dataset (1.4 million social housing households), the average actual and theoretical energy consumptions (gas and electricity) of different household types and characteristics (income level, type of income, number of occupants and their age) were compared for each energy label. Additionally, the 10% highest and lowest energy-consuming groups were analysed. The use of combinations of occupant characteristics instead of individual occupant characteristics provides new insights into the influence of the occupant on energy demand. For example, in contrast to previous studies, low-income households consume more gas per m2 (space heating and hot water) than households with a high income for all types of housing. Furthermore, the performance gap is caused not only by the occupant but also by the assumed building characteristics.

With the aim of reducing the environmental burden of decentralized wastewater treatment plants in India, this project investigated five primary materials (stainless steel (SS), mild steel (MS), glass fibre reinforced polymer (GFRP), high density polyethylene (HDPE), and reinforced concrete cement (RCC)) in terms of the relative environmental impact that each would incur across 13 midpoint and 4 endpoint impact categories during the early life stages. The results showed that SS demonstrated substantially higher impact in total (5.47 Pt) and across each of the endpoint categories, most notably human health (3.12 Pt). Further investigations demonstrated that this was largely fed by the respiratory inorganics midpoint category that accounted for 50 % of the total impact (2.75 Pt), while global warming (0.93 Pt), non-renewable energy (0.70 Pt) and terrestrial ecotoxicity (0.62 Pt) were the only other considerable impacts. GFRP incurred the second greatest impact overall (2.32 Pt), while MS, RCC and HDPE followed with 1.82 Pt, 0.78 Pt, and 0.39 Pt respectively. HDPE afforded the greatest efficiency in all midpoint categories except carcinogens where RCC incurred the least environmental cost. Results were then compared with previous work and likely causal factors highlighted. Further study is recommended to investigate the longevity of the alternative materials in a wastewater containment role to support these results.

In Lebanon, fragility and hybridity — manifested in violent conflict and sectarian oligopolies — undermine entrepreneurial activities, among others through its impact on electricity supply. It is found here that hybrid governance results in inflated electricity prices and reduced electricity reliability, undermining business competitiveness and limiting innovative entrepreneurship. The article's contributions lie in providing a deeper understanding of entrepreneurship in fragile states and showing not merely that, but precisely how, governance matters for developmental entrepreneurship.

It can support sustainable development by aiming to safeguard the resources to mitigate negative impacts on the climate and the environment and to sustain our current and future generations. Yet, despite progress with circular economy initiatives, there is a risk of focusing on incremental innovations with little real impact, and possibly even creating serious negative rebound effects. This study suggests that the concept of “sufficiency” is inadequately represented in the current circular economy discourse and innovations, and this may be undermining real progress. In this paper, the Sufficiency-based Circular Economy paradigm is introduced. We investigate the following questions: What is the role of business in the sufficiency-based circular economy? What are the institutional limitations to the role of business as a driver for the transition and how might these be overcome? We conduct a “practice research” by analyzing company cases of sufficiency practices in a business context. We analyse 150 business cases to identify how their organizational strategies support sufficiency and what type of innovations they exemplify within this transition. We investigate seven core business elements for economic transformation (purpose, ownership, governance, finance, networks, scale-up and impact) of these businesses to understand how they drive the value propositions and their impact on the wider transition. This is followed by a discussion on a broader business and policy perspective of the Sufficiency-based Circular Economy.

AbstractIntroductionFoetal alcohol spectrum disorders (FASD) are a set of preventable conditions where the foetus is exposed to alcohol in utero and as a result suffers adverse consequences. To develop a public health strategy related to FASD, it is important to first establish what is known by the public about this condition. This study aimed to assess the current level of knowledge about FASD in the UK general population.MethodsA mixed methodology study was conducted using a 17‐item questionnaire and focus group sessions. Four focus groups were held with an average of 10 people in each group. Semi‐structured questions and thematic analysis of interviews alongside quantitative analysis of the questionnaire data was completed. The research was approved by an National Health service (NHS) research ethical committee.ResultsA total of 674 people responded to the questionnaire and a majority (86.7%) had heard about FASD, with most receiving their information from the media (26.2%) or from their work (27.7%). Four broad themes emerged. Overall these were: a general lack of knowledge about the subject; information about the subject needed to be personally relevant; there was a need for further education; and there was a lack of clarity in the current guidance on alcohol use in pregnancy.Discussion and conclusionsCurrently there appears to be a superficial level of knowledge about FASD in the UK general public. More detailed work in subgroups, such as young women, to identify their specific needs may be necessary before targeted public health and educational interventions can be developed to meet the needs of the general public.

Reducing greenhouse gas (GHG) emissions from agriculture has become a critical target in national climate change policies. More than 80% of the countries in Sub-Saharan Africa (SSA) refer to the reduction of agricultural emissions, including livestock, in their nationally determined contribution (NDC) to mitigate climate change. The livestock sector in Kenya contributes largely to the gross domestic product and to GHG emissions from the land use sector. The government has recently pledged in its NDC to curb total GHG emissions by 30% by 2030. Quantifying and linking the mitigation potential of farm practices to national targets is required to support realistically the implementation of NDCs. Improvements in feed and manure management represent promising mitigation options for dairy production. This study aimed (i) to assess mitigation and food production benefits of feed and manure management scenarios, including land use changes covering Kenya's entire dairy production region and (ii) to analyse the contribution of these practices to national targets on milk production and mitigation, and their biophysical feasibility given the availability of arable land. The results indicate that improving forage quality by increasing the use of Napier grass and supplementing dairy concentrates supports Kenya's NDC target, reduces emission intensities by 26%–31%, partially achieves the national milk productivity target for 2030 by 38%–41%, and shows high feasibility given the availability of arable land. Covering manure heaps may reduce emissions from manure management by 68%. In contrast, including maize silage in cattle diets would not reduce emission intensities due to the risk of ten-fold higher emissions from the conversion of land required to grow additional maize. The shortage of arable land may render the implementation of these improved feed practices largely infeasible. This assessment provides the first quantitative estimates of the potential of feed intensification and manure management to mitigate GHG emissions and to increase milk yields at sectoral-level and at a high spatial resolution for an SSA country. The scientific evidence is tailored to support actual policy and decision-making processes at the national level, such as 'Nationally Appropriate Mitigation Actions'. Linking feed intensification and manure management strategies with spatially-explicit estimates of mitigation and food production to national targets may help the sector to access climate financing while contributing to food security.
