The purpose of the present study was to determine the acute effects of the anticraving compound acamprosate (calcium acetylhomotaurinate) and the closely related compound homotaurine on ethanol intake and ethanol-stimulated dopamine release in the nucleus accumbens. Male rats were treated with acamprosate (200 or 400 mg/kg intraperitoneally, i.p.) or homotaurine (10, 50, or 100 mg/kg i.p.) 15 min prior to access to 10% ethanol and water for 1 h in a two-bottle choice restricted access paradigm. A separate group of rats was implanted with microdialysis probes in the nucleus accumbens and given an acute injection of ethanol (1.5 g/kg i.p.) that was preceded by saline, acamprosate, or homotaurine. Acamprosate and homotaurine dose-dependently reduced ethanol intake and preference. These compounds also delayed or suppressed ethanol-stimulated increases in nucleus accumbens dopamine release, suggesting that acamprosate and homotaurine may reduce ethanol intake by interfering with the ability of ethanol to activate the mesolimbic dopamine reward system.

Kulinski; Matthew Lee Smith; SangNam Ahn; SangNam Ahn; Kate Lorig;With a near 20-year developmental history as an evidence-based program, the suite of Chronic Disease Self-Management Education (CDSME) programs were selected in 2010 for grand-scale dissemination in a federally supported initiative to improve the health of older Americans. The primary charge of this national effort was to establish a sustainable program delivery system for empowering American adults with one or more chronic conditions to better manage their health. The current study focused on a series of dissemination and implementation science research questions to: (1) examine the geographic distribution of participation in this initiative across the Unites States; (2) describe workshop characteristics engaged to reach program participants in various settings; and (3) describe personal characteristics of the first 100,000 participants. Each subsequent entering cohort was descriptively examined to indicate whether there was constancy or change in delivery sites and populations reached over time. Findings show a strengthening of the workshop delivery infrastructure in that it took 9.4 months to reach the first 25,000 participants in 853 counties compared to 5.4 months to reach the last 25,000 participants in 1,109 counties. The workshop delivery characteristics and participant characteristics remained relatively consistent across increments of 25,000 participants reached, although general trends were observed for some variables. For example, after reaching the first 25,000 participants, subsequent groups of 25,000 participants were reached more quickly. Additionally, workshops were increasingly delivered in ZIP Codes with higher percentages of families residing below the federal poverty line. As more participants were reached, more participants with chronic conditions were enrolled. This national translational study illustrates the rapid expansion of CDSME programs throughout the United States and capability to reach diverse populations in a variety of settings.

AbstractMicroalgae represent one of the most promising groups of candidate organisms for replacing fossil fuels with contemporary primary production as a renewable source of energy. Algae can produce many times more biomass per unit area than terrestrial crop plants, easing the competing demands for land with food crops and native ecosystems. However, several aspects of algal biology present unique challenges to the industrial‐scale aquaculture of photosynthetic microorganisms. These include high susceptibility to invading aquatic consumers and weeds, as well as prodigious requirements for nutrients that may compete with the fertiliser demands of other crops. Most research on algal biofuel technologies approaches these problems from a cellular or genetic perspective, attempting either to engineer or select algal strains with particular traits. However, inherent functional trade‐offs may limit the capacity of genetic selection or synthetic biology to simultaneously optimise multiple functional traits for biofuel productivity and resilience. We argue that a community engineering approach that manages microalgal diversity, species composition and environmental conditions may lead to more robust and productive biofuel ecosystems. We review evidence for trade‐offs, challenges and opportunities in algal biofuel cultivation with a goal of guiding research towards intensifying bioenergy production using established principles of community and ecosystem ecology.

Consumers are increasingly demanding more nutritious food products with less environmental degradation, particularly related to water and fertilizer nitrogen (N) inputs. While triticale (× Triticosecale) is often referenced as having superior abiotic stress tolerance compared to wheat, few studies have compared crop productivity and resource use efficiencies under a range of N-and water-limited conditions. Because previous work has shown that blending wheat with triticale in a 40:60 ratio can yield acceptable and more nutritious baked products, we tested the hypothesis that increasing the use of triticale grain in the baking supply chain would reduce the environmental footprint for water and N fertilizer use. Using a dataset comprised of 37 site-years encompassing normal and stress-induced environments in California, we assessed yield, yield stability, and the efficiency of water and fertilizer N use for 67 and 17 commercial varieties of wheat and triticale, respectively. By identifying environments that favor one crop type over the other, we then quantified the sustainability implications of producing a mixed triticale-wheat flour at the regional scale. Results indicate that triticale outyielded wheat by 11% (p < 0.05) and 19% (p < 0.05) under average and N-limited conditions, respectively. However, wheat was 3% (p < 0.05) more productive in water-limited environments. Overall, triticale had greater yield stability and produced more grain per unit of water and N fertilizer inputs, especially in high-yielding environments. We estimate these differences could translate to regional N fertilizer savings (up to 555 Mg N or 166 CO2-eq kg ha−1) in a 40:60 blending scenario when wheat is sourced from water-limited and low-yielding fields and triticale from N-limited and high-yielding areas. Results suggest that optimizing the agronomic and environmental benefits of triticale would increase the overall resource use efficiency and sustainability of the agri-food system, although such a transition would require fundamental changes to the current system spanning producers, processors, and consumers.

Aryana;Type 2 resistant starch (RS2) has been shown to improve glycemic control and some cardiovascular endpoints in rodent and human studies.The aim of this study was to perform one of the first randomized clinical trials in adults with prediabetes and one of the longest trials to test whether RS2 can improve cardiometabolic health.68 overweight [body mass index (BMI) ≥27 kg/m2] adults aged 35-75 y with prediabetes were randomized to consume 45 g/d of high-amylose maize (RS2) or an isocaloric amount of the rapidly digestible starch amylopectin (control) for 12 wk. At baseline and postintervention, ectopic fat depots (visceral adipose tissue, intrahepatic lipids, and intramyocellular lipids) were measured by magnetic resonance imaging/spectroscopy, energy metabolism by respiratory chamber, and carbohydrate metabolism by glycated hemoglobin (HbA1c), an intravenous glucose tolerance test, and a meal tolerance test. Cardiovascular risk factors-serum lipids, blood pressure, heart rate, and inflammatory markers (high-sensitivity C-reactive protein [hs-CRP], interleukin-6, and tumor necrosis factor [TNF]-α)-were also measured. The primary endpoints were insulin sensitivity, insulin secretion, ectopic fat, and markers of inflammation. Data were primarily analyzed as treatment effects via a linear mixed model both with and without the addition of covariates.Relative to the control group, RS2 lowered HbA1c by a clinically insignificant 0.1 ± 0.2% (Δ = -1 ± 2 mmol/mol; P = 0.05) but did not affect insulin secretion, insulin sensitivity, the disposition index, or glucose or insulin areas under the curve relative to baseline (P ≥ 0.23). RS2 decreased heart rate by 5 ± 9 beats/min (P = 0.02) and TNF-α concentrations by 2.1 ± 2.7 pg/mL (P = 0.004), relative to the control group. In eastern Africa droughts can cause crop failure and lead to food insecurity. With increasing temperatures, there is an a priori assumption that droughts are becoming more severe. However, the link between droughts and climate change is not sufficiently understood. Here we investigate trends in long-term agricultural drought and the influence of increasing temperatures and precipitation deficits. Using a combination of models and observational datasets, we studied trends, spanning the period from 1900 (to approximate pre-industrial conditions) to 2018, for six regions in eastern Africa in four drought-related annually averaged variables: soil moisture, precipitation, temperature, and evaporative demand (E0). In standardized soil moisture data, we found no discernible trends. The strongest influence on soil moisture variability was from precipitation, especially in the drier or water-limited study regions; temperature and E0 did not demonstrate strong relations to soil moisture. However, the error margins on precipitation trend estimates are large and no clear trend is evident, whereas significant positive trends were observed in local temperatures. The trends in E0 are predominantly positive, but we do not find strong relations between E0 and soil moisture trends. Nevertheless, the E0 trend results can still be of interest for irrigation purposes because it is E0 that determines the maximum evaporation rate. We conclude that until now the impact of increasing local temperatures on agricultural drought in eastern Africa is limited and we recommend that any soil moisture analysis be supplemented by an analysis of precipitation deficit.

We have previously shown that rats exposed to 20% ethanol intermittently in a two-bottle choice paradigm will consume two times more ethanol than those given continuous access without the use of water deprivation or sucrose fading (5-6 g/kg every 24 h vs 2-3 g/kg every 24 h, respectively). In this study, we have adapted the model to an operant self-administration paradigm. Long-Evans rats were given access to 20% ethanol in overnight sessions on one of two schedules: (1) intermittent (Monday, Wednesday, and Friday) or (2) daily (Monday through Friday). With the progression of the overnight sessions, both groups showed a steady escalation in drinking (3-6 g/kg every 14 h) without the use of a sucrose-fading procedure. Following the acquisition phase, the 20% ethanol groups consumed significantly more ethanol than did animals trained to consume 10% ethanol with a sucrose fade (1.5 vs 0.7 g/kg every 30 min) and reached significantly higher blood ethanol concentrations. In addition, training history (20% ethanol vs 10% ethanol with sucrose fade) had a significant effect on the subsequent self-administration of higher concentrations of ethanol. Administration of the pharmacological stressor yohimbine following extinction caused a significant reinstatement of ethanol-seeking behavior. Both 20% ethanol models show promise and are amenable to the study of maintenance, motivation, and reinstatement. Furthermore, training animals to lever press for ethanol without the use of sucrose fading removes a potential confound from self-administration studies.

Phase change in gas flow channels is explained, and a simplified approach accounting for phase change is incorporated into the fuel cell model. It is found that the liquid water contours in the GDL are similar along flow channels when the channels are subject to two-phase flow. Analysis is performed on a dimensionless parameter Da0 introduced in our previous paper [Y. Wang and K. S. Chen, Chemical Engineering Science 66 (2011) 3557–3567] and the parameter is further evaluated in a realistic fuel cell. We found that the GDL's liquid water (or liquid-free) region is determined by the Da0 number which lumps several parameters, including the thermal conductivity and operating temperature. By adjusting these factors, a liquid-free GDL zone can be created even though the channel stream is two-phase flow. Such a liquid-free zone is adjacent to the two-phase region, benefiting local water management, namely avoiding both severe flooding and dryness.

The purpose of the present study was to determine the acute effects of the anticraving compound acamprosate (calcium acetylhomotaurinate) and the closely related compound homotaurine on ethanol intake and ethanol-stimulated dopamine release in the nucleus accumbens. Male rats were treated with acamprosate (200 or 400 mg/kg intraperitoneally, i.p.) or homotaurine (10, 50, or 100 mg/kg i.p.) 15 min prior to access to 10% ethanol and water for 1 h in a two-bottle choice restricted access paradigm. A separate group of rats was implanted with microdialysis probes in the nucleus accumbens and given an acute injection of ethanol (1.5 g/kg i.p.) that was preceded by saline, acamprosate, or homotaurine. Acamprosate and homotaurine dose-dependently reduced ethanol intake and preference. These compounds also delayed or suppressed ethanol-stimulated increases in nucleus accumbens dopamine release, suggesting that acamprosate and homotaurine may reduce ethanol intake by interfering with the ability of ethanol to activate the mesolimbic dopamine reward system.

The primary charge of this national effort was to establish a sustainable program delivery system for empowering American adults with one or more chronic conditions to better manage their health. The current study focused on a series of dissemination and implementation science research questions to: (1) examine the geographic distribution of participation in this initiative across the Unites States; (2) describe workshop characteristics engaged to reach program participants in various settings; and (3) describe personal characteristics of the first 100,000 participants. Each subsequent entering cohort was descriptively examined to indicate whether there was constancy or change in delivery sites and populations reached over time. Findings show a strengthening of the workshop delivery infrastructure in that it took 9.4 months to reach the first 25,000 participants in 853 counties compared to 5.4 months to reach the last 25,000 participants in 1,109 counties. The workshop delivery characteristics and participant characteristics remained relatively consistent across increments of 25,000 participants reached, although general trends were observed for some variables. For example, after reaching the first 25,000 participants, subsequent groups of 25,000 participants were reached more quickly. Additionally, workshops were increasingly delivered in ZIP Codes with higher percentages of families residing below the federal poverty line. As more participants were reached, more participants with chronic conditions were enrolled. This national translational study illustrates the rapid expansion of CDSME programs throughout the United States and capability to reach diverse populations in a variety of settings.

AbstractMicroalgae represent one of the most promising groups of candidate organisms for replacing fossil fuels with contemporary primary production as a renewable source of energy. Algae can produce many times more biomass per unit area than terrestrial crop plants, easing the competing demands for land with food crops and native ecosystems. However, several aspects of algal biology present unique challenges to the industrial‐scale aquaculture of photosynthetic microorganisms. These include high susceptibility to invading aquatic consumers and weeds, as well as prodigious requirements for nutrients that may compete with the fertiliser demands of other crops. Most research on algal biofuel technologies approaches these problems from a cellular or genetic perspective, attempting either to engineer or select algal strains with particular traits. However, inherent functional trade‐offs may limit the capacity of genetic selection or synthetic biology to simultaneously optimise multiple functional traits for biofuel productivity and resilience. We argue that a community engineering approach that manages microalgal diversity, species composition and environmental conditions may lead to more robust and productive biofuel ecosystems. We review evidence for trade‐offs, challenges and opportunities in algal biofuel cultivation with a goal of guiding research towards intensifying bioenergy production using established principles of community and ecosystem ecology.

Lundy;Wheat (Triticum aestivum L.) is a major global commodity and the primary source for baked products in agri-food supply chains. Consumers are increasingly demanding more nutritious food products with less environmental degradation, particularly related to water and fertilizer nitrogen (N) inputs. While triticale (× Triticosecale) is often referenced as having superior abiotic stress tolerance compared to wheat, few studies have compared crop productivity and resource use efficiencies under a range of N-and water-limited conditions. Because previous work has shown that blending wheat with triticale in a 40:60 ratio can yield acceptable and more nutritious baked products, we tested the hypothesis that increasing the use of triticale grain in the baking supply chain would reduce the environmental footprint for water and N fertilizer use. Using a dataset comprised of 37 site-years encompassing normal and stress-induced environments in California, we assessed yield, yield stability, and the efficiency of water and fertilizer N use for 67 and 17 commercial varieties of wheat and triticale, respectively. By identifying environments that favor one crop type over the other, we then quantified the sustainability implications of producing a mixed triticale-wheat flour at the regional scale. Results indicate that triticale outyielded wheat by 11% (p < 0.05) and 19% (p < 0.05) under average and N-limited conditions, respectively. However, wheat was 3% (p < 0.05) more productive in water-limited environments. Overall, triticale had greater yield stability and produced more grain per unit of water and N fertilizer inputs, especially in high-yielding environments. We estimate these differences could translate to regional N fertilizer savings (up to 555 Mg N or 166 CO2-eq kg ha−1) in a 40:60 blending scenario when wheat is sourced from water-limited and low-yielding fields and triticale from N-limited and high-yielding areas. Results suggest that optimizing the agronomic and environmental benefits of triticale would increase the overall resource use efficiency and sustainability of the agri-food system, although such a transition would require fundamental changes to the current system spanning producers, processors, and consumers.

Lundy;Wheat (Triticum aestivum L.) is a major global commodity and the primary source for baked products in agri-food supply chains. Consumers are increasingly demanding more nutritious food products with less environmental degradation, particularly related to water and fertilizer nitrogen (N) inputs. While triticale (× Triticosecale) is often referenced as having superior abiotic stress tolerance compared to wheat, few studies have compared crop productivity and resource use efficiencies under a range of N-and water-limited conditions. Because previous work has shown that blending wheat with triticale in a 40:60 ratio can yield acceptable and more nutritious baked products, we tested the hypothesis that increasing the use of triticale grain in the baking supply chain would reduce the environmental footprint for water and N fertilizer use. Using a dataset comprised of 37 site-years encompassing normal and stress-induced environments in California, we assessed yield, yield stability, and the efficiency of water and fertilizer N use for 67 and 17 commercial varieties of wheat and triticale, respectively. By identifying environments that favor one crop type over the other, we then quantified the sustainability implications of producing a mixed triticale-wheat flour at the regional scale. Results indicate that triticale outyielded wheat by 11% (p < 0.05) and 19% (p < 0.05) under average and N-limited conditions, respectively. However, wheat was 3% (p < 0.05) more productive in water-limited environments. Overall, triticale had greater yield stability and produced more grain per unit of water and N fertilizer inputs, especially in high-yielding environments. We estimate these differences could translate to regional N fertilizer savings (up to 555 Mg N or 166 CO2-eq kg ha−1) in a 40:60 blending scenario when wheat is sourced from water-limited and low-yielding fields and triticale from N-limited and high-yielding areas. Results suggest that optimizing the agronomic and environmental benefits of triticale would increase the overall resource use efficiency and sustainability of the agri-food system, although such a transition would require fundamental changes to the current system spanning producers, processors, and consumers.

At baseline and postintervention, ectopic fat depots (visceral adipose tissue, intrahepatic lipids, and intramyocellular lipids) were measured by magnetic resonance imaging/spectroscopy, energy metabolism by respiratory chamber, and carbohydrate metabolism by glycated hemoglobin (HbA1c), an intravenous glucose tolerance test, and a meal tolerance test. Cardiovascular risk factors-serum lipids, blood pressure, heart rate, and inflammatory markers (high-sensitivity C-reactive protein [hs-CRP], interleukin-6, and tumor necrosis factor [TNF]-α)-were also measured. The primary endpoints were insulin sensitivity, insulin secretion, ectopic fat, and markers of inflammation. Data were primarily analyzed as treatment effects via a linear mixed model both with and without the addition of covariates.Relative to the control group, RS2 lowered HbA1c by a clinically insignificant 0.1 ± 0.2% (Δ = -1 ± 2 mmol/mol; P = 0.05) but did not affect insulin secretion, insulin sensitivity, the disposition index, or glucose or insulin areas under the curve relative to baseline (P ≥ 0.23). In eastern Africa droughts can cause crop failure and lead to food insecurity. With increasing temperatures, there is an a priori assumption that droughts are becoming more severe. However, the link between droughts and climate change is not sufficiently understood. Here we investigate trends in long-term agricultural drought and the influence of increasing temperatures and precipitation deficits. Using a combination of models and observational datasets, we studied trends, spanning the period from 1900 (to approximate pre-industrial conditions) to 2018, for six regions in eastern Africa in four drought-related annually averaged variables: soil moisture, precipitation, temperature, and evaporative demand (E0). In standardized soil moisture data, we found no discernible trends. The strongest influence on soil moisture variability was from precipitation, especially in the drier or water-limited study regions; temperature and E0 did not demonstrate strong relations to soil moisture. However, the error margins on precipitation trend estimates are large and no clear trend is evident, whereas significant positive trends were observed in local temperatures. The trends in E0 are predominantly positive, but we do not find strong relations between E0 and soil moisture trends. Nevertheless, the E0 trend results can still be of interest for irrigation purposes because it is E0 that determines the maximum evaporation rate. We conclude that until now the impact of increasing local temperatures on agricultural drought in eastern Africa is limited and we recommend that any soil moisture analysis be supplemented by an analysis of precipitation deficit.

We have previously shown that rats exposed to 20% ethanol intermittently in a two-bottle choice paradigm will consume two times more ethanol than those given continuous access without the use of water deprivation or sucrose fading (5-6 g/kg every 24 h vs 2-3 g/kg every 24 h, respectively). In this study, we have adapted the model to an operant self-administration paradigm. Long-Evans rats were given access to 20% ethanol in overnight sessions on one of two schedules: (1) intermittent (Monday, Wednesday, and Friday) or (2) daily (Monday through Friday). With the progression of the overnight sessions, both groups showed a steady escalation in drinking (3-6 g/kg every 14 h) without the use of a sucrose-fading procedure. Following the acquisition phase, the 20% ethanol groups consumed significantly more ethanol than did animals trained to consume 10% ethanol with a sucrose fade (1.5 vs 0.7 g/kg every 30 min) and reached significantly higher blood ethanol concentrations. In addition, training history (20% ethanol vs 10% ethanol with sucrose fade) had a significant effect on the subsequent self-administration of higher concentrations of ethanol. Administration of the pharmacological stressor yohimbine following extinction caused a significant reinstatement of ethanol-seeking behavior. Both 20% ethanol models show promise and are amenable to the study of maintenance, motivation, and reinstatement. Furthermore, training animals to lever press for ethanol without the use of sucrose fading removes a potential confound from self-administration studies.

Phase change in gas flow channels is explained, and a simplified approach accounting for phase change is incorporated into the fuel cell model. It is found that the liquid water contours in the GDL are similar along flow channels when the channels are subject to two-phase flow. Analysis is performed on a dimensionless parameter Da0 introduced in our previous paper [Y. Wang and K. S. Chen, Chemical Engineering Science 66 (2011) 3557–3567] and the parameter is further evaluated in a realistic fuel cell. We found that the GDL's liquid water (or liquid-free) region is determined by the Da0 number which lumps several parameters, including the thermal conductivity and operating temperature. By adjusting these factors, a liquid-free GDL zone can be created even though the channel stream is two-phase flow. Such a liquid-free zone is adjacent to the two-phase region, benefiting local water management, namely avoiding both severe flooding and dryness.
