Abstract

EWP-5の第一制限アミノ酸はTrpであるので, その制限度を調べるためにTrpの利用率の悪いニコチン酸欠, ショ糖食を用い, 幼若ラットの成長からEWP-5のTrpの制限度を求めた。その結果, EWP-5食群の成長は, ニコチン酸もしくはTrpの添加により有意に増加し, EWP-5はTrpが制限になっていることが見いだされた。しかし, 糖質源をショ糖からα-コーンスターチに変えることによっても, ニコチン酸添加もしくはTrp添加と同じ体重増加量を示したので, EWP-5のTrp制限はそれほど強いものではないと思われた。投与Trpの形態の違い, すなわちタンパク態Trp, 低分子態Trp, 遊離Trpの相違が, どの程度Trpからのナイアシンへの変換に影響するのかを, 血液および肝臓NADレベルの比較, ならびに尿中へのN1-メチルニコチンアミドの排泄量の比較から調べた。その結果, ニコチン酸欠, ショ糖食における血中NADレベルは, 三つの形態による違いは認められなかったが, 肝NADレベルは, EW食群に比しEWP-5食群およびA.A. 食群は低い値を示したが, これはTrp含量の差異に起因するものであろう。しかし, 尿中へのN1-メチルニコチンアミド排泄量はEWP-5食群>EW 食群>A.A. 食群であった。また, ニコチン酸含有のα-コーンスターチ: ショ糖 (2: 1) 食においては, 血液および肝臓NADレベルは, EWP-5食群>EW食群>A.A. 食群であった。N1-メチルニコチンアミド排泄量も, EWP-5食群が他の2群に比し高い傾向が認められた。以上の事実から, ペプチド態Trpが最もよくナイアシンに変換され, 次にタンパク態Trpで, 遊離Trpは最も低いと考えられた。
