In this study, we aimed at studying the variation and genetic control of WUE and the respective role of its components (plant biomass and transpiration) in a perennial fruit crop. We explored an INRA apple core collection grown in a phenotyping platform to screen one-year-old scions for their accumulated biomass, transpiration and WUE under optimal growing conditions. Plant biomass was decompose into morphological components related to either growth or organ expansion. For each trait, nine mixed models were evaluated to account for the genetic effect and spatial heterogeneity inside the platform. The Best Linear Unbiased Predictors of genetic values were estimated after model selection. Mean broad-sense heritabilities were calculated from variance estimates. Heritability values indicated that biomass (0.76) and WUE (0.73) were under genetic control. This genetic control was lower in plant transpiration with an heritability of 0.54. Across the collection, biomass accounted for 70% of the WUE variability. A Hierarchical Ascendant Classification of the core collection indicated the existence of six groups of genotypes with contrasting morphology and WUE. Differences between morphotypes were interpreted as resulting from differences in the main processes responsible for plant growth: cell division leading to the generation of new organs and cell elongation leading to organ dimension. Although further studies will be necessary on mature trees with more complex architecture and multiple sinks such as fruits, this study is a first step for improving apple plant material for the use of water.

Bacterial contamination causes yield reduction during ethanol production from molasses. To prevent contamination, construction of a fermentation system using acetate-tolerant yeast under an acetate-containing condition was attempted. Schizosaccharomyces pombe was screened as an acetate-tolerant strain. Bacterial contamination was significantly prevented by the combined use of Sc. pombe and acetate.

The rocket plants were cultivated on an infested substrate (4 log CFU g-1) and a non-infested substrate over three cycles. Pots were placed in six phytotrons in order to simulate different environmental conditions: 1) 400-450 ppm CO2, 18-22°C; 2) 800-850 ppm CO2, 18-22°C; 3) 400-450 ppm CO2, 22-26°C, 4) 800-850 ppm CO2, 22-26°C, 5) 400-450 ppm CO2, 26-30°C; 6) 800-850 ppm CO2, 26-30°C. Substrates from the infested and control samples were collected from each phytotron at 0, 60 and 120 days after transplanting. The disease index, microbial abundance, leaf physiological performances, root exudates and variability in the fungal profiles were monitored. The disease index was found to be significantly influenced by higher levels of temperature and CO2. Plate counts showed that fungal and bacterial development was not affected by the different CO2 and temperature levels, but a significant decreasing trend was observed from 0 up to 120 days. Conversely, the F. oxysporum f.sp. conglutinans plate counts did not show any significantly decrease from 0 up to 120 days. The fungal profiles, evaluated by means of polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE), showed a relationship to temperature and CO2 on fungal diversity profiles. Different exudation patterns were observed when the controls and infested plants were compared, and it was found that both CO2 and temperature can influence the release of compounds from the roots of rocket plants. In short, the results show that global climate changes could influence disease incidence, probably through plant-mediated effects, caused by soilborne pathogens.

Nipa palms, a plant species in mangrove forest, are valuable and beneficial for the local community’s economy as well as the conservation in the southern region of Thailand. This study aimed to investigate the use of nipa palms in Khanap Nak and focused on the type of products made from nipa palms and the yield of this plant through focus group discussion and interview with the farmers maintaining nipa palm forest. The results suggested that nipa palms in Khanap Nak can yield for 5–100 years. Apart from the benefits to the community in terms of sustainability, as it prevents erosion, it provides sources for the production of food (molasses, granulated sugar, syrup, and vinegar from sap) and raw materials (roofing material and cigarette paper from leaves and stem), which can generate income to community members. It was found that most of the communities earn 90–130 USD/day from palm sugar production. The quantity of nipa palm products varies in each season, resulting in different prices. In this regard, they can produce high quantity of products made from sap from January to March. However, the production of different types of sugar requires local knowledge and wisdom to ensure good quality. Nipa palm production is the sustainable way to utilize mangrove forest resources, leading to effective conservation and good life quality. Regarding problems and difficulties in farming, it was found that natural disaster is a major threat, such as drought, excessive amount of salt or freshwater in certain periods, and insect pests.

Allison;AbstractThe study reports on the effect of anaerobic digestate derived composts on the metabolite composition and thermal behaviour of rosemary (Rosmarinus officinalis L.). Plants were cultivated in semiarid soil under four different fertiliser treatments (composts of anaerobic digested cattle (C) or pig slurry (P) at 30t/ha and 60 t/ha, and two control treatments (inorganic fertiliser and no fertiliser application). Samples of leaves and stems were analysed to investigate the effect of treatment on chemical composition and thermochemical properties. Three orthogonal analytical approaches were used, namely: Fourier transform mid infrared spectroscopy (FTIR), gas chromatography/mass spectrometry (GC/MS) and thermochemical gravimetric analysis (TGA). FTIR and GC/MS showed fertiliser treatment resulted in tissue specific changes in sample metabolite composition. Fertiliser treatment was detected to change the thermogravimetric properties of the leaf samples and from inorganic and composted pig slurry digestate treatments had greater ash content and lower proportions of fixed carbon compared with samples from the unfertilised control treatment. This study provides information on how the composition of rosemary might be altered by fertiliser application in regions of poor soil, and has implications for biomass quality when rosemary is grown on semi-wild sites for the purpose of soil improvement.

In eucalyptus plantations, water availability is a key factor limiting productivity. However, the molecular mechanisms underlying the adaptation of eucalyptus to water shortage remain unclear. In this study, we compared the molecular responses of two commercial eucalyptus hybrids during the dry season. Both hybrids differ in productivity when grown under water deficit. Results Pyrosequencing of RNA extracted from shoot apices provided extensive transcriptome coverage - a catalog of 129,993 unigenes (49,748 contigs and 80,245 singletons) was generated from 398 million base pairs, or 1.14 million reads. The pyrosequencing data enriched considerably existing Eucalyptus EST collections, adding 36,985 unigenes not previously represented. Digital analysis of read abundance in 14,460 contigs identified 1,280 that were differentially expressed between the two genotypes, 155 contigs showing differential expression between treatments (irrigated vs. non irrigated conditions during the dry season), and 274 contigs with significant genotype-by-treatment interaction. The more productive genotype displayed a larger set of genes responding to water stress. Moreover, stress signal transduction seemed to involve different pathways in the two genotypes, suggesting that water shortage induces distinct cellular stress cascades. Similarly, the response of functional proteins also varied widely between genotypes: the most productive genotype decreased expression of genes related to photosystem, transport and secondary metabolism, whereas genes related to primary metabolism and cell organisation were over-expressed. Conclusions For the most productive genotype, the ability to express a broader set of genes in response to water availability appears to be a key characteristic in the maintenance of biomass growth during the dry season. Its strategy may involve a decrease of photosynthetic activity during the dry season associated with resources reallocation through major changes in the expression of primary metabolism associated genes. Further efforts will be needed to assess the adaptive nature of the genes highlighted in this study.

After accumulation in the liver of animals they inhibit eukaryotic serine/threonine protein phosphatases, causing liver disease or death. Accurate detection/quantification of microcystins is essential to ensure safe water resources and to enable research on this toxin. Previous methodological comparisons have focused on detection and extraction techniques, but have not investigated the commonly used biomass enrichment steps. These enrichment steps could modulate toxin production as recent studies have demonstrated that high cyanobacterial cell densities cause increased microcystin levels. In this study, three microcystin-producing strains were processed using no cell enrichment steps (by direct freezing at three temperatures) and with biomass enrichment (by centrifugation or GF/C filtration). After extraction, microcystins were analyzed using liquid chromatography-tandem mass spectrometry. All processing methods tested, except GF/C filtration, resulted in comparable microcystin quotas for all strains. The low yields observed for the filtration samples were caused by adsorption of arginine-containing microcystins to the GF/C filters. Whilst biomass enrichment did not affect microcystin metabolism over the time-frame of normal sample processing, problems associated with GF/C filtration were identified. The most widely applicable processing method was direct freezing of samples as it could be utilized in both field and laboratory environments.

The stability of ethyl glucuronide (EtG) under conditions of degradation was examined in urine samples of nine volunteers and in post-mortem tissue (liver, skeletal muscle) and blood taken from seven corpses at autopsies. Analysis was performed via LC-MS/MS. EtG concentrations in urine samples ranged from 2.5 to 296.5 mg/l. When stored at 4 degrees C in airtight test tubes, EtG concentrations remained relatively constant; when stored at room temperature (RT) for 5 weeks in ventilated vials, variations of EtG concentrations ranged from a 30% decrease to an 80% increase, with an average of 37.5% increase. Liver and skeletal muscle tissue of three corpses with positive blood alcohol concentrations (BAC; ranging from 0.106 to 0.183 g%) were stored for 4 weeks and analysed periodically. EtG concentrations decreased 27.7% on average in 4 weeks storage at RT but EtG was still detectable in all samples with initial EtG concentrations higher than 1 mug/g. Blood and liver samples of four corpses with negative BACs were stored at RT after addition of 0.1 g% ethanol, and no new formation of EtG was observed.

The leaf features like trichome density, gradient grooves, and leaf wettability determine the efficiency to capture air moisture for self-irrigation in the wheat plant. Plants in water-scarce environments evolved to capture air moisture for their water needs either directly or indirectly. Structural features like cones, hairs, and grooves assist water capture. The morphology of crops such as wheat can promote self-irrigation under drought. To examine this further, 34 wheat genotypes were characterized for leaf traits in near optimal conditions in the field using a randomized complete block design with 3 replications. An association was found between morphological and physiological traits and yield using simple correlation plots. A core set of nine genotypes was subsequently evaluated for moisture harvesting ability and leaf wettability. Results showed that variation among genotypes exists for fog harvesting ability attributed to structural leaf features. Physiological traits, especially photosynthesis and water use efficiency, were positively associated with yield, negatively correlated with soil moisture at booting, and positively correlated with soil moisture at anthesis. The genotypes with deep to medium leaf grooves and dense hairs on the edges and adaxial surfaces (genotypes 7 and 18) captured the most moisture. This was a function of higher water drop rolling efficiency resulting from lower contact angle hysteresis. These results can be exploited to develop more heat and drought-tolerant crops.

Phagotrophic protists are an important mortality factor of prokaryotes in most aquatic habitats. However, no study has assessed protistan grazing as loss factor of bacterial biomass across the stratification gradient of a temperate freshwater meromictic lake. Protistan grazing effect was quantified in the mixolimnion, the transition zone, and the sulfidic anoxic monimolimnion of Lake Alatsee (Germany). Grazing experiments were performed using prey analogues from the natural prokaryotic assemblage. Daily grazing effect declined from the mixolimnion to the monimolimnion. Heterotrophic flagellates were phagotrophically active in all three water horizons and the main grazers in the monimolimnion. Pigmented flagellates accounted for 70% of total grazing in the mixolimnion and ciliates only for a small fraction of grazing in each depth. Prokaryotic biomass removal peaked in the interface, but protistan impact on the respective prokaryotic abundance was low. Grazing in the anoxic monimolimnion was negligible, with prokaryotic turnover rate being only 0.4% of standing stock. Our results support the assumption that protistan predation in anoxic waters is lower than in oxygenated ones and identify the interface as a microhabitat that supports high grazer biomass, pinpointing the importance of purple sulfur bacteria as carbon source for the upper mixolimnion and the bottom monimolimnion.

In this study, we aimed at studying the variation and genetic control of WUE and the respective role of its components (plant biomass and transpiration) in a perennial fruit crop. We explored an INRA apple core collection grown in a phenotyping platform to screen one-year-old scions for their accumulated biomass, transpiration and WUE under optimal growing conditions. Plant biomass was decompose into morphological components related to either growth or organ expansion. For each trait, nine mixed models were evaluated to account for the genetic effect and spatial heterogeneity inside the platform. The Best Linear Unbiased Predictors of genetic values were estimated after model selection. Mean broad-sense heritabilities were calculated from variance estimates. Heritability values indicated that biomass (0.76) and WUE (0.73) were under genetic control. This genetic control was lower in plant transpiration with an heritability of 0.54. Across the collection, biomass accounted for 70% of the WUE variability. A Hierarchical Ascendant Classification of the core collection indicated the existence of six groups of genotypes with contrasting morphology and WUE. Differences between morphotypes were interpreted as resulting from differences in the main processes responsible for plant growth: cell division leading to the generation of new organs and cell elongation leading to organ dimension. Although further studies will be necessary on mature trees with more complex architecture and multiple sinks such as fruits, this study is a first step for improving apple plant material for the use of water.

Bacterial contamination causes yield reduction during ethanol production from molasses. To prevent contamination, construction of a fermentation system using acetate-tolerant yeast under an acetate-containing condition was attempted. Schizosaccharomyces pombe was screened as an acetate-tolerant strain. Bacterial contamination was significantly prevented by the combined use of Sc. pombe and acetate.

The rocket plants were cultivated on an infested substrate (4 log CFU g-1) and a non-infested substrate over three cycles. Pots were placed in six phytotrons in order to simulate different environmental conditions: 1) 400-450 ppm CO2, 18-22°C; 2) 800-850 ppm CO2, 18-22°C; 3) 400-450 ppm CO2, 22-26°C, 4) 800-850 ppm CO2, 22-26°C, 5) 400-450 ppm CO2, 26-30°C; 6) 800-850 ppm CO2, 26-30°C. Substrates from the infested and control samples were collected from each phytotron at 0, 60 and 120 days after transplanting. The disease index, microbial abundance, leaf physiological performances, root exudates and variability in the fungal profiles were monitored. The disease index was found to be significantly influenced by higher levels of temperature and CO2. Plate counts showed that fungal and bacterial development was not affected by the different CO2 and temperature levels, but a significant decreasing trend was observed from 0 up to 120 days. Conversely, the F. oxysporum f.sp. conglutinans plate counts did not show any significantly decrease from 0 up to 120 days. The fungal profiles, evaluated by means of polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE), showed a relationship to temperature and CO2 on fungal diversity profiles. Different exudation patterns were observed when the controls and infested plants were compared, and it was found that both CO2 and temperature can influence the release of compounds from the roots of rocket plants. In short, the results show that global climate changes could influence disease incidence, probably through plant-mediated effects, caused by soilborne pathogens.

Nipa palms, a plant species in mangrove forest, are valuable and beneficial for the local community’s economy as well as the conservation in the southern region of Thailand. This study aimed to investigate the use of nipa palms in Khanap Nak and focused on the type of products made from nipa palms and the yield of this plant through focus group discussion and interview with the farmers maintaining nipa palm forest. The results suggested that nipa palms in Khanap Nak can yield for 5–100 years. Apart from the benefits to the community in terms of sustainability, as it prevents erosion, it provides sources for the production of food (molasses, granulated sugar, syrup, and vinegar from sap) and raw materials (roofing material and cigarette paper from leaves and stem), which can generate income to community members. It was found that most of the communities earn 90–130 USD/day from palm sugar production. The quantity of nipa palm products varies in each season, resulting in different prices. In this regard, they can produce high quantity of products made from sap from January to March. However, the production of different types of sugar requires local knowledge and wisdom to ensure good quality. Nipa palm production is the sustainable way to utilize mangrove forest resources, leading to effective conservation and good life quality. Regarding problems and difficulties in farming, it was found that natural disaster is a major threat, such as drought, excessive amount of salt or freshwater in certain periods, and insect pests.

Plants were cultivated in semiarid soil under four different fertiliser treatments (composts of anaerobic digested cattle (C) or pig slurry (P) at 30t/ha and 60 t/ha, and two control treatments (inorganic fertiliser and no fertiliser application). Samples of leaves and stems were analysed to investigate the effect of treatment on chemical composition and thermochemical properties. Three orthogonal analytical approaches were used, namely: Fourier transform mid infrared spectroscopy (FTIR), gas chromatography/mass spectrometry (GC/MS) and thermochemical gravimetric analysis (TGA). FTIR and GC/MS showed fertiliser treatment resulted in tissue specific changes in sample metabolite composition. Fertiliser treatment was detected to change the thermogravimetric properties of the leaf samples and from inorganic and composted pig slurry digestate treatments had greater ash content and lower proportions of fixed carbon compared with samples from the unfertilised control treatment. This study provides information on how the composition of rosemary might be altered by fertiliser application in regions of poor soil, and has implications for biomass quality when rosemary is grown on semi-wild sites for the purpose of soil improvement.

Results Pyrosequencing of RNA extracted from shoot apices provided extensive transcriptome coverage - a catalog of 129,993 unigenes (49,748 contigs and 80,245 singletons) was generated from 398 million base pairs, or 1.14 million reads. The pyrosequencing data enriched considerably existing Eucalyptus EST collections, adding 36,985 unigenes not previously represented. Digital analysis of read abundance in 14,460 contigs identified 1,280 that were differentially expressed between the two genotypes, 155 contigs showing differential expression between treatments (irrigated vs. non irrigated conditions during the dry season), and 274 contigs with significant genotype-by-treatment interaction. The more productive genotype displayed a larger set of genes responding to water stress. Moreover, stress signal transduction seemed to involve different pathways in the two genotypes, suggesting that water shortage induces distinct cellular stress cascades. Similarly, the response of functional proteins also varied widely between genotypes: the most productive genotype decreased expression of genes related to photosystem, transport and secondary metabolism, whereas genes related to primary metabolism and cell organisation were over-expressed. Conclusions For the most productive genotype, the ability to express a broader set of genes in response to water availability appears to be a key characteristic in the maintenance of biomass growth during the dry season. Its strategy may involve a decrease of photosynthetic activity during the dry season associated with resources reallocation through major changes in the expression of primary metabolism associated genes. Further efforts will be needed to assess the adaptive nature of the genes highlighted in this study.

After accumulation in the liver of animals they inhibit eukaryotic serine/threonine protein phosphatases, causing liver disease or death. Accurate detection/quantification of microcystins is essential to ensure safe water resources and to enable research on this toxin. Previous methodological comparisons have focused on detection and extraction techniques, but have not investigated the commonly used biomass enrichment steps. These enrichment steps could modulate toxin production as recent studies have demonstrated that high cyanobacterial cell densities cause increased microcystin levels. In this study, three microcystin-producing strains were processed using no cell enrichment steps (by direct freezing at three temperatures) and with biomass enrichment (by centrifugation or GF/C filtration). After extraction, microcystins were analyzed using liquid chromatography-tandem mass spectrometry. All processing methods tested, except GF/C filtration, resulted in comparable microcystin quotas for all strains. The low yields observed for the filtration samples were caused by adsorption of arginine-containing microcystins to the GF/C filters. Whilst biomass enrichment did not affect microcystin metabolism over the time-frame of normal sample processing, problems associated with GF/C filtration were identified. The most widely applicable processing method was direct freezing of samples as it could be utilized in both field and laboratory environments.

The stability of ethyl glucuronide (EtG) under conditions of degradation was examined in urine samples of nine volunteers and in post-mortem tissue (liver, skeletal muscle) and blood taken from seven corpses at autopsies. Analysis was performed via LC-MS/MS. EtG concentrations in urine samples ranged from 2.5 to 296.5 mg/l. When stored at 4 degrees C in airtight test tubes, EtG concentrations remained relatively constant; when stored at room temperature (RT) for 5 weeks in ventilated vials, variations of EtG concentrations ranged from a 30% decrease to an 80% increase, with an average of 37.5% increase. Liver and skeletal muscle tissue of three corpses with positive blood alcohol concentrations (BAC; ranging from 0.106 to 0.183 g%) were stored for 4 weeks and analysed periodically. EtG concentrations decreased 27.7% on average in 4 weeks storage at RT but EtG was still detectable in all samples with initial EtG concentrations higher than 1 mug/g. Blood and liver samples of four corpses with negative BACs were stored at RT after addition of 0.1 g% ethanol, and no new formation of EtG was observed.

The leaf features like trichome density, gradient grooves, and leaf wettability determine the efficiency to capture air moisture for self-irrigation in the wheat plant. Plants in water-scarce environments evolved to capture air moisture for their water needs either directly or indirectly. Structural features like cones, hairs, and grooves assist water capture. The morphology of crops such as wheat can promote self-irrigation under drought. To examine this further, 34 wheat genotypes were characterized for leaf traits in near optimal conditions in the field using a randomized complete block design with 3 replications. An association was found between morphological and physiological traits and yield using simple correlation plots. A core set of nine genotypes was subsequently evaluated for moisture harvesting ability and leaf wettability. Results showed that variation among genotypes exists for fog harvesting ability attributed to structural leaf features. Physiological traits, especially photosynthesis and water use efficiency, were positively associated with yield, negatively correlated with soil moisture at booting, and positively correlated with soil moisture at anthesis. The genotypes with deep to medium leaf grooves and dense hairs on the edges and adaxial surfaces (genotypes 7 and 18) captured the most moisture. This was a function of higher water drop rolling efficiency resulting from lower contact angle hysteresis. These results can be exploited to develop more heat and drought-tolerant crops.

Phagotrophic protists are an important mortality factor of prokaryotes in most aquatic habitats. However, no study has assessed protistan grazing as loss factor of bacterial biomass across the stratification gradient of a temperate freshwater meromictic lake. Protistan grazing effect was quantified in the mixolimnion, the transition zone, and the sulfidic anoxic monimolimnion of Lake Alatsee (Germany). Grazing experiments were performed using prey analogues from the natural prokaryotic assemblage. Daily grazing effect declined from the mixolimnion to the monimolimnion. Heterotrophic flagellates were phagotrophically active in all three water horizons and the main grazers in the monimolimnion. Pigmented flagellates accounted for 70% of total grazing in the mixolimnion and ciliates only for a small fraction of grazing in each depth. Prokaryotic biomass removal peaked in the interface, but protistan impact on the respective prokaryotic abundance was low. Grazing in the anoxic monimolimnion was negligible, with prokaryotic turnover rate being only 0.4% of standing stock. Our results support the assumption that protistan predation in anoxic waters is lower than in oxygenated ones and identify the interface as a microhabitat that supports high grazer biomass, pinpointing the importance of purple sulfur bacteria as carbon source for the upper mixolimnion and the bottom monimolimnion.
