L.;An aerobic granular bioreactor was operated for over 4months, treating a synthetic wastewater with a high ammonium content (100mgNL-1). The inoculum was collected from a bioreactor performing simultaneous partial nitrification and aromatic compounds biodegradation. From day-56 onwards, 2-fluorophenol (2-FP) (12.4mgL-1) was added to the feeding wastewater and the system was bioaugmented with a 2-FP degrading bacteria (Rhodococcus sp. FP1). By the end of operation, complete 2-FP biodegradation and partial nitrification were simultaneously achieved. Aerobic granules remained stable over time. During the 2-FP loading, a shift in the community structure occurred, coinciding with the improvement of 2-FP degradation. DGGE analysis did not allow to infer on the bioaugmented strain presence but pyrosequencing analysis detected Rhodococcus genus by the end of operation. Together with other potential phenolic-degraders within granules, these microorganisms were probably responsible for 2-FP degradation.

The use of heavy metals (HM) contaminated soils to grow energy crops can diminish the negative impact of HM in the environment improving land restoration. The effect of two PGPR (B1--Chryseobacterium humi ECP37(T) and B2--Pseudomonas reactans EDP28) and an AMF (F--Rhizophagus irregularis) on growth, Cd and Zn accumulation, and nutritional status of energy maize plants grown in a soil collected from an area adjacent to a Portuguese mine was assessed in a greenhouse experiment. Both bacterial strains, especially when co-inoculated with the AMF, acted as plant growth-promoting inoculants, increasing root and shoot biomass as well as shoot elongation. Cadmium was not detected in the maize tissues and a decrease in Zn accumulation was observed for all microbial treatments in aboveground and belowground tissues--with inoculation of maize with AMF and strain B2 leading to maximum reductions in Zn shoot and root accumulation of up to 48 and 43%, respectively. Although microbial single inoculation generally did not increase N and P levels in maize plants, co-inoculation of the PGPR and the AMF improved substantially P accumulation in roots. The DGGE analysis of the bacterial rhizosphere community showed that the samples inoculated with the AMF clustered apart of those without the AMF and the Shannon-Wiener Index (H') increased over the course of the experiment when both inoculants were present. This work shows the benefits of combined inoculation of AMF and PGPR for the growth energy maize in metal contaminated soils and their potential for the application in phytomanagement strategies.

Cadmium is one of the most toxic heavy metals and its accumulation in the upper layers of forest soils affects plants, microorganisms and their interactions. Adequate strategies for the reforestation of metal contaminated sites are of vital importance. The aim of this work was to evaluate the response of Pinus pinaster seedlings to Cd exposure and to assess the effect of inoculation with two selected ectomycorrhizal fungi, Suillus bovinus and Rhizopogon roseolus on that response. Seedlings were exposed to soil contaminated at 15 and 30 mg Cd kg(-1). Shoot biomass of P. pinaster decreased ca. 36% when exposed to 15 mg Cd kg(-1). Overall, colonization by S. bovinus significantly enhanced shoot development up to 30% in contaminated soil while colonization by R. roseolus produced no significant effect at both Cd concentrations tested and significantly increased the level of Cd in the shoots at both Cd concentrations. Metal accumulation in the shoots and roots of non-inoculated and S. bovinus-inoculated seedlings increased at the higher Cd levels whereas R. roseolus-inoculated seedlings were not sensitive to Cd variation in the soil. The results from our research show that inoculation with ECM fungi has a significant impact on metal uptake and development of P. pinaster seedlings; the differential response induced by the two tested species highlights the importance of selecting the appropriate strains for nursery inoculation, and, as such, this biological tool ought to be considered in reforestation processes of heavy metal contaminated areas by woody species.

Zn accumulation in Solanum nigrum grown in naturally contaminated soil in the presence of different types of organic amendments was assessed. Under the same conditions, the response of the plant to inoculation with two different isolates of arbuscular mycorrhizal fungi (AMF) (Glomus claroideum and Glomus intraradices) was also evaluated. S. nigrum grown in the non-amended soil always presented higher Zn accumulation in the tissues, with the addition of amendments inducing reductions of up to 80 and 40%, for manure and compost, respectively, and enhancing plant biomass yields. The establishment of S. nigrum in the Zn contaminated soil combined with the application of amendments led to a 70-80% reduction in the amount of Zn leached through the soil. The use of S. nigrum in combination with manure appeared as an effective method for reducing the effects of soil contamination, diminishing Zn transfer to other environmental compartments via percolation.

Zn tissue accumulation in Solanum nigrum grown in a non-contaminated and a naturally contaminated Zn matrix and the effect of inoculation with different arbuscular mycorrhizal fungi (AMF) on metal uptake were assessed. S. nigrum grown in the contaminated soil always presented higher Zn accumulation in the tissues, accumulating up to 1622 mg Zn kg(-1). The presence of both Glomus claroideum and Glomus intraradices enhanced the uptake and accumulation of Zn by S. nigrum (up to 83 and 49% higher Zn accumulation, respectively). The main deposits of the metal were found in the intercellular spaces and in the cell walls of the root tissues, as revealed by autometallography, with the inoculation with different AMF species causing no differences in the location of Zn accumulation. These findings indicate that S. nigrum inoculated with selected heavy metal tolerant AMF presents extracting and accumulating capacities, constituting a potentially suitable remediation method for Zn polluted soils.

An up-flow fixed bed reactor (UFBR) was established to investigate the biodegradation of fluorobenzene (FB) under a number of operating conditions, which included variation in the concentration of FB in the feed stream (up to 180 mg l(-1)) and temporary suspension of feeding. Degradation of FB was followed for a period of 8 months under a continuous flow regime. During the operation of the UFBR, FB was never detected in the reactor effluent, being biodegraded by the microbial biofilm or adsorbed to the granular activated carbon (GAC). Biodegradation of FB was observed from the beginning of the reactor operation, and overall, it accounted for 50% of the total amount fed to the bioreactor. High organic loads of FB (210-260 mg d(-1) dm(-3)) were found to affect the biological removal efficiency, possibly due to an inhibitory effect caused by the higher FB concentrations fed to the bioreactor (149-179 mg l(-1)). When FB feeding was suspended for 1 month, biodegradation continued, indicating that the adsorbed FB became bioavailable. Biofilm bacterial dynamics were followed throughout the UFBR operation by denaturing gradient gel electrophoresis and plate-counting techniques, showing that a quite stable community was found in the bioreactor, and this was mainly attributed to the high selective pressure exerted by the presence of FB.

Microalgae are ubiquitous in aquatic environments, including aquaculture farms, but few studies have delved into their phytoplankton diversity and bioremediation potential. In this study, the cultivable phytoplankton of a rainbow trout freshwater aquaculture farm was isolated, phylogenetically analysed and used to assemble a consortium to polish an aquaculture-derived effluent, with low concentrations of ammonium, nitrite and nitrate.Through standard plating in different selective media, a total of 15 microalgae strains were isolated from sludge from a rotary drum filtering system which removes suspended solids from the water exiting the facility. Based on 18S rRNA gene sequences, isolates were assigned to nine different genera of the Chlorophyta phylum: Asterarcys, Chlorella, Chloroccocum, Chlorosarcinopsis, Coelastrella, Desmodesmus, Micractinium, Parachlorella and Scenedesmus. Species from most of these genera are known to inhabit freshwater systems in Galicia and continental Spain, but the Coelastrella, Asterarcys or Parachlorella genera are not usually present in freshwater streams. In an onsite integrative approach, the capacity of a consortium of native microalgae isolates to grow on aquaculture-derived effluents and its nutrient removal capacity were assessed using a raceway pond. After 7 days, removal efficiencies of approximately 99%, 92% and 49% for ammonium, nitrite and nitrate, respectively, were achieved concomitantly with a microalgae biomass increase of ca. 17%.Sludge from the aquaculture filtering system presents a high diversity of microalgae species from the Chlorophyta phylum, whose application in a consortial approach revealed to be efficient to polish aquaculture-derived effluents with low nutrient content.The use of native microalgae consortia from aquaculture systems can contribute to the development of efficient treatment systems for low-nutrient wastewater, avoiding nutrients release to the environment and promoting water recirculation. This may further strengthen the use of phycoremediation at the industrial scale, as an environment-friendly strategy.

Toxicity tests were performed to assess the effect of tannery wastewater with different treatment levels on two wetland plants, Phragmites australis and Typha latifolia, which are frequently used in constructed wetlands (CWs) for water treatment, and thus deepen the knowledge on their capacity to withstand the application of industrial wastewater. Trifolium pratense, a plant generally used as an indicator in toxicity tests, was included as a control. End points measured were germination percentage, shoot length, root elongation, and biomass growth of the plants. When tannery effluent, with a low treatment level, was supplied to the wetland plants germination occurred even at effluent concentrations of 100%, whereas germination of T. pratense was completely inhibited, almost invariably, at effluent concentration of 50%. Higher germination levels were achieved when the plants were exposed to effluent originating from the outlet of constructed wetland pilot units, allowing germination of all tested plants, indicating a significant decrease in its toxicity level. Experiments conducted with the same plants using different growing substrata as the germination matrix, namely expanded clay aggregates (Filtralite MR 3-8 and Filtralite NR 3-8) and two types of sand (fine gravel and standard sand) have shown that higher germination levels were achieved in standard sand and that P. australis was the plant species showing higher germination in all cases, reinforcing the robustness of this plant to environmental stress. The phytoextraction potential of P. australis, was evaluated by subjecting the plant to tannery wastewater supplemented with 50 and 150 mg Cr/L. After 6 weeks of exposure, levels up to 4825, 883, and 627 mg Cr/kg were found in the rhizome, shoot, and leaves, respectively, although phytotoxic signs in the plant were evident. This plant might not be considered a chromium hyperacumulator, but the potential to extract and accumulate this metal on its rhizomes is high.

A field survey of the arbuscular mycorrhizal status of herbaceous plant species was conducted in a highly alkaline anthropogenic sediment resulting from the disposal of waste from an acetylene and polyvinyl chloride factory. Most plant species found at the site were mycorrhizal and the dominant mycotrophic plant species was Conyza bilbaoana. Fungal species richness was assessed by identification of spores extracted from the sediment and from continuously propagated trap pot cultures. All of the six species of arbuscular mycorrhizal fungi (AMF) found were from the genus Glomus. Glomus intraradices and G. mosseae were found in field-collected sediment samples and also occurred most frequently in trap cultures. To test the symbiotic effectiveness of these two fungi, seedlings of C. bilbaoana were inoculated with either native G. intraradices BEG163 or G. mosseae BEG198 and non-native G. intraradices BEG75 or G. mosseae BEG25 isolates in sterile and non-sterile sediment collected from the study site. All four isolates were able to colonise C. bilbaoana. However, AMF native to the target sediments were generally more effective than the non-native fungi in promoting plant establishment and growth under highly alkaline conditions. The non-native G. intraradices was, however, more effective than the non-native G. mosseae. The results of this study suggest the use of adapted AMF as inoculants for phytorestoration of alkaline anthropogenic-stressed sediments.

The biosorption of three most prescribed FQs, ofloxacin (OFL), norfloxacin (NOR) and ciprofloxacin (CPF) by activated sludge (AS) and aerobic granular sludge (AGS) was investigated. Biosorption assays were conducted with FQs concentrations within the range of 100–700 ng mL−1, to mimic environmental conditions. At neutral pH and at the end of 48 h, AS showed higher biosorption capacity than AGS. For AS, a maximum biosorption of 1.50 ± 0.03, 3.24 ± 0.05 and 3.39 ± 0.06 mg gTSS−1 was observed for OFL, NOR and CPF respectively, whereas for AGS the maximum amount of FQs biosorbed was 1.18 ± 0.03, 2.73 ± 0.02 and 2.94 ± 0.03 mg gTSS−1. Langmuir isotherm was more applicable for describing FQs biosorption equilibrium by AS while for AGS, the Freundlich isotherm was more adequate. Given the AGS technology innovative character, the effect of change of pH on the biosorbed FQs was evaluated. FQs could be desorbed from AGS at pH 3, pH 8 and pH 9 but at pH 4 the biosorption process was promoted. This study allows a better understanding of the FQs biosorption processes. Moreover, the data from biosorption/desorption from AGS may be useful for management and operation of AGS bioreactors.

The inoculum was collected from a bioreactor performing simultaneous partial nitrification and aromatic compounds biodegradation. From day-56 onwards, 2-fluorophenol (2-FP) (12.4mgL-1) was added to the feeding wastewater and the system was bioaugmented with a 2-FP degrading bacteria (Rhodococcus sp. FP1). By the end of operation, complete 2-FP biodegradation and partial nitrification were simultaneously achieved. Aerobic granules remained stable over time. During the 2-FP loading, a shift in the community structure occurred, coinciding with the improvement of 2-FP degradation. DGGE analysis did not allow to infer on the bioaugmented strain presence but pyrosequencing analysis detected Rhodococcus genus by the end of operation. Together with other potential phenolic-degraders within granules, these microorganisms were probably responsible for 2-FP degradation.

The use of heavy metals (HM) contaminated soils to grow energy crops can diminish the negative impact of HM in the environment improving land restoration. The effect of two PGPR (B1--Chryseobacterium humi ECP37(T) and B2--Pseudomonas reactans EDP28) and an AMF (F--Rhizophagus irregularis) on growth, Cd and Zn accumulation, and nutritional status of energy maize plants grown in a soil collected from an area adjacent to a Portuguese mine was assessed in a greenhouse experiment. Both bacterial strains, especially when co-inoculated with the AMF, acted as plant growth-promoting inoculants, increasing root and shoot biomass as well as shoot elongation. Cadmium was not detected in the maize tissues and a decrease in Zn accumulation was observed for all microbial treatments in aboveground and belowground tissues--with inoculation of maize with AMF and strain B2 leading to maximum reductions in Zn shoot and root accumulation of up to 48 and 43%, respectively. Although microbial single inoculation generally did not increase N and P levels in maize plants, co-inoculation of the PGPR and the AMF improved substantially P accumulation in roots. The DGGE analysis of the bacterial rhizosphere community showed that the samples inoculated with the AMF clustered apart of those without the AMF and the Shannon-Wiener Index (H') increased over the course of the experiment when both inoculants were present. This work shows the benefits of combined inoculation of AMF and PGPR for the growth energy maize in metal contaminated soils and their potential for the application in phytomanagement strategies.

Cadmium is one of the most toxic heavy metals and its accumulation in the upper layers of forest soils affects plants, microorganisms and their interactions. Adequate strategies for the reforestation of metal contaminated sites are of vital importance. The aim of this work was to evaluate the response of Pinus pinaster seedlings to Cd exposure and to assess the effect of inoculation with two selected ectomycorrhizal fungi, Suillus bovinus and Rhizopogon roseolus on that response. Seedlings were exposed to soil contaminated at 15 and 30 mg Cd kg(-1). Shoot biomass of P. pinaster decreased ca. 36% when exposed to 15 mg Cd kg(-1). Overall, colonization by S. bovinus significantly enhanced shoot development up to 30% in contaminated soil while colonization by R. roseolus produced no significant effect at both Cd concentrations tested and significantly increased the level of Cd in the shoots at both Cd concentrations. Metal accumulation in the shoots and roots of non-inoculated and S. bovinus-inoculated seedlings increased at the higher Cd levels whereas R. roseolus-inoculated seedlings were not sensitive to Cd variation in the soil. The results from our research show that inoculation with ECM fungi has a significant impact on metal uptake and development of P. pinaster seedlings; the differential response induced by the two tested species highlights the importance of selecting the appropriate strains for nursery inoculation, and, as such, this biological tool ought to be considered in reforestation processes of heavy metal contaminated areas by woody species.

Zn accumulation in Solanum nigrum grown in naturally contaminated soil in the presence of different types of organic amendments was assessed. Under the same conditions, the response of the plant to inoculation with two different isolates of arbuscular mycorrhizal fungi (AMF) (Glomus claroideum and Glomus intraradices) was also evaluated. S. nigrum grown in the non-amended soil always presented higher Zn accumulation in the tissues, with the addition of amendments inducing reductions of up to 80 and 40%, for manure and compost, respectively, and enhancing plant biomass yields. The establishment of S. nigrum in the Zn contaminated soil combined with the application of amendments led to a 70-80% reduction in the amount of Zn leached through the soil. The use of S. nigrum in combination with manure appeared as an effective method for reducing the effects of soil contamination, diminishing Zn transfer to other environmental compartments via percolation.

Zn tissue accumulation in Solanum nigrum grown in a non-contaminated and a naturally contaminated Zn matrix and the effect of inoculation with different arbuscular mycorrhizal fungi (AMF) on metal uptake were assessed. S. nigrum grown in the contaminated soil always presented higher Zn accumulation in the tissues, accumulating up to 1622 mg Zn kg(-1). The presence of both Glomus claroideum and Glomus intraradices enhanced the uptake and accumulation of Zn by S. nigrum (up to 83 and 49% higher Zn accumulation, respectively). The main deposits of the metal were found in the intercellular spaces and in the cell walls of the root tissues, as revealed by autometallography, with the inoculation with different AMF species causing no differences in the location of Zn accumulation. These findings indicate that S. nigrum inoculated with selected heavy metal tolerant AMF presents extracting and accumulating capacities, constituting a potentially suitable remediation method for Zn polluted soils.

An up-flow fixed bed reactor (UFBR) was established to investigate the biodegradation of fluorobenzene (FB) under a number of operating conditions, which included variation in the concentration of FB in the feed stream (up to 180 mg l(-1)) and temporary suspension of feeding. Degradation of FB was followed for a period of 8 months under a continuous flow regime. During the operation of the UFBR, FB was never detected in the reactor effluent, being biodegraded by the microbial biofilm or adsorbed to the granular activated carbon (GAC). Biodegradation of FB was observed from the beginning of the reactor operation, and overall, it accounted for 50% of the total amount fed to the bioreactor. High organic loads of FB (210-260 mg d(-1) dm(-3)) were found to affect the biological removal efficiency, possibly due to an inhibitory effect caused by the higher FB concentrations fed to the bioreactor (149-179 mg l(-1)). When FB feeding was suspended for 1 month, biodegradation continued, indicating that the adsorbed FB became bioavailable. Biofilm bacterial dynamics were followed throughout the UFBR operation by denaturing gradient gel electrophoresis and plate-counting techniques, showing that a quite stable community was found in the bioreactor, and this was mainly attributed to the high selective pressure exerted by the presence of FB.

Microalgae are ubiquitous in aquatic environments, including aquaculture farms, but few studies have delved into their phytoplankton diversity and bioremediation potential. In this study, the cultivable phytoplankton of a rainbow trout freshwater aquaculture farm was isolated, phylogenetically analysed and used to assemble a consortium to polish an aquaculture-derived effluent, with low concentrations of ammonium, nitrite and nitrate.Through standard plating in different selective media, a total of 15 microalgae strains were isolated from sludge from a rotary drum filtering system which removes suspended solids from the water exiting the facility. Based on 18S rRNA gene sequences, isolates were assigned to nine different genera of the Chlorophyta phylum: Asterarcys, Chlorella, Chloroccocum, Chlorosarcinopsis, Coelastrella, Desmodesmus, Micractinium, Parachlorella and Scenedesmus. Species from most of these genera are known to inhabit freshwater systems in Galicia and continental Spain, but the Coelastrella, Asterarcys or Parachlorella genera are not usually present in freshwater streams. In an onsite integrative approach, the capacity of a consortium of native microalgae isolates to grow on aquaculture-derived effluents and its nutrient removal capacity were assessed using a raceway pond. After 7 days, removal efficiencies of approximately 99%, 92% and 49% for ammonium, nitrite and nitrate, respectively, were achieved concomitantly with a microalgae biomass increase of ca. 17%.Sludge from the aquaculture filtering system presents a high diversity of microalgae species from the Chlorophyta phylum, whose application in a consortial approach revealed to be efficient to polish aquaculture-derived effluents with low nutrient content.The use of native microalgae consortia from aquaculture systems can contribute to the development of efficient treatment systems for low-nutrient wastewater, avoiding nutrients release to the environment and promoting water recirculation. This may further strengthen the use of phycoremediation at the industrial scale, as an environment-friendly strategy.

Toxicity tests were performed to assess the effect of tannery wastewater with different treatment levels on two wetland plants, Phragmites australis and Typha latifolia, which are frequently used in constructed wetlands (CWs) for water treatment, and thus deepen the knowledge on their capacity to withstand the application of industrial wastewater. Trifolium pratense, a plant generally used as an indicator in toxicity tests, was included as a control. End points measured were germination percentage, shoot length, root elongation, and biomass growth of the plants. When tannery effluent, with a low treatment level, was supplied to the wetland plants germination occurred even at effluent concentrations of 100%, whereas germination of T. pratense was completely inhibited, almost invariably, at effluent concentration of 50%. Higher germination levels were achieved when the plants were exposed to effluent originating from the outlet of constructed wetland pilot units, allowing germination of all tested plants, indicating a significant decrease in its toxicity level. Experiments conducted with the same plants using different growing substrata as the germination matrix, namely expanded clay aggregates (Filtralite MR 3-8 and Filtralite NR 3-8) and two types of sand (fine gravel and standard sand) have shown that higher germination levels were achieved in standard sand and that P. australis was the plant species showing higher germination in all cases, reinforcing the robustness of this plant to environmental stress. The phytoextraction potential of P. australis, was evaluated by subjecting the plant to tannery wastewater supplemented with 50 and 150 mg Cr/L. After 6 weeks of exposure, levels up to 4825, 883, and 627 mg Cr/kg were found in the rhizome, shoot, and leaves, respectively, although phytotoxic signs in the plant were evident. This plant might not be considered a chromium hyperacumulator, but the potential to extract and accumulate this metal on its rhizomes is high.

A field survey of the arbuscular mycorrhizal status of herbaceous plant species was conducted in a highly alkaline anthropogenic sediment resulting from the disposal of waste from an acetylene and polyvinyl chloride factory. Most plant species found at the site were mycorrhizal and the dominant mycotrophic plant species was Conyza bilbaoana. Fungal species richness was assessed by identification of spores extracted from the sediment and from continuously propagated trap pot cultures. All of the six species of arbuscular mycorrhizal fungi (AMF) found were from the genus Glomus. Glomus intraradices and G. mosseae were found in field-collected sediment samples and also occurred most frequently in trap cultures. To test the symbiotic effectiveness of these two fungi, seedlings of C. bilbaoana were inoculated with either native G. intraradices BEG163 or G. mosseae BEG198 and non-native G. intraradices BEG75 or G. mosseae BEG25 isolates in sterile and non-sterile sediment collected from the study site. All four isolates were able to colonise C. bilbaoana. However, AMF native to the target sediments were generally more effective than the non-native fungi in promoting plant establishment and growth under highly alkaline conditions. The non-native G. intraradices was, however, more effective than the non-native G. mosseae. The results of this study suggest the use of adapted AMF as inoculants for phytorestoration of alkaline anthropogenic-stressed sediments.

Afonso;Abstract Fluoroquinolones (FQs) have been reported in trace amounts in different environmental matrices. The biosorption of three most prescribed FQs, ofloxacin (OFL), norfloxacin (NOR) and ciprofloxacin (CPF) by activated sludge (AS) and aerobic granular sludge (AGS) was investigated. Biosorption assays were conducted with FQs concentrations within the range of 100–700 ng mL−1, to mimic environmental conditions. At neutral pH and at the end of 48 h, AS showed higher biosorption capacity than AGS. For AS, a maximum biosorption of 1.50 ± 0.03, 3.24 ± 0.05 and 3.39 ± 0.06 mg gTSS−1 was observed for OFL, NOR and CPF respectively, whereas for AGS the maximum amount of FQs biosorbed was 1.18 ± 0.03, 2.73 ± 0.02 and 2.94 ± 0.03 mg gTSS−1. Langmuir isotherm was more applicable for describing FQs biosorption equilibrium by AS while for AGS, the Freundlich isotherm was more adequate. Given the AGS technology innovative character, the effect of change of pH on the biosorbed FQs was evaluated. FQs could be desorbed from AGS at pH 3, pH 8 and pH 9 but at pH 4 the biosorption process was promoted. This study allows a better understanding of the FQs biosorption processes. Moreover, the data from biosorption/desorption from AGS may be useful for management and operation of AGS bioreactors.
