Abstract

Представлен метод определения гидазепама в крови и биологических тканях по его фармакологически активному метаболиту N1-дезалкилгидазепаму в варианте двумерной хроматомасспектрометрии. С целью изолирования N1-дезалкилгидазепама предложено два варианта экстракции – жидкостная и твердофазная. Показано, что в случае жидкостной экстракции оптимальным экстрагентом является смесь 1-хлорбутана и этилацетата в соотношении 9:1. Эффективность изолирования жидкостной и твердофазной экстракцией составляет 98 и 94 % соответственно. Линейный диапазон определяемых концентраций составил 20-1000 нг/мл в крови, и 0,2-10 мкг/г в тканях органов. Предел обнаружения в крови и тканях составил соответственно 3 и 5 нг.

review methods of determination gidazepam in blood and biological tissues on its awake metabolite N1-desalkylgidazepam by two-dimensional gas chromatography-mass spectrometry. For isolate N1-desalkylgidazepam were applied two techniques –solvent and solid-phase extraction. We found, what extraction mixture 1-chlorbutane-ethylacetic ether (9:1), is a optimal for isolate N1-desalkylgidazepam. Extraction yield by solvent and solid-phase extraction were 98 and 94 % respectively. The working range for blood was 20-1000 ng/mL and for tissues was 0,2-10 μg/g/ The limit of detection and limit of quantitative for blood and tissues were 3 and 5 ng respectively.
