ABSTRACTEnzymatic wheat gluten hydrolysis at high solid concentrations is advantageous from an environmental and economic point of view. However, increased wheat gluten concentrations result in a concentration effect with a decreased hydrolysis rate at constant enzyme‐to‐substrate ratios and a decreased maximum attainable degree of hydrolysis (DH%). We here identified the underlying factors causing the concentration effect. Wheat gluten was hydrolyzed at solid concentrations from 4.4% to 70%. The decreased hydrolysis rate was present at all solid concentrations and at any time of the reaction. Mass transfer limitations, enzyme inhibition and water activity were shown to not cause this hydrolysis rate limitation up to 50% solids. However, the hydrolysis rate limitation can be, at least partly, explained by a second‐order enzyme inactivation process. Furthermore, mass transfer impeded the hydrolysis above 60% solids. Addition of enzyme after 24 h at high solid concentrations scarcely increased the DH%, suggesting that the maximum attainable DH% decreases at high solid concentrations. Reduced enzyme activities caused by low water activities can explain this DH% limitation. Finally, a possible influence of the plastein reaction on the DH% limitation is discussed. Biotechnol. Systemic delivery of therapeutic agents remains ineffective against diffuse intrinsic pontine glioma (DIPG), possibly due to an intact blood-brain-barrier (BBB) and to dose-limiting toxicity of systemic chemotherapeutic agents. Convection-enhanced delivery (CED) into the brainstem may provide an effective local delivery alternative for DIPG patients.The aim of this study is to develop a method to perform CED into the murine brainstem and to test this method using the chemotherapeutic agent carmustine (BiCNU). To this end, a newly designed murine CED catheter was tested in vitro and in vivo. After determination of safety and distribution, mice bearing VUMC-DIPG-3 and E98FM-DIPG brainstem tumors were treated with carmustine dissolved in DW 5% or carmustine dissolved in 10% ethanol.Our results show that CED into the murine brainstem is feasible and well tolerated by mice with and without brainstem tumors. CED of carmustine dissolved in 5% DW increased median survival of mice with VUMC-DIPG-3 and E98FM-DIPG tumors with 35% and 25% respectively. Dissolving carmustine in 10% ethanol further improved survival to 45% in mice with E98FM-DIPG tumors.Since genetically engineered and primary DIPG models are currently only available in mice, murine CED studies have clear advantages over CED studies in other animals.CED in the murine brainstem can be performed safely, is well tolerated and can be used to study efficacy of chemotherapeutic agents orthotopically. These results set the foundation for more CED studies in murine DIPG models.

SummaryThis paper describes the metabolic adaptation of Lactococcus lactis during the transition from a growing to a non‐growing state using retentostat cultivation. Under retentostat cultivation, the specific growth rate decreased from 0.025 h−1 to 0.0001 h−1 in 42 days, while doubling time increased to more than 260 days. Viability of the overall culture was maintained above 90% but included approximately 20% damaged cells, which had lost their colony forming capacity on solid media. Although culture biomass and viability had reached a steady‐state after 14 days of retentostat cultivation, the morphology of the cells changed from coccus‐to‐rod shape at later stages of retentostat cultivation, by which the cell's surface to volume ratio was estimated to increase 2.4‐fold. Furthermore, the metabolic patterns switched between homolactic and mixed‐acid fermentation during the retentostat cultivation. Retentostat cultivation enabled the calculation of accurate substrate‐ and energy‐related maintenance coefficients and biomass yields under non‐growing conditions, which were in good agreement with those calculated by extrapolation from chemostat cultivations at high dilution rates. In this study, we illustrate how retentostat cultivation allows decoupling of growth and non‐growth associated processes in L. lactis, enabling the analysis of quantitative physiological responses of this bacterium to near zero‐specific growth rates.

Improved understanding of recent changes in crop water demand in the dry season is important for the water resources management in the region. A study was carried out to determine the potential impacts of recent climate change during last three decades on trends of water requirements of Boro rice. The reference crop evapotranspiration (ETo), potential crop water requirement (∑ETC), effective rainfall during the crop growing period (ER), potential irrigation requirement for crop evapotranspiration (∑ETC − ER) and net irrigation requirement of Boro rice were estimated using observed daily climate data in the CropWat model for the period of 1980 to 2013 for four North-West districts. Significant decreasing trends of ETo were observed in most of the dry months due to increasing relative humidity and decreasing wind-speed and sun-shine hours. The results showed decreasing trends of potential crop water requirement, i.e. the total crop evapotranspiration (∑ETC), of Boro rice due to decreasing reference crop evapotranspiration and shorter crop growing periods. The variations in trends of potential irrigation requirement for crop evapotranspiration (∑ETC − ER) found among different districts, are mainly linked to variations in trends of changes in effective rainfall. The net irrigation requirement of Boro rice has decreased, by 11% during the last three decades at an average rate of −4.4 mm year−1, instead of decreasing effective rainfall, mainly because of high rate of decrease of crop evapotranspiration (−5.9 mm year−1). Results indicate that a warming climate does not always result in higher agricultural water use and that climate change can also result in reduced water demands because of changes in humidity, wind-speed and sun-shine hours.

In order for this landscape transformation to be sustainable, renewable energy technologies should not cause critical trade-offs between the provision of energy and that of other ecosystem services such as food production. This literature review advances the body of knowledge on sustainable energy transition with special focus on ecosystem services-based approaches and methods. Two key issues emerge from this review: only one sixth of the published applications on the relation between renewable energy and landscape make use of the ecosystem service framework. Secondly, the applications that do address ecosystem services for landscape planning and design lack efficient methods and spatial reference systems that accommodate both cultural and regulating ecosystem services. Future research efforts should be directed to further advancing the spatial reference systems, the use of participatory mapping and landscape visualizations tools for cultural ecosystem services and the elaboration of landscape design principles.

Verdegem;This paper compares the quantity and quality of solid waste obtained from traditional ponds and recirculating aquaculture systems (RAS) for striped catfish and evaluates methane and compost production from these wastes. Striped catfish sludge was collected from four commercial ponds along the Mekong river and from three indoor RAS. The amount of sludge dry matter produced per kilogram of fish in ponds was 6 times higher than that in RAS. However, the concentration of nutrients in solid waste from RAS was much higher, with better compost quality and higher methane yield than that of sludge from ponds. Out of the collected 381 L biogas, the methane yield of striped catfish's solid waste in RAS systems was 201 L per kg chemical oxygen demand (COD). In ponds, the collected 267 L biogas yielded 125 L CH4 per kg COD. The higher methane production from RAS sludge concurred with higher digestibility of COD: 58% for RAS versus 38% for ponds. The quality and quantity of methane from striped catfish sludge were lower than that for other animal manures, and the resulting electricity yield was low. Considering the higher nutrient concentration in RAS-sludge, we recommend the combination RAS and composting in reusing sludge, which is presently the best option for a more sustainable and cleaner striped catfish production system.

Mollenhorst; I.J.M. de Boer;Several dairy farms in the Netherlands aim at reducing their environmental impact by improving the internal nutrient cycle (INC) at farm level. Practices to improve nutrient cycling at these INC farms, however, might not only reduce the environmental impact on-farm, but alter also the off-farm environmental impact associated with supply chain processes (production and transport) related to inputs entering the farm, such as purchased feed or fertilizer or the economic or societal performance of these farms. We compared, therefore, a set of sustainability indicators of nine INC farms with a group of benchmark farms, comparable in terms of farm size, intensity and site-specific circumstances. This benchmark group was composed using statistical matching to exclude the effect of these characteristics on economic, environmental and societal performance. Economic indicators used were: farm income per unpaid annual working unit and the costs to revenues ratio. Environmental indicators used were derived from a cradle-to-farm-gate life cycle assessment: land occupation (LO), non-renewable energy use (NREU), global warming potential (GWP), acidification potential (AP) and eutrophication potential (EP), expressed per kg fat-and-protein-corrected milk (FPCM). In addition, we quantified the soil content of organic carbon and phosphorus, and the soil nitrogen supply. Societal indicators used were: payments for agri-environmental measures, grazing hours and penalties for aberrant milk composition. Results showed that INC farms had a lower non-renewable energy use per kg FPCM, higher soil organic carbon content and received higher annual payments for agri-environmental measures, whereas economic and other environmental, societal indicators did not differed. Furthermore, we demonstrated the need for a sound benchmark to assess the effect of INC-farming on the economic, environmental and societal performance. Statistical matching enabled us to define, for each INC farm, a benchmark group with similar farm characteristics, which are known to affect sustainability indicators. Observed differences in sustainability indicators between both farm groups, therefore, truly resulted from aiming at internal nutrient cycling, and not from differences in other farm characteristics.

Contrary to cellulose, the structure of hemicellulose is quite complex. Therefore, the biodegradation of xylan needs the cooperation of many enzymes. For industrial production of xylanase multienzyme complexes (cocktails) and selected monocomponent xylanases, different Trichoderma reesei mutants and recombinants are used. T. reesei QM 6a (wild-type parent of best existing mutants) was selected as a starting material in the 1960s when the modern in-depth analytical methods were not yet in use. Therefore, screening of fungi genetically close to T. reesei in biodegradation of xylan may have a scientific value. Fifteen different strains from Trichoderma section Longibrachiatum have been tested for extracellular xylan-degrading enzyme production on three carbon sources (wheat straw, corn fiber, and eucalyptus wood) in shake flask cultivation. The enzyme activities were evaluated by traditional colorimetric enzyme assays and by HPLC and matrix assisted laser desorption/ionization time-of-flight mass spectrometry. Degradation of xylan was studied on four different xylan-rich model substrates. T. reesei CPK 155, Trichoderma parareesei TUB F-2535, and Trichoderma gracile TUB F-2543 isolates were equally good or better in degradation of the wheat arabinoxylan (WAX) and corn fiber alcohol insoluble solids as hydolysis substrates than the well-known T. reesei QM 6a and RUT C30 strains. Though Trichoderma saturnisporum ATCC 18903 gave relatively low volumetric enzyme activities by traditional colorimetric assays, it could release quite large amount of hydrolysis products (mono- and oligosaccharides) from WAX. It is shown that more than half of the exergy loss takes place in the liquefaction unit and almost one-third in the air compression unit. Minor exergy losses are taking place in the distillation unit and the main heat exchanger. The major cause of exergy loss is the use of compressors and to a lesser extent the use of turbines. Especially, the relatively low rational efficiency of the turbines operating in the cryogenic region is striking. Improvements are suggested which save one-fourth of the exergy loss. For more substantial reductions of the exergy losses in air separation alternative processes have to be used or developed.

ABSTRACTEnzymatic wheat gluten hydrolysis at high solid concentrations is advantageous from an environmental and economic point of view. However, increased wheat gluten concentrations result in a concentration effect with a decreased hydrolysis rate at constant enzyme‐to‐substrate ratios and a decreased maximum attainable degree of hydrolysis (DH%). We here identified the underlying factors causing the concentration effect. Wheat gluten was hydrolyzed at solid concentrations from 4.4% to 70%. The decreased hydrolysis rate was present at all solid concentrations and at any time of the reaction. Mass transfer limitations, enzyme inhibition and water activity were shown to not cause this hydrolysis rate limitation up to 50% solids. However, the hydrolysis rate limitation can be, at least partly, explained by a second‐order enzyme inactivation process. Furthermore, mass transfer impeded the hydrolysis above 60% solids. Addition of enzyme after 24 h at high solid concentrations scarcely increased the DH%, suggesting that the maximum attainable DH% decreases at high solid concentrations. Reduced enzyme activities caused by low water activities can explain this DH% limitation. Finally, a possible influence of the plastein reaction on the DH% limitation is discussed. Biotechnol. Systemic delivery of therapeutic agents remains ineffective against diffuse intrinsic pontine glioma (DIPG), possibly due to an intact blood-brain-barrier (BBB) and to dose-limiting toxicity of systemic chemotherapeutic agents. Convection-enhanced delivery (CED) into the brainstem may provide an effective local delivery alternative for DIPG patients.The aim of this study is to develop a method to perform CED into the murine brainstem and to test this method using the chemotherapeutic agent carmustine (BiCNU). To this end, a newly designed murine CED catheter was tested in vitro and in vivo. After determination of safety and distribution, mice bearing VUMC-DIPG-3 and E98FM-DIPG brainstem tumors were treated with carmustine dissolved in DW 5% or carmustine dissolved in 10% ethanol.Our results show that CED into the murine brainstem is feasible and well tolerated by mice with and without brainstem tumors. CED of carmustine dissolved in 5% DW increased median survival of mice with VUMC-DIPG-3 and E98FM-DIPG tumors with 35% and 25% respectively. Dissolving carmustine in 10% ethanol further improved survival to 45% in mice with E98FM-DIPG tumors.Since genetically engineered and primary DIPG models are currently only available in mice, murine CED studies have clear advantages over CED studies in other animals.CED in the murine brainstem can be performed safely, is well tolerated and can be used to study efficacy of chemotherapeutic agents orthotopically. These results set the foundation for more CED studies in murine DIPG models.

SummaryThis paper describes the metabolic adaptation of Lactococcus lactis during the transition from a growing to a non‐growing state using retentostat cultivation. Under retentostat cultivation, the specific growth rate decreased from 0.025 h−1 to 0.0001 h−1 in 42 days, while doubling time increased to more than 260 days. Viability of the overall culture was maintained above 90% but included approximately 20% damaged cells, which had lost their colony forming capacity on solid media. Although culture biomass and viability had reached a steady‐state after 14 days of retentostat cultivation, the morphology of the cells changed from coccus‐to‐rod shape at later stages of retentostat cultivation, by which the cell's surface to volume ratio was estimated to increase 2.4‐fold. Furthermore, the metabolic patterns switched between homolactic and mixed‐acid fermentation during the retentostat cultivation. Retentostat cultivation enabled the calculation of accurate substrate‐ and energy‐related maintenance coefficients and biomass yields under non‐growing conditions, which were in good agreement with those calculated by extrapolation from chemostat cultivations at high dilution rates. In this study, we illustrate how retentostat cultivation allows decoupling of growth and non‐growth associated processes in L. lactis, enabling the analysis of quantitative physiological responses of this bacterium to near zero‐specific growth rates.

Improved understanding of recent changes in crop water demand in the dry season is important for the water resources management in the region. A study was carried out to determine the potential impacts of recent climate change during last three decades on trends of water requirements of Boro rice. The reference crop evapotranspiration (ETo), potential crop water requirement (∑ETC), effective rainfall during the crop growing period (ER), potential irrigation requirement for crop evapotranspiration (∑ETC − ER) and net irrigation requirement of Boro rice were estimated using observed daily climate data in the CropWat model for the period of 1980 to 2013 for four North-West districts. Significant decreasing trends of ETo were observed in most of the dry months due to increasing relative humidity and decreasing wind-speed and sun-shine hours. The results showed decreasing trends of potential crop water requirement, i.e. the total crop evapotranspiration (∑ETC), of Boro rice due to decreasing reference crop evapotranspiration and shorter crop growing periods. The variations in trends of potential irrigation requirement for crop evapotranspiration (∑ETC − ER) found among different districts, are mainly linked to variations in trends of changes in effective rainfall. The net irrigation requirement of Boro rice has decreased, by 11% during the last three decades at an average rate of −4.4 mm year−1, instead of decreasing effective rainfall, mainly because of high rate of decrease of crop evapotranspiration (−5.9 mm year−1). Results indicate that a warming climate does not always result in higher agricultural water use and that climate change can also result in reduced water demands because of changes in humidity, wind-speed and sun-shine hours.

In order for this landscape transformation to be sustainable, renewable energy technologies should not cause critical trade-offs between the provision of energy and that of other ecosystem services such as food production. This literature review advances the body of knowledge on sustainable energy transition with special focus on ecosystem services-based approaches and methods. Two key issues emerge from this review: only one sixth of the published applications on the relation between renewable energy and landscape make use of the ecosystem service framework. Secondly, the applications that do address ecosystem services for landscape planning and design lack efficient methods and spatial reference systems that accommodate both cultural and regulating ecosystem services. Future research efforts should be directed to further advancing the spatial reference systems, the use of participatory mapping and landscape visualizations tools for cultural ecosystem services and the elaboration of landscape design principles.

Verdegem;This paper compares the quantity and quality of solid waste obtained from traditional ponds and recirculating aquaculture systems (RAS) for striped catfish and evaluates methane and compost production from these wastes. Striped catfish sludge was collected from four commercial ponds along the Mekong river and from three indoor RAS. The amount of sludge dry matter produced per kilogram of fish in ponds was 6 times higher than that in RAS. However, the concentration of nutrients in solid waste from RAS was much higher, with better compost quality and higher methane yield than that of sludge from ponds. Out of the collected 381 L biogas, the methane yield of striped catfish's solid waste in RAS systems was 201 L per kg chemical oxygen demand (COD). In ponds, the collected 267 L biogas yielded 125 L CH4 per kg COD. The higher methane production from RAS sludge concurred with higher digestibility of COD: 58% for RAS versus 38% for ponds. The quality and quantity of methane from striped catfish sludge were lower than that for other animal manures, and the resulting electricity yield was low. Considering the higher nutrient concentration in RAS-sludge, we recommend the combination RAS and composting in reusing sludge, which is presently the best option for a more sustainable and cleaner striped catfish production system.

Mollenhorst; I.J.M. de Boer;Several dairy farms in the Netherlands aim at reducing their environmental impact by improving the internal nutrient cycle (INC) at farm level. Practices to improve nutrient cycling at these INC farms, however, might not only reduce the environmental impact on-farm, but alter also the off-farm environmental impact associated with supply chain processes (production and transport) related to inputs entering the farm, such as purchased feed or fertilizer or the economic or societal performance of these farms. We compared, therefore, a set of sustainability indicators of nine INC farms with a group of benchmark farms, comparable in terms of farm size, intensity and site-specific circumstances. This benchmark group was composed using statistical matching to exclude the effect of these characteristics on economic, environmental and societal performance. Economic indicators used were: farm income per unpaid annual working unit and the costs to revenues ratio. Environmental indicators used were derived from a cradle-to-farm-gate life cycle assessment: land occupation (LO), non-renewable energy use (NREU), global warming potential (GWP), acidification potential (AP) and eutrophication potential (EP), expressed per kg fat-and-protein-corrected milk (FPCM). In addition, we quantified the soil content of organic carbon and phosphorus, and the soil nitrogen supply. Societal indicators used were: payments for agri-environmental measures, grazing hours and penalties for aberrant milk composition. Results showed that INC farms had a lower non-renewable energy use per kg FPCM, higher soil organic carbon content and received higher annual payments for agri-environmental measures, whereas economic and other environmental, societal indicators did not differed. Furthermore, we demonstrated the need for a sound benchmark to assess the effect of INC-farming on the economic, environmental and societal performance. Statistical matching enabled us to define, for each INC farm, a benchmark group with similar farm characteristics, which are known to affect sustainability indicators. Observed differences in sustainability indicators between both farm groups, therefore, truly resulted from aiming at internal nutrient cycling, and not from differences in other farm characteristics.

Contrary to cellulose, the structure of hemicellulose is quite complex. Therefore, the biodegradation of xylan needs the cooperation of many enzymes. For industrial production of xylanase multienzyme complexes (cocktails) and selected monocomponent xylanases, different Trichoderma reesei mutants and recombinants are used. T. reesei QM 6a (wild-type parent of best existing mutants) was selected as a starting material in the 1960s when the modern in-depth analytical methods were not yet in use. Therefore, screening of fungi genetically close to T. reesei in biodegradation of xylan may have a scientific value. Fifteen different strains from Trichoderma section Longibrachiatum have been tested for extracellular xylan-degrading enzyme production on three carbon sources (wheat straw, corn fiber, and eucalyptus wood) in shake flask cultivation. The enzyme activities were evaluated by traditional colorimetric enzyme assays and by HPLC and matrix assisted laser desorption/ionization time-of-flight mass spectrometry. Degradation of xylan was studied on four different xylan-rich model substrates. T. reesei CPK 155, Trichoderma parareesei TUB F-2535, and Trichoderma gracile TUB F-2543 isolates were equally good or better in degradation of the wheat arabinoxylan (WAX) and corn fiber alcohol insoluble solids as hydolysis substrates than the well-known T. reesei QM 6a and RUT C30 strains. Though Trichoderma saturnisporum ATCC 18903 gave relatively low volumetric enzyme activities by traditional colorimetric assays, it could release quite large amount of hydrolysis products (mono- and oligosaccharides) from WAX. It is shown that more than half of the exergy loss takes place in the liquefaction unit and almost one-third in the air compression unit. Minor exergy losses are taking place in the distillation unit and the main heat exchanger. The major cause of exergy loss is the use of compressors and to a lesser extent the use of turbines. Especially, the relatively low rational efficiency of the turbines operating in the cryogenic region is striking. Improvements are suggested which save one-fourth of the exergy loss. For more substantial reductions of the exergy losses in air separation alternative processes have to be used or developed.
